Macrophages and nitric oxide in leishmania - sandfly - host interactions

Leishmania reside fagolysosome of macrophages immediately after their entry to host where they multiply and consequently infect other macrophages or eventually other cells. A synthesis of a reactive reactant of oxygen and nitrogen is one of the mechanisms that some mammal cells are equipped with and that also contributes to eradication of leishmania. Nitric oxide rising during a metabolic change of L-arginine under the catalysis of NO synthase is of a large importance. Beyond cytotoxic function, nitric oxide is involved in signalling pathways for a neurotransmission (nNOS) and vasorelaxation (eNOS). Not all types of macrophages have ability to produce NO (iNOS). It is a heterogeneous group differing in immunological function and also in physiology. A group of classical activated macrophages represents an effective APC capable of efficient killing of intracellular pathogens. In addition to NO, they also secrete an inflammatory cytokines, which evolve an immune reaction towards to Th1. Contrary to this, a group of alternative activated macrophages is not capable of any efficient antigen presentation and nitric oxide production but produces L-ornithine, which is a precursor of polyamines, which leishmania utilizes for its own intracellular growth. Biochemistry of polyphenols effects in the treatment of vascular disease

This Ph.D. thesis deals with the deep analysis of polyphenols effects toward vascular disorders. This work provides a number of experimental results of studying both the effects of natural bioactive compounds in red wines and potatoes, and their application to the experiments which includes experimental animal models with spontaneous hypertension. Methods and equipment used in experimental studies allowed us to make several new statements regarding the universal nature of the relationships between the antioxidant capacity and the polyphenolic content in examined foodstuffs. Besides this it is also recognized that food is a source of minerals which also contribute to the total antioxidant capacity and therefore may have influence the endogenous antioxidant enzyme system by providing the essential cofactors. Experimentally we have found that the concentrations of magnesium known for its therapeutic action in wine and red wine extract are comparable to the mineral waters recommended for the treatment and prevention of cardiovascular and metabolic diseases. Moreover, synergistic interactions between selected minerals have been found. Somatisms in Czech Phraseology (The Respiratory System)

The bachelor thesis deals with the linguistic picture of the world of somatic idioms with components of the respiratory system, namely the nose, throat, lungs, diaphragm, and bronchi. The theoretical part explains the basic concepts and fundamentals of cognitive and cultural linguistics, primarily focusing on cognitive linguistics, the linguistic picture of the world and its definition and properties, anthropocentrism, the theory of conceptual / imaginary schemes, metaphors, metonymy and its types, and last but not least, phraseology. The practical part is organised into chapters, each of which revolves around a single corresponding somatism. Their etymological and anatomical characteristics are discussed, as are particular semantic areas and idioms in which the somatisms play a primary role. Lastly, those imaginary schemes thar are involved in the conceptualization of a given organ in the linguistic picture of the world are presented. The thesis is supplemented with the knowledge of psychosomatics which, given the premises of some idioms being conditioned directly by psychosomatics itself, have turned out to be of great utility.
Pharmacological animal model of Alzheimer's disease (rat model Samaritan) and mediator system of N-methyl-D-aspartate receptor and nitric oxide

Alzheimer's disease is a neurodegenerative disorder with the highest prevalence in the population and for which we do not have a cure so far. The aim of this thesis was to test the mediator system of the N-methyl-D-aspartate receptor and nitric oxide in an animal model of sporadic form of Alzheimer's disease (Samaritan Alzheimer's Rat Model; Taconic Pharmaceuticals, USA). Then compare these results with changes in hippocampal cholinergic system and cognitive tests. The Samaritan rat model is based on the unilateral in vivo application of β-amyloid42 and the pro-oxidative substances (ferrous sulfate heptahydrate and L-buthionine-(S,R)-sulfoximine). Neurochemical methods included testing of the NR1/NR2A/NR2B subunits of the N-methyl-D-aspartate receptor and activity of nitric oxide synthases (neuronal, endothelial, inducible) in the cortex, in both cases in the right and left hemisphere separately. Our results show that Samaritan rats exhibited significant changes in expression of NR2A/NR2B subunits of the N-methyl-D-aspartate receptor and activity of inducible nitric oxide synthase in cortex compared to control rats. The results of glutamatergic system are consistent with changes in activity of cholinergic transporter and cognitive tests (Morris water maze and active allothetic place avoidance)....
Nose in the Czech sign language in comparison with Czech

I continue my work on the cognitive-linguistic research of somatisms (names of body parts) and conceptual profiles, into which I bring research findings of Czech sign language. I performed a structural analysis of the signs, where a place of articulation is on the nose. Then I categorized these signs using conceptual profiles: GESTURE, APPEARANCE/ MANIFESTATION, FUNCTION and LOCALIZATION. On the basis of such profiles are shown new possibilities etymological interpretation of a relatively large number of collected signs. The created linguistic picture of the nose in the Czech sign language I compared with the image with the linguistic picture of the nose in the Czech language. I also conducted preliminary probes into other sign languages, which I have served as a guide for effective grasp of the issues.

The thesis tries to grasp the meaning of lexeme nos "the nose" in cognitively-linguistic perspective, namely in particular through colocation and idioms, which contains the lexeme nos. The goal of this thesis is to define so-called semantic profiles of concept NOSE and to formulate cognitively cultural definition of nose consequential from its location in the Czech linguistic picture of the world. For this purpose nose first will be subjected to analysis from two different perspectives - from medical perspective, which represent the scientific view on the world, and from the perspective of a common user of the language, which represent so- called naive picture of the world. Data relating to the lexeme nos will be excerpted from Czech monolingual, synonymous, etymological and phraseological dictionaries, the semic analysis will be done and the lexeme nos will be included to systemic relationships in Czech vocabulary. Nose also will be introduced in a ligth of an additional perspectives of an alternative medicine and a pseudoscientific disciplines: the phrenology and the physiognomy.
