Wnt/β-catenin signalling in the development of the marine annelid Platynereis dumerilii

Radim Žídek "Wnt/β-catenin signalling in the development of the marine annelid Platynereis dumerilii" (dissertation) Abstract: Wnt/β-catenin signalling is absolutely crucial for the early embryonic development of metazoan animals from the establishment of body axes, through the specification of germ layers and tissues to the development of organ systems. I used pharmacological manipulations of the Wnt/β-catenin pathway activity in the planktonic larvae of the marine polychaete annelid Platynereis dumerilii, the representative of the clade Spiralia, to investigate the role of Wnt/β- catenin signalling in the development and evolution of three hallmarks of Bilateria: the central nervous system, the body segmentation and the digestive tube. Wnt proteins are produced in all three aforementioned systems in Platynereis where they trigger the Wnt/β-catenin pathway in neighbouring cells. I describe here, for the first time in Platynereis, a homologue of the endpoint transcription factor of the entire pathway, Pdu-Tcf, which is subjected to an alternative splicing and along with a Wnt target gene Pdu-Axin is expressed in tissues with the active Wnt signalling - in the brain ganglia, in the neuroectoderm along the ventral midline, in segments, in the posterior growth zone and in the gut. TITLE: Leeches (Hirudinea) and Their Usage in Teaching AUTHOR: Bc. Anna Koldová DEPARTMENT: Biology and Environmental Studies, Faculty od Education Charles University SUPERVISOR: Mgr. Dagmar Říhová, Ph.D. ANNOTATION This diploma thesis is focused on leeches (Annelida: Hirudinea) in the Czech Republic. It provides a description of all species occurring in the Czech Republic, as well as the key to their classification into families. A detailed guideline about the organization of field trip and implementation of extracurricular teaching is part of this thesis. The field trip is focused on sampling, observation and determination of leeches in an excursion to a site with a traditional occurrence of the European medical leech Hirudo medicinalis - national nature reservation Hrabanovská černava. In addition, the work defines the advantages, disadvantages and other possibilities of application of the proposed program, and the inclusion of extracurricular education. Teaching of Annelids in Podkrušnohorské gymnázium (The Case Study)

The main object of this thesis is to evaluate the results of a case study of teaching the topic of annelids. The study was carried for three years at Podkušnohorské gymnasium grammar school in Most. Another objective is to find the connection between obtained data, activity of students during classes and other chosen factors that may affected a success in the classified test and post-test. The thesis is divided into two parts. First part presents the research part which is devoted to a brief overview of textbooks for second grade of elementary and high school. The second, analytical part, is divided into three subsections. The first subsection is devoted to the evaluation of teaching methods for individual teachers Most schools with an emphasis on the level of activity of students in biology classes. The second subsection analyzes the results of the investigation and commented the results on, especially the success rate of students in the various test phases. The third subsection suggests, based on the data obtained, the best teaching methods to the topic of annelids. Branchiobdellidan population on alien species of crayfish in the Czech Republic

The thesis summarizes the current knowledge about the class Branchiobdellae including their anatomy, ecology and geographic distribution with respect to the presence of introduced species in Europe. Using samples of Branchiobdellidan obtained from two introduced species of crayfish caught in a pond near Čáslavice u Moravských Budějovic (signal crayfish, Pacifastacus leniusculus) and in Černíský stream (eastern crayfish, Orconectes limosus), two out of seven European species of Branchiobdellidan were identified, namely B. parasita with 100% presence in the eastern crayfish and 58.7% presence in the signal crayfish, and B. pentodonta with 41.3% presence. Based on data collected from the caught crayfish and samples of Branchiobdellidan, the individual weight of eastern crayfish was proved to influence the quantity and biomass of Branchiobdellidan on the body of crayfish (p<0.001), i.e. increased quantity and biomass of Branchiobdellidan was found on more robust specimens of crayfish, however the effect of gender was not statistically significant. With respect to the signal crayfish, a significant difference was observed in the quantity and biomass of Branchiobdellidan depending on the weight of the crayfish (p<0.001), and the influence of the gender of the crayfish was detected (p<0.001), i.e. the quantity and biomass of Branchiobdellidan was higher on the surface of male specimens of crayfish. The presence of eggs in some female signal crayfish had no significant effect on the quantity and biomass of Branchiobdellidan (p=0.052), however in combination with the weight, the influence of the presence or absence of eggs was proved (p=0.011), more robust females showed increased quantity and biomass of Branchiobdellidan.
