Biological age prediction using DNA analysis

Eye colour, hair colour and biogeographical origin can be predicted based on DNA analysis. This is used in forensic genetics when an unknown perpetrator leaves a biological material on a crime scene. Prediction of biological age is the subject of further research. Nowadays, there is no standardised model, which can be used for accurate prediction. The best option for biological age prediction is DNA methylation - stable epigenetic modification. Many markers have been evaluated. Methylation of these markers correlates with biological age positively or negatively. Methylation has tissue specificity, so one model cannot be applied on all types of tissues. Model for biological age prediction can be applied during investiganting of a criminal activity, when the identity of a person commiting a crime is unknown. This master thesis deals with forensic genetics. Primarily is forensic genetics put into the historical context. Then, there is described a biological material and the possibility of detecting biological traces. Further work is focused on the analysis of DNA, while there are elaborated in detail the techniques used as nucleic acid isolation, quantification of DNA, as well as types of polymerase chain reaction (PCR). Also the thesis deals with methods of obtaining DNA profiles - electrophoresis and sequencing and their variants. Finally, the thesis concern the issues of interpretation of forensic genetic data. Forensic biological exercises for high school students are processed in the didactic part. It has been tested in practice in biological seminars at Jiráskovo gymnázium in Náchod. Significance of repetitive sequences in clinical and forensic genetics

Repetitive sequences are important compartments of the genome and they are important for the whole organism as well. These non-coding sequences take a lot of space in the genome and they are called "junk DNA". However, they are valuable in many science sectors, especially because of their polymorphic character among individuals and also among tissues of one individual. This work pursues the significance of microsatellite repeats in clinical and forensic genetics. Some tumors have microsatellite instability (MSI) when compared to the repeats in the healthy tissue. This can be used to diagnose cancer. MSI sometimes appears before the disease fully breaks out, which could lead into an early diagnosis of cancer. MSI is associated with better prognosis. Forensic genetics takes advantage of microsatellite polymorphism among individuals. DNA profiling is used to identify persons in criminal investigations but also in parental testing or protection of wild animals.
Role of polymorphic DNA markers in personal identification and determination of selected phenotypic traits

Nowadays intensive research is conducted for application of genetic polymorphisms for degraded samples analysis, identification and kinship determination. Another area of research in forensic genetics is biogeographical and phenotypic traits (eye, hair and skin color) determination. First part of presented work dealt with population study on the Czech popu- lation using Investigator DIPplex (QIAGEN, Germany) marker set containing 30 autosomal insertion-deletion polymorphisms. Power of Discrimination (PD), which is the probability of random selection of two persons with different genotypes, was 99.9999999999% for the whole marker set. This part of study concluded that ana- lyzed marker set is suitable as an additional marker panel for identification and kinship determination in the Czech Republic. Second part of the presented study was devoted to population research of Cen- tral Croatia using Mentype Argus X-8 kit (QIAGEN, Germany) containing 8 short tandem repeat polymorhisms located on X choromosomes (X-STR) divided into 4 linkage groups. PD for the whole kit reached 99.9999% and 99.99999999% for males and females, respectively. DNA analysis serves as a highly efficient identification tool when the biological material to be identified is heavily damaged or does not carry sufficient morphological characteristics. It is employed for human, but also for non-human identification in numerous cases such as crime investigation and prevention, populational studies, kinship analysis, paternity testing, mass grave body recognition or endangered species rescue. For successful identification, it is necessary to collect and maintain DNA profile databases. Such databases hold records of people involved in criminal activity or sequences from non-human sources. This work intends to review how identification and kinship analysis using genetic material works. Next, the most important databases collecting DNA profiles are summarised. Although the benefits of such databases are vast, there are many ethical concerns about misuse of such private information. For convenient database queries, a dedicated set of command line tools has been implemented by the author. These are used for an exemplary species identification of an unknown sequence.
Age estimation from human biological material based on DNA methylation

DNA methylation is one of the epigenetics modifications which performs many functions in human body by silencing genes and maintaining genomic stability. Changes in DNA methylation accumulate during life and current studies indicate that the level of methylation in certain loci (CpG dinucletotides) correlates with chronological age. However, the methylation pattern changes not only with increasing age but it can also be affected by our lifestyle choices and biogeographical origin. Therefore, the key for succesful age estimation is finding such CpG loci that are independent (or as little dependent as possible) on other variables. This bachelor thesis describes the issue of age estimation based on DNA analysis of specific methylated DNA loci with focus on its application in forensic practice.
