Occurrence of human pharmaceuticals in the middle part of the Yangtze River. Literature rewiew compared to results from passive samplers POCIS.
KUBATA, Michal
Occurrence of human pharmaceuticals in the middle part of the Yangtze River. Literature review compared to results from passive samplers POCIS. Pollution of water environment by pharmaceuticals is becoming a major problem in recent years. Amount of discharged pharmaceuticals and personal care products (PPCP) is continually increasing in number and volume as well. The Yangtze basin is one of the world's most populated areas and its development is still growing. The goal of this thesis is to compare results from previous studies with concentrations of pharmaceuticals which were detected in POCIS samplers. Passive samplers were deployed in 10 localities in the middle part of the Yangtze River from October 12 to October 22, 2013. Temperature of water was from 20 to 21°C. Instrumental analysis was based on LC-MS/MS method. The passive sampling method was very succesful in detecting of pharmaceuticals. Range of detected substances was 39 PPCP. In our study, the total concentration of target substances ranged from 36 to 757 ng . POCIS-1 in respektive sites. The most abundant group were antibiotics. Several studies, which were aimed to asses the presence of pharmaceuticals in the river have been made to date. Authors of cited studies detected 36 pharmaceutical compounds. Influential substances were sulfamethoxazole, carbamazepine, indomethacine and ibuprofen. Their maximal concentrations reached more than 200 ng . l-1. Methods of the previous measurements consisted in collecting samples of surface water or sediments. Detection of polar micropollutants by using passive sampling is relatively new approach. Unfortunately it is not possible to compare results from water samples with data analysed in POCIS directly. There are calculations available only for limited range of pharmaceuticals and pesticides. Moreover most sampling rates were calculated in controled laboratory conditions only. There are many factors that can influence results of the filed measurements for example water temperature, turbulence conditions, fluctuation of concentration, biochemical degradation of substances during storage, etc. Amounts of concentration are mostly influenced by dilution in the river. Presence of individual substances depends on composition of using pharmaceuticals in particular area.

Analysis of climatological time series for selected places in Europe in the years 1961-2010
Míka, Tomáš ; Helman, Karel (advisor) ; Bašta, Milan (referee)
This bachelor thesis preoccupied with the analysis climatological time series mesured at seven selected meterological stations located throughout the European continent. The underlying data for this analysis were obtained from publicly available European database of ECA&D. The analysis is focused on calculation of the basic charakteristic and decomposition of time series. The aim of this bachelor thesis is to examine development of the time series of average monthly temperatures and precipation in the period 1961 - 2010 for individual weather stations and these data analyzed to compare with each other and explore similiraties and differences in temperature and precipation from individual station, not only in term of time but also terms of space. This thesis is divided into two parts. The first part is theoretical and generally discribes statistical methods. The second key part (practical) is devoted data processing and analysis of the time series.

Vondráčková, Ilona ; Chýlková, Jaromíra (referee) ; Hroch, Martin (referee) ; Kráčmar, Stanislav (referee) ; Vávrová, Milada (advisor)
In the presented PhD. thesis, problems covering occurrence of polybrominated diphenyl ethers (PBDE) and perfluorinated compounds in the environment were solved. The study was focused on identification and further verification of optimal methods in order to determine PBDEs and perfluorinated compounds sampled from the aquatic ecosystem. Polybrominated diphenyl ethers belong to persistent compounds classified as main pollutants; within the environmental constituents, they have been observed particularly in the last decade. For these studies, there were selected surface water matrices and sediments taken in various localities within the river Svratka basin. The analyses were to demonstrate whether they accumulate and remain in these specific matrices for longer time. There we assessed the congeners of polybrominated diphenyl ethers as follows: BDE-28, 47, 66, 85, 99, 100, 153, 154 and 183. In order to isolate them from matrices, various extraction techniques were used, i.e., ultrasonic extraction, microwave extraction, and pressure solvent extraction. Gas chromatography method with electron capture detection (GC/ECD) was selected for determination. The accomplished studies also assessed basic chemical, physical and environmental characteristics of diphenyl ethers in the environmental constituents. The attention was also paid to perfluorooctanoic compounds (PFOA), (PFOS) and (FOSA); physico-chemical properties of these organic pollutants were characterised and their toxicological and environmental aspects were evaluated. Usability of extraction techniques (ultrasonic extraction, pressure solvent extraction, solid phase extraction) applied to PFCs isolation from sediments samples was assessed. Identification and quantification of these analytes were performed using a high performance liquid chromatography/mass spectrometry method (HPLC/MS). After that, the optimized methods applied to real samples. Surface water and sediments were sampled within the river Svratka basin, in particular, from 19 sampling localities. PBDE occurrence was confirmed in sediments samples from the Svratka river (g.kg-1), PBDE were not detected in the surface water samples, their concentrations were below the detection limit, resp.; perfluorinated compounds were not detected in sediments and water samples at all.

The paper presents a method for filtration of particle count measurement in the time domain. The goal of this method is to remove extremal values caused by short-time instrument malfunction but preserve values representing fast events. The method offers interpolation of particle size distribution so that in can correlated with other variables measured at different time intervals.

Oslejová, Kateřina ; Kubíček, Vladimír (advisor) ; Lázníčková, Alice (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of biophysics and physical chemistry Candidate: Kateřina Oslejová Supervisor: ing. Vladimír Kubíček, CSc. Title of diploma thesis: HPLC Analysis of Benzimidazoles Using Core-shell Columns I The aim of this work was to transfer the HPLC method of determination of flubendazole and its metabolite, i.e. reduced flubendazole, from a classical C18 column (250×3,0 mm; 5 µm) to core-shell column Ascentis® Express C18 (10×3,0; 2,7 µm) and to find a suitable internal standard. Experiments were performed using liquid chromatograph Agilent 1200 Series® with both DAD and fluorescence detection. Flubendazole was detected with the DAD detector. Low concentrations of reduced flubendazole were detected with the fluorimetric detector. Several mobile phase compositions were tested. The mobile phase consisting of phosphate buffer (pH = 3,0; 0,025 mol/l) and acetonitrile (70/30, v/v) appeared to be a good choice for the separation under study. Two internal standards were exploited for the quantification, depending on the detection principle. Mebendazole served as the internal standard in the case of DAD detection, while oxibendazole was utilized for fluorimetric detection. The proposed method was successfully applied for analyses of real samples.
