Diagnostika parazitárních onemocnění a úspěšnost vybraných terapeutických postupů u lichokopytníků a sudokopytníků

This dissertation dealt with the diagnosis and therapeutic procedures for the treatment of parasitic diseases in odd-toed and even-toed ungulates with selected anthelmintics. Between 2020 - 2023, 5.359 artiodactyls and 819 solipeds, namely cattle, sheep, goats, aurochs, mouflon, deer, bison, bison bonasus, horses, donkeys and ponies, were microscopically examined for the presence of nematodes, trematodes or cestodes in faecal samples in the Czech Republic. Molecularly, 76 cattle samples, 26 bison bonasus samples and 52 bison samples were examined for the presence of trematodes and C. daubneyi was detected. Parasitic infections were monitored in naturally infected individuals, and during the study no clinical signs suggestive of parasitemia were observed in any of the monitored individuals. The most parasitic infections were evaluated for H. contortus, O. ostertagi, M. benedeni, C. daubneyi and A. perfoliata. As part of the dissertation, a total of ten anthelmintics available in the Czech Republic were chosen, focusing on individual categories of parasitic infections in various animal species. Based on laboratory analyzes of LDA tests, the species of nematode was determined according to the developmental stage of the larvae, which helped to choose the appropriate anthelmintic during treatment. In conclusion, it can be stated that almost all anthelmintics have been shown to be effective in therapeutically treated farm animals.
Enterobius vermicularis

The aim of this survey is to find out informations about the progress, diagnosis and treatment of this infection and public awareness of this disease. Methods: Questionnaire study among parents, whose children have ever been infected with Enterobius vermicularis. Results: Pinworm infection is the most common among children aged 4-7 years. Half of the children who visited a pediatrician were examined. In almost 25 % of the cases, the stool sample was collected. The children were treated mostly with prescribed medication, the parents rarely chose home-treatment. In one third of the cases, the other members of the household were infected too. In the half of the cases the whole family was treated preventively. The most common symptom is rectal itching (83 %). Complications are exceptional. 60 % of the respondents incorrectly think that it is possible to get infected with pinworms from animals. Most of these drugs are administered by the oral route, where absorption into the systemic circulation occurs through the intestinal barrier. However, the course and extent of this absorption may be limited by biotransforming enzymes and transport proteins, in particular the family of so-called ATP-binding cassette transporters. These transporters are capable of returning many xenobiotics, including many drugs, back into the lumen of the gut and are the first line of defence against the entry of these substances into the body. An important representative of this group of transporters is P-glycoprotein, which is known for its broad substrate specificity. On this transporter, drugs can act as substrates but also as inhibitors and/or inducers, which may lead to the risk of drug-drug interactions. Lenka Martínková Supervisor: RNDr. Lucie Raisová Stuchlíková, Ph.D. Title of diploma thesis: Biotransformation of fenbedazole in soya (Glycine max) Veterinary drugs are used in large amounts in modern husbandry for treatment and prevention of diseases in animals. Anthelmintics administered to animals enter into environment primarily through its excretion in faeces or urine. Following excretion, drugs may persist in the environment and impact non-target organisms. Plants are able to uptake xenobiotics, including drugs, and detoxify them via biotransformation. However, only drug biotransformation into non-toxic and stable metabolites and their consequent accumulation in plants represent drug detoxification. For that reasons, knowledge of biotransformation pathways of drug in plants is very important. Soybean plants, eventually seeds, are further used in agriculture as feed for cattle and absorbed anthelmintics including their metabolites can enter the food chain. The results showed that fenbendazole entered plant and enzymatic systems of plant were able to biotransform fenbendazole via several reactions. We found differences in the metabolites between the roots, leaves, seeds and pods of soybean.

Infection with these parasites mainly with the barber's pole worm (Haemonchus contortus) causes considerable economic losses and has a significant impact on sheep productivity. Anthelmintics, including ivermectin, are used for treatment. Ivermectin belongs to the class of macrocyclic lactones and is characterised by broad spectrum and low toxicity. On the other hand, it poses a risk to the environment in form of residues that are excreted in feces by treated individuals. The aim of this study was to determine the excretion profile of ivermectin in sheep subcutaneously administered in a standard dose 0,2 mg/kg of body weight. HPLC Determination of Benzimidazoles

Title of diploma thesis: HPLC Determination of Benzimidazoles In this thesis we have investigated the possibility of establishing benzimidazole with HILIC chromatography using a chromatographic column Ascentis Express HILIC 10.0 cm x 3.0 mm; 2.7 microns. Two groups of benzimidazoles were tested. The first one was focused on albendazole and its metabolites. Experiments with these substances did not produce satisfactory results, because the mobile phase composition which enables separation of the studied analytes was not found. The second group was formed by flubendazole and its reduced and hydrolyzed form. In this case, several mobile phase compositions were tested. Finally, distribution of individual analytes in a mixture, using a mobile phase ACN:HCOOH 0.03 mol/l (90:10), was successfully achieved. Ricobendazol was chosen as an internal standard. When separation conditions were found, calibration curve for the determination of reduced flubendazole in biological samples was subsequently constructed using ricobendazol as the internal standard with the use of fluorescence detection.
