Preparation of chicken antibodies against phosphoserine residues of phosphoproteins
Benýšek, Jakub ; Tichá, Marie (advisor) ; Entlicher, Gustav (referee)
New type of immunogen for the preparation of chicken antibodies specific for O- phosphorylated amino acid residues of phosphoproteins was prepared: the α-casein phosphopeptide mixture was coupled to maleinylated derivative of bovine serum albumin. Affinity chromatography on immobilized Fe(III) ions (Fe(III)-IDA-Sepharose) was used for the separation of the α-casein phosphopeptide fraction from the α-casein peptide mixture obtained by the proteolytic digestion using trypsin. The presence of coupled phosphopeptide was shown by means of MALDI-TOF MS analysis. After chicken immunisaion with the prepared immunogen the immunoglobulin fraction was isolated from egg yolks that was further purified using affinity chromatography on α-casein-Sepharose and bovine serum albumin immobilized on Sepharose (BSA-Sepharose). The specificity of obtaine immunoglobulin fractions was tested by means of ELISA tests. The obtained results showed, that the prepared chicken antibodies and their fractions obtained by affinity chromatography on α-casein-Sepharose do not recognize the O-fosforyl-L-serine residues of phosphoproteins (α-casein, phosvitin, ovalbumin). Using the prepared different antigens it has been shown, that prepared chicken antibodies interact with maleinylated chain coupled to carrier proteins. .

Tooth extractions in patients with coagulation disorder
Houba, Robert ; Houbová, Helena (advisor) ; Vaněk, Jiří (referee) ; Stanko, Peter (referee)
The study deals with the question of minor oral surgery procedures, dental extractions especially, in patients suffering from haemocoagulation disorder. The aim of this study was 1) the clinical attestation of the risk of bleeding that may follow the dental extraction in patients with non-interrupted oral administration of drugs with the anticoagulant and antiplatelet effects and 2) suggestion of the schedules of surgical procedures suitable for this kind of patients and for the realization in the standard dental office. The opening theoretical part analyses the principle of haemocoagulation, the surway of individual inherited and acquired bleeding disorders and the main principles of treatment of bleeding patients in dental offices with the emphasis on the local security of the surgical wound. In the proper study we present the observation and evaluation of bleeding complications after the standard dental extractions in 3 groups of persons (age range, 19 to 93 years). In all patients, one or more extractions were performed. 99 patients were treated with oral anticoagulant therapy without any interruption and underwent 199 extractions. 149 patients, treated with antiplatelet therapy (acetylsalicylic acid) without interruption, underwent 347 extractions. 160 healthy subjects with 316 extractions served as a...

The legal nature of derivatives
Votýpka, Petr ; Kotáb, Petr (advisor) ; Dřevínek, Karel (referee)
Legal nature of derivatives This diploma thesis deals with the legal nature of derivatives. To the legal aspects of the nature of derivatives has not been yet devoted much attention in Czech or foreign literature, even if it is the legal nature that could provide the answers to many questions, seeking the cause of the current economic crisis in an incorrect and unprofessional use of derivative contracts. The thesis is divided into three separate parts. The first chapter is devoted to the issue that arises from the definition of derivatives. Next part classifies derivatives into several groups, especially in terms of their place where they are traded on those that are traded on the market and those they are traded over the counter. The second part focuses on the legal nature of derivatives. First, their similarity is analyzed in terms of character as a gambling contract, and then in terms of their purpose as an insurance policy. This section concludes with the list of various derivative contracts such as swaps, forwards and options and their legal characterization, with regard to the provisions relating to the Czech law, applicable in the case of trading in derivative contracts. In all parts of dealing with the legal principle is allowed for future regulation of private law contained in the new Civil Code....

Study of cytotoxicity of potential antituberculotics using selected methods on liver and kidney cell line
Katrnošková, Simona ; Ramos Mandíková, Jana (advisor) ; Bárta, Pavel (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Pharmacology and Toxicology Student: Mgr. Simona Katrnošková Supervisor: PharmDr. Jana Ramos Mandíková, Ph.D. Title of Thesis: Study of cytotoxicity of potential antituberculotics using selected methods on liver and kidney cell line During pharmacologic therapy, tissues of liver and kidney are frequently exposed to high doses of xenobiotics. Via in vitro assays during preclinical testing, we are able to predict potential toxicity which helps to prevent the possible serious adverse drug reactions in clinical practice. The aim of this rigorous thesis was to state the cytotoxic profile of 4 potential antituberculotic drug candidates using appropriate cell models and in vitro assays. Another aim was to compare the cytotoxic effect of the tested compounds among themselves and to 4-aminosalicylic acid (PAS) which antituberculotic activity is known for many years. The tested substances were 3 salicylanilide diethyl phosphate-based derivatives and 4-(trifluormethyl)benzoic acid. For the cytotoxic potential assessment, we used the parameter half maximal inhibitory concentration IC50. We have used methods determining the cell metabolic activity, aminopeptidase activity, lactate dehydrogenase leakage and activity of 3/7 caspases in this...

Cross-border cooperation between Czech Rebulic and Slovakia during 2007-2013 programming period 2007-2013
Vašák, Viktor ; Khendriche Trhlínová, Zuzana (advisor) ; Kouřilová, Jana (referee)
As the time goes by, one former Czech-Slovak territory has separated, forming two independent republics and members of European union. Division of common territory by a border, however, cannot affect cultural and linguistic proximity, which can be clearly identified in cross-border regions. The purpose of this thesis is to evaluate the cooperation amongst six regions located near mutual border during 2007-2013 period. By the means of publicly available content, an analysis of allocation of financial resources from ERDF and national budgets is executed. The qualitative evaluation of cross-border cooperation projects is delivered by the proposal of matrix for territorial impact assessment. According to percentual fulfillment of given goals, impact on barrier effect reduction and selected projects analysis, the overall added value of cross-border cooperation between Czech and Slovak republic during 2007-2013 period is specified. Main findings are then a result of combination of qualitative analysis and quantitative indicators for the researched period.

Monolayers of platinum nanoparticles prepared by dip-coating
Černohorský, Ondřej ; Grym, Jan ; Yatskiv, Roman ; Pham, V.H. ; Hudry, D. ; Dickerson, J.H.
Platinum is a transition metal known for its catalytic properties, which are further enhanced when employed in a nanoparticle form. We have recently shown that a monolayer of Pt nanoparticles deposited on semiconductor substrates forms high quality Schottky diodes, which were used in sensitive hydrogen sensors with a detection limit of 1 ppm of H-2 in N-2. Preparation of ordered monolayers of Pt nanoparticles is essential for the understanding of the behaviour of such an interface. To obtain a hexagonal closed-packed nanoparticle array, we prepared Pt nanoparticles stabilized by oleylamine and oleic acid with a narrow size distribution and uniform shapes. A monolayer prepared by dip-coating of Si substrate in the suspension containing Pt nanoparticles showed hexagonal arrangement within separate domains with the surface coverage up to 90%. The increase of the surface coverage with increasing withdrawal speed of the dip-coating process was observed
