Stanovení stresové odezvy smrku ztepilého na suché a zamokřené prostředí

The diploma thesis deals with the effects of abiotic stress on seedlings of Norway spruce (Picea abies (L.) Karsten), the impact of water deficit and abundance. In the theore-tical part is described the importance of water in plant, stress and its impact on plants and the cultivation of plants in vitro. Experimental part consists of experiments in vitro. Waterlogging conditions were simulated the medium without agar, drought conditions were created by increasing concentration of sucrose and polyethylene glycol. In the course of experiment were measured plant responses to stress, concentration of ethylene, ethane and CO2 in the culture vessels. At the end of the experiment was measured concentration of O2, quantum yield of photosynthesis, plant growth parameters, content of plant pigments in needles and anatomical changes in the stem and root of Norway spruce.

This bachelor thesis is focused on natural dyes primarily from plant sources. In thesis are analysed the most important groups of plant colourants according to their structure – tetrapyrroles (chlorophyll, phycobilins), betalains, flavonoids (anthocyanins), carotenoids and curcumin. In each group are discussed sources, structure, reactions with other components and application. There are not ommited benefits of consumation and effect on human health.
Refinery and characterization of grape oil

This bachelor thesis deals with the topic concerning the extraction of wine oil from grape seeds, the refining of the extracted oil and the characterization of grape oil. Grape oil was extracted from the grape seeds obtained from the harvest in September 2019 by the winery Vavricek (Brezi u Mikulova, Czech Republic). The received grape pomace was the mixture of Green Veltliner and Blanc Saugvinon. The oil was extracted from the separated seeds by using liquid extraction in n-Hexane. The extraction yields of grape oil were compared with the literature, involving the different extraction methods as well as different oil plants. For the refining of the grape oil have been applied more steps aiming for the removal of the lipophilic pigments. Different adsorbents of pigments have been applied. The purification efficiency has been proved by the investigation with the thin layer and the high-performance chromatography. Moreover, the gas chromatography was used to characterize the composition of the oil. Finally, the composition profile of the oil obtained by the liquid extraction before and after purification has been compared with the commercial grape oil, which has been received by cold pressing. The results of the work show that the winery waste produced in South Moravia is a valuable raw material, which can be used for the production and different products, e.g., grape oil and pigments (chlorophyll).
Chemical experiments for high school - chromatography with natural substances.

This thesis concerns experiments with natural compounds and vitamins. In the first part I described these substances, which I proved by chromatography or I made other experiments with them in the second part. I created four worksheets with instruction for laboratory practise. There are thein solutions in the supplement. I verified these four experiments at Grammar School in Prague 9.
Textile dyeing by vegetal dyes

1 ABSTRACT: Theoretical part of this bachelor thesis deals briefly with colour perception and history of dyeing with natural dyes. Also it devotes to dyer's plants, their use and colouring agents. Then the thesis deals with classification of natural dyes, their structure, use and organisms that contain these dyes. The accent is put on social important dyes and mainly on dyes that were analysed in the Experimental part. In the theoretical part of this thesis the principle of the used mass spectrometric method is explained. In the Experimental part the dyeing of small clothes by the selected plants is described. The the textiles were analysed by LDI MS (Laser Desorption/Ionisation - Time Of Flight Mass Spectrometry). The goal of this thesis was by using of the method of mass spectrometry determine the known dyestuffs in black elder (Sambucus nigra), blueberry (Vaccinium myrtillus), onion (Allium cepa), red currant (Ribes rubrum), walnut (Juglans regia) and St John's wort (Hypericum perforatum). Inquiry-based science education of plant pigments in upper primary school

This thesis is focused on interconnection between education of plant pigments and inquiry- based science education in upper primary school. The theoretical section of this thesis deals with the current quality of education and knowledge level of Czech pupils in science subjects, as well as characteristic of inquiry based science education and description of various types of plant pigments. Evaluation of publications focused on occurrence of plant pigments in curricular document is another part of this thesis. The study revealed that plant pigments represent only a marginal topic of these publications. The next section of this thesis is a questionnaire survey about utilization of inquiry based science education among the teachers of biology in upper primary school. The results showed that most of the teachers were well acquainted with the method and applied it during their teaching. The last section of this thesis contains two proposals for experimental tasks of inquiry-based science education of plant pigments, which are focused on interface of biology and chemistry. Both tasks were successfully solved.
