Analysis of the cytosol of thyroid gland and its contribution in the differential diagnosis of nodular lesions
Pikner, Richard ; Topolčan, Ondřej (advisor) ; Fínek, Jindřich (referee) ; Třeška, Vladislav (referee)
Thyroid nodules represent the most frequent endocrine lession in our population and it is neccessary to differentiate malignant lessions from them. The aim of the study was to validate determination of selected angiogenic, proliferative, and appptotic markers in cytosol tissue extracts. We analysed 166 tissue samples (85 goitres, benign adenomas and 10 malignat tumours in which VEGF, bFGF, Endostatin, Thymidinkinase ans TPS were determined. Main limitation of cytosolic analysis is tissue sample volume, that must be about 1cm3 and interindividual variability caused by tissue sample heterogeneity. Best way is to compare normal with pathological tissue samples from one patient. We fund significant differences amog histological groups in VEGF, bFGF, Endostatin and maily Tymidinkinase and TPS. These differences are not sufficiently huge to distinguish goitres and benign lessions . We also did not find any correlation between cytosolic markers and iminuhistochemistry markers . Cytosol analysis is not able to measure local expression and its differences in anylysed tissue, but it is able to quantitatively determine mean levels of selected markers.

The analysis of aberrations in the localization of the radiated fields during irradiation of the patients with rectum tumors by EPID method
HARTMANOVÁ, Magda
The presented thesis deals with the analysis of deviations in radiation fields during the irradiation of the patients with rectal tumors. The first part of the thesis is focused on the current problems connected with rectal and rectosigmoid cancer both in the Czech Republic and worldwide. It also includes the description of risk factors, characterisation of the given type oftumor, manifestation of the disease as well as the prognosis. What is also tackled is the prevention, which is important and often neglected. It was necessary to mention the possibilities of screening and dispensarisation programs. The next part deals with the diagnostic process (from anamnesis to specialised examinations) as well as the therapy. Chemotherapy and surgery are mentioned only marginally whereas radiotherapy, including its planning and methods, is dealt with in a more detailed way. The main part consists of the analysis of deviations in radiation fields. The deviations have been obtained by means of EPID (electron portal image device) in a sample of patients with rectal and rectosigmoid cancer who are treated with the use of radiotherapy at the oncological ward of the Hospital České Budějovice, Inc. The maximum, minimum and average deviation in the localization of radiation fields were assessed statistically, by means of the khi-quadrate test. The results obtained will be submitted to the oncological ward of the Hospital České Budějovice, Inc., where they can contribute to the assessment of radiotherapy quality. The thesis includes supplements such as portal images and tables with graphs indicating the development of the incidence and mortality for the chosen diagnosis within the entire population.

The use of the DMS 48 test for the diagnosis of the mild cognitive impairment and the initiatory phase of dementia
Nikolai, Tomáš ; Loneková, Katarína (referee) ; Kulišťák, Petr (advisor)
This thesis presents a contribution towards the diagnosis of the Mild Cognitive Impairment and to the initiatory phase of dementia. We have used the DMS 48 test to analyze the differences in the area of memory capabilities damage. The research has been carried out with the sample of 50 patients with MCI, who have been diagnosed according to the actual Petersen concept. There has been a control group of 34 persons created with no exposure of the cognitive function damage. The research has proved that the achievements of the patients with MCI have been in the DMS 48 test significantly lower than the achievements of the control group. The achievements of the patients with MCI in the DMS 48 test have been also compared with the results of patients that have been tested in other chosen neuropsychological tests with the aim to confirm the ability of this test to identify hippocampal memory damage. We have chosen 2 neuropsychological memory subtests, which according to the theoretical constructs can measure the hippocampal memory damage and another two subtests, which are not able to distinguish the hipocampalnian memory damage from other types of memory damage. The results have proved that the DMS 48 has significant correlations only with the results of the subtests measured by the theoretical construct of the...

Solar cell non-destructive analysis
Černý, Michal ; Škarvada, Pavel (referee) ; Macků, Robert (advisor)
This thesis briefly analyse contemporary situation of photovoltaic systems and their actual developemenet. It shows a typical properties of most produced up-to-date types. Decision point of this thesis consist in nondestructive analyse of monocristalic unijunction solar cells. It describes two main diagnostic aproaches. First – through the use of measuring of VA characteristics, second – through the use of measuring noise characteristics. With selected samples are performed experimental measurements and estabilished basic parameters. In case of drone analysis considerably attention is devoted to experimental equipment for automatized measuring of spectral power density. This experimental equipment is consequently used for measuring some samples where we can observe microplazmatic noise.

Financial analysis of used cars market
Jinek, Daniel ; Sieber, Patrik (advisor) ; Kouklík, Michal (referee)
Thesis deals with analysis of the largest companies working on used cars market. The first phase of the practical part of this work consists of analysis of preselected sample of companies that are of service to create reference averages. The largest companies are then compared with these averages and are evaluated. In the second phase of the practical part, all the samples are compared using intercompany comparison methods and the results are established. Outcome of this work says there is only little to none relation between the size and the rank in the comparison. The small companies tend to reach high values. Another outcome of this work is that the largest company at used cars market in Czech Republic, AAA Auto, is ultimately the most powerful company of all that were tested in the sample.

Laboratory diagnostic of the thrombofilic diseases.
Studená, Šárka ; Blažek, Martin (referee) ; Pecka, Miroslav (advisor)
The thesis describes the occurance of trombophilic attacs and about their diagnostic possibilities in women with recurent spontaneous abortions. In the study, the sample of women tested were the ones who subscribed for treatment in the 2nd internal disease clinic - Department of clinical hematology of Faculty hospital and Medical faculty Univerzity Karlovy in Hradec Kralove in years 1995-2007. I have also used the materials from archive whilst investigating some samples from past. I was actively involved in this investigation in last couple of years. During writing conclusion of the knowledge gained, following criteria from the literature of our choice and search were used based on the described importance of trombophylic attacs with rising incidence of is capture: age - in this part we used the median of age from the observed sample number of undertaken abortions - this parameter has a great significancy, however it is needed to overview whole period of pregnancy, afterwhich the abortions arose (in first trimester is crucial third abortion and in second trimester the second abortion has been under the count) number of newborn children - the risk of spontaneous abortion is rising with every child the period of using contraception - the usage of contraceptives can in an early stage show the...

Fluorescent analysis of the differential protein expression in normozoospermic and asthenozoospermic sperm samples
Děd, Lukáš ; Čapková, Jana ; Kubátová, Alena ; Teplá, O. ; Pěknicová, Jana
Asthenozoospermia is one of the main seminal pathologies underlying male infertility. Previous proteomic studies have demonstrated the significant differences in the protein profiles between normozoospermic and asthenozoospermic sperm samples. Since these studies were primarily focused on the identification of differentially expressed proteins by mass spectrometry, we aimed to evaluate the ability of our diagnostic antibodies to detect the differential expression of selected protein markers by fluorescent microscopy and flow cytometry techniques. Therefore, we analyzed sperm samples from 30 men with normal and 30 men with astheno spermiograms, average by the panel of our diagnostic anti-human sperm (Hs) antibodies. Fluorescent microscopy and flow cytometry analysis revealed quantitative differences in the protein abundances between normo and astheno sperm samples, namely, in GAPDHs, evaluated with Hs-8 MoAb, VCP, evaluated with Hs-14 MoAb, and ATP synthase, evaluated with MoAb Hs-36. From the methodological point of view, we observed very high correlation between the data obtained by fluorescent microscopy and flow cytometry techniques and therefore both methods are useful for evaluation of protein differences associated with asthenozoospermia. From the clinical point of view, we observed the strong association of the low sperm motility in the sample with the expression of proteins, playing an important role in sperm energy metabolism (expected), but also with the expression of all tested intra-acrosomal proteins. These findings further demonstrate asthenozoospermia as a complex semen disorder frequently associated with other semen pathologies, which are not diagnosed by basic semen analysis, and the possibility to use monoclonal antibodies as a tool for diagnosis of protein associated sperm pathologies in the semen with the low sperm motility.

Mechanizmy upravy kupnej ceny
Gajdošech, Martin ; Smrčka, Luboš (advisor) ; Strouhal, Jiří (referee)
My diploma thesis focuses on the M&A transaction closing mechanisms. Their function is to reflect the value changes of the target company into the purchase price. Value change occurs during the time lag between the date of the financial statements and the date of the transaction closing. Throughout history, there have been two major approaches developed. The "Completion Accounts Mechanism" uses post-completion price adjustments to reflect the change of the net working capital and net debt during the interim period. The "Locked Box Mechanism", using fixed price, assures non-fluctuation of target value in the interim period by imposing strict restrictions on the seller's activities. In this research, I have analyzed 44 transactions closed in the Czech Republic. I have challenged the theoretical foundations and described the application of the mechanisms in practice. At the end, I have compared the Czech and European practices. Thesis findings: 1. All the theoretical features of the mechanisms were proven by an analyzed sample of closed transactions. The completion accounts mechanism is buyer-friendly, while the locked box is a seller-friendly mechanism. 2. The main driver in mechanism selection is bargaining power. Buyers were in a better negotiating position in 69% of the completion account transactions. On the contrary, the sellers had bargaining power in 100% of the locked box transactions. 3. The Czech Republic is an environment where buyers (big multinational companies) dominate. They have preferred the completion account mechanism that provides them with higher protection from value leakage or other risks associated with small CEE economies. A total of 91% of analyzed transactions were executed by the completion account mechanism in the Czech Republic between 2011 and 2012.

Gluten-free diet - reality in school facilities
JIRÁČKOVÁ, Marie
The topic of this bachelor thesis is gluten-free diet - reality in schools facilities. The goal of this work was to determine the number of school cafeterias in the Vysocina Region which provide gluten-free meals, to evaluate awareness of the canteen staff about preparation of the gluten-free diet and to describe the cooperation between the school cafeteria and family of the pupil with gluten-free diet. School meals are important to provide optimal intake of nutrition during the time the child spends in school, to establish proper eating habits and to better integrate the child into the group of its peers. Gluten-free diet is a necessary medical precaution especially for those with coeliac disease, in which case gluten can lead to villous atrophy, and for those with gluten allergy for whom gluten intake can cause anaphylactic shock. Adherence to the gluten-free diet is recommended also to those with non-coeliac gluten sensitivity diagnosis, although gluten causes only short-term complications, usually intestinal. The method of quantitative and qualitative research was used to make this thesis. The research sample consisted of 8 school cafeterias from the Vysocina Region which provide gluten-free meal to their pupils. Awareness about gluten-free diet principles of the managers of the cafeterias or of the cooks responsible for preparation of gluten-free meal was determined by means of semi-structured interviews. The calculation of the two-day menu of each cafeteria by means of the Nutriservis Profesional program was also part of this thesis. The results were compared with the recommended daily intake of energy, proteins, fat, and saccharides for pupils of elementary and secondary schools. The results of the quantitative research show that gluten-free diet is provided by only a small percentage of school cafeterias in the Vysocina Region. The qualitative research prove sufficient awareness of the cooks about gluten-free diet, with awareness of those cooks directly responsible for preparation of gluten free meals being at much higher level. It was also determined that the cooperation with parents of pupils with gluten-free diet is a usually one time occasion limited to an arrangement of nonperishable gluten free meal packages.
