In vitro cultures of medicinal of plants - VIII

In vitro cultures of medicinal of plants - VIII Method of elicitation is one of the possibilities how to increase production of secondary metabolites in cultures in vitro. The effect of abiotic elicitor (3-jodo-4-methylfenyl)amide 5- terc-butyl-6-chloropyrazine-2-karboxylic acid in three concentrations on the flavonolignans and flavonoid taxifolin production in Silybum marianum callus and suspension culture was tested in this study. Elicitation was run in this time intervals: 6, 12, 24, 48, 72 and 168 hours. Cultivation proceeded on Murashige - Skoog medium with the addition of 10 mg/l of α- naphtylacetic acid. The content of flavonolignans and flavonoid taxifolin was determined by HPLC. The maximum content of flavonolignans (0,05%) in callus culture was found after 168 hours of elicitor treatment in concentration of c2 = 2,33 x 10-4 mol/l. The maximum production of silychristin (0,04%) was found after 6 hours of affecting elicitor c1 = 2,33 x 10-3 mol/l. There wasn't statistical significant increasing of flavonolignans and taxifolin production in suspension culture. The maximum production of silybin A (0,01%) in medium of suspension culture was found after 24 hours of elicitor treatment in concentration of c1 = 2,33 x 10-3 mol/l.

The presented thesis deals with development and preparation of solubilisation systems for potentially photodynamically active substances. The aim was to determine the solubility of lipophilic photosensitizers of phthalocyanines azaanalogues type in several oils. MCT have been found suitable for ZIP21Zn and MUZ21Zn, and castor oil for P9Zn. These PS-in-oil solutions were incorporated into microemulsions using Cremophor® EL as the emulsifier. Molar extinction coefficients of photosenzitizers in selected oils were also measured. For photosenzitizer ZIP21Zn in MCT was found molar extinction coefficient 354400 l·mol-1 ·cm-1 at the wavelength of 653 nm; for MUZ21Zn in MCT was found the coefficient 416000 l·mol-1 ·cm-1 (746 nm), and for P9Zn in castor oil 352200 l·mol-1 ·cm-1 (703 nm). Novel thin scintillation films for 2D imaging screens

In this work we studied properties of Ce doped garnet scintillator layers (CexLu3-xAl5O12, CexY3-xAl5O12). They were prepared by liquid phase epitaxy. Studied materials show fast response, high quantum efficiency and good chemical and mechanical stability. Thus they are ideal for use in devices for 2D imaging. We measured absorption, excitation and emission spectra and kinetics of luminescence. Aim was to compare properties of grown layers with properties of single crystals grown by Czochralski method. We looked for the impact of melt compounds to measured layer properties. We also tried to determine optimal amount of Ce in layer. We used flux PbO - B2O3 and flux BaO - BaF2 - B2O3. In those fluxes we succeeded to grow garnet layers at temperatures as low as 1000řC. This lead to less intrinsic defects in crystalic lattice of layer in comparison to single crystals grown by Czochralski method (1900řC). Our layers were from 1 to 30 mm thick with higher concentration of Ce than singlecrystal. Due to higher concentration of impurities in layers grown from PbO - B2O3 flux we grown layers from BaO - BaF2 - B2O3 flux too. Flux BaO - BaF2 - B2O3 leaves less impurities in layers.


Oligoester matrices with antimicrobial drugs III.

Klára Lazařová Diploma thesis 2008 The aim of this work was the study of the fluconazol and terbinafine release from oligoester carriers branched with various concentration of either mannitol (3M, 5M, 8M) or dipentaerythritol (3D, 5D, 8D) and plasticized with 30 % triethyl citrate (TEC). Theoretical part is occupied with drug delivery systems and the conditions of the coupling of the macromolecules with the biological substrates. The 150,0 mg matrices composed of carrier, drug of 4% and plasticizer TEC of 30% were prepared. They were put to the static dissolution test using phosphate citrate buffer pH 5,0 at 37řC. The drug release was determined spectrophotometricaly at 261 nm for Fluconazole and 283 nm for Terbinafine. Fluconazole release was the fastest from carrier 5D. The carriers 5D and 8D released the drug during three stages, after burst effect was observed constant liberation followed slowing the process. The carriers branched using mannitol released the drug depending of the concentration of the mannitol.

Benefit of assesment of cytokines in inflammatory postoperative complications

Infection and the resulting sepsis continue to be important causes of morbidity in surgical patients. Cytokine network causes activation of neutrophils and vascular endothelium with increased upregulation of adhesion molecules, setting at a train of pathogenic inflammatory reactions in the host. Increased concentrations of inflammatory cytokines observed in the initial phase of postoperative complications have a great significance in forecasting a potentially fatal outcome of the disease and can be used for an early diagnosis of systemic complications.Prospective, clinical trial. Levels of cytokines, procalcitonin (PCT), leptin and acute phase proteins (APP) were measured before and after the operation (planned resection of colorectal cancer) and compared with levels in sepsis. Cytokine levels were measured by ELISA method. APP were estimated by nephelometric analysis.PCT was measured by immunoluminometric analysisDuring the first 24 h after the planned resection of colorectal cancer there was a significant increase in serum concentration of interleukin (IL)-6 which declined in next 48 to 72 h. Via in vitro assays during preclinical testing, we are able to predict potential toxicity which helps to prevent the possible serious adverse drug reactions in clinical practice. The aim of this rigorous thesis was to state the cytotoxic profile of 4 potential antituberculotic drug candidates using appropriate cell models and in vitro assays. Another aim was to compare the cytotoxic effect of the tested compounds among themselves and to 4-aminosalicylic acid (PAS) which antituberculotic activity is known for many years. The tested substances were 3 salicylanilide diethyl phosphate-based derivatives and 4-(trifluormethyl)benzoic acid. Vývoj metody na stanovení steroidních látek ve vodách

The theoretical part of the Master´s thesis describes steroids, their features, and the methods for their determination in waters. In experimental part three steroid hormones, namely 1,4-androstadiene-3,17-dione (ADD), 1,4-androstadiene-3-one-17(beta)-ol (boldenone) and 17(beta)-estradiol were selected. The compounds were concentrated from water by LLE and SPE extraction and determined by gas chromatography coupled to mass spectrometry. Several experiments were conducted to assess the recovery of specific extraction for studied compound.

The main aim of this thesis is to describe the complex transaction that is de-merger. It outlines the legal process of divisions, accounting and tax procedures. From the legal point of view, the main emphasis is put on the decisive date and draft terms of division. The accounting part concentrates on valuation and deferred tax. There is also a brief characterization of corporate income tax regarding divisions. The last part of this thesis describes some examples from accounting practice and their analysis and comparison.

Estimation of Radioactive Aerosol Particles Size Distribution.

The main purpose of these experiments is to compare results of the size distribution measurements using conventional aerosol methods with results of radioactive aerosols measurements. Additional purpose was to compare our results with published data on dependency of unattached fraction on aerosol particle concentration. Our data show the limitations of the commonly used approximation to this relationship.
