Remote WakeOnLan

This work deals with construction of an embedded system with remote access, whitch controls computers in the same network in order to reduce their energy consumption. For this purpose is used the WakeOnLAN technique. The device also uses two power components called SSR and  these are used for switching AC power to them attached appliances. The work discusses the TCP/IP architecture, system design and finally the firmware implementation.

Control system overhaul of a granulate transport equippment

This thesis deals with the design and overhaul of the suction system which was part of the extruder dosing device from Reifenheuser. The first part describes the pneumatic transport and its main parts and control possibilities. Moreover, the thesis deals with the evaluation of the original solution of the control system, its advantages and disadvantages, the requirements for modernization from the client and proposals for its implementation. The last part focuses on the selection of suitable components and subsequently on the final implementation of the entire project, which consists of the design of the switchboard, the creation of electrical documentation and PLC programming.
Controller of hot runner for injection molds

The thesis focuses on implementation of a hot runner controller for injection molds for plastic materials with a maximum number of 56 regulated zones. The first part of the thesis deals with already existing and used solutions of the given subject. The following part of the thesis describes the principles of temperature measurement by means of thermoelectric sensors, including their linearization as well as communication protocols MODBUS, EUROMAP 17. The identification of a regulated system ( heating zone) has been carried out before the hardware design. The development process is divided into some parts : system conception, hardware design, software development. The controller consists of measuring modules designed to collect data and handle power controllers for management of performance of heating elements. The necessary calculation power is integrated into the controllling computer. All parts of a hot runner controller are implemented taking into account the costs.
Adjustable timer for three-phase motor

The thesis describes several potential solutions of an adjustable timer, such as monostable multivibrators, asynchronous (ripple) and synchronous counters and microcontrollers. It also deals with issue of switching three-phase asynchronous motors with limit a high starting current on starting - star-delta switch, softstarter, frequency inverter. Particular attention is focused on the conceptual design of the timer using micro- controller Atmel (timer control unit), LCD (display device) and control switches. The microcontroller will be complemented by external 32,768 kHz watch crystal, necessary for real-time function - Real Time Counter (RTC). The cause for this choice is not only simplicity and clarity of control to the operator, but also reliability, low power consumption and also the price of the device itself.
Evaluation of genetic strains polymorphism of \kur{Beauveria bassiana} based on analyses of using microsatellite markers

Microsatellites, or Simple Sequence Repeats (SSRs), are highly polymorphic loci present in nuclear and organellar DNA that consist of repeating units of 1-6 base pairs in length. Microsatellites are very effective molecular markers, which allow us to examine various aspects of organisms. The aim of thesis is to carry out analyzes of microsatellites and describe the genetic relationships between individual strains and monosporic isolates, which were derived from these strains. Usind SSR I would like to asses the degree of genetic polymorphism between \kur{Beauveria bassiana} strains, collected during monitoring in the locality of Černá Hora in Šumava. I am also trying to optimize the method of DNA isolation and method of microsatellites analysis. 44 strains of \kur{Beauveria bassiana} isolated from \kur{Ips typographus} were analyzed in total using four primers. Overall, the results that I obtained show high similarity of evaluated strains, and therefore low level of genetic polymorphism. For further work it would be appropriate to enlarge the area, which will be sampled and include sets of strains that were obtained from different developmental stages of the host, as well as other reference strains of entomopathogeníc fungi collected from other localities with similar conditions. For more detailed study it would be useful to use the analysis of ribosomal DNA. Thus selected and evaluated population is likely to provide a more veritable image of strain diversity in observed area.

Hybridization of crab apple with domestic apple trees

This bachelor thesis summarizes known information about hybridization (anthropohybridization) crab apple with orchard apple. Next it follows up mainly genetic and morphometric methods that haven't been used in the hybridization studies yet. Crab apple (Malus sylvestris) is the only one wild species in Malus genus in Europe. However in the last few decades these populations are endangered by orchard apple (Malus ×domestica), that originates from central Asia. It often outspreads into the free nature and thereby closer to wild apple plants, that leads to easier hybridization of these two woods. This process leads up to decreasing number of genetically pure crab apple individuals and also to slow hybrid plants expansion. This trend, which was called anthropohybridization, can be observed also in the Prunus genus, where there hybridizes wild species European dwarf cherry Prunus fruticosa with tart cherry Prunus cerasus. Recent studies though refers to following genetic aspects of Malus domestica, namely formation by triple hybridisation. Several locations of crab apple were observed and analysed in Europe in the last few years for study of anthropohybridization degree with Malus ×domestica. Molekulární fylogeneze a genetická diverzita nejbližších příbuzných rodu Pisum

The topic of this work was studied genetic diversity and phylogeny of selected wild species Lathyrus neurolobus Boiss. & Heldr., which is closely related to cultivated pea, using modern methods of molecular phylogenetics. Genotypes of L. neurolobus were obtained from Department of Botany at Palacký University in Olomouc, genomic DNA was isolated of these genotypes and samples were subjected to analysis using SSR and iPBS markers. It was found that for assessment of genetic diversity is suitable method of iPBS markers, where the average value of polymorphic information content is very high (PIC=0,928). According to evaluated markers polymorphism and assembled dendrograms was found that for search of relationship between place of origin and placement in dendrogram, will be appropriate the method of SSR markers, but only in case of selection informative markers. There was also confirmed transferability of SSR markers designed for pea to wild species of another genus.
