Climate Monitoring at Home

Cieľom tejto bakalárskej práce je vyvinúť, zostaviť a otestovať zariadenie na meranie enviromentálnych veličín v interiéri. Zariadenie obsahuje senzory teploty, relatívnej vlhkosti, tlaku a koncentrácie CO2. Aktuálne hodnoty su zobrazené na vstavanej obrazovke spolu s časom a odosielané pomocou IoT protokolu MQTT na server. Práca obsahuje aj riešenie tohoto serveru. Dáta sú ním spracovávané, ukladané do SQL databázy a ďalej sprístupnené cez JSON API implementované v jazyku Python. Aplikácia vyvinutá pomocou niekoľkých webových technológií ponúka prehľad aktuálnych aj historických hodnôt. Serverové riešenie je hostované na http serveri. Demonštračná verzia riešenia využíva server NGINX ktorý tiež poskytuje zabezpečenie komunikácie so serverom pomocou TLS pre všetky služby. Celá práca je spracovaná s dôrazom na otvorenosť, modularitu a rozšíriteľnosť. V práci sú preferenčne používané Libre a Open Source technológie.
Field sensor network for microclimatological measurements

Tato práce je zaměřena na návrh terénní bezdrátové senzorové sítě pro mikroklimatologická měření, zejména výronů juvenilního oxidu uhličitho v oblasti hydrotermílního krasu, založené na LoRaWAN. V teoretické části práce je předložen přehled cílové oblasti, možností řešení měření, získávání a přenosu dat. Praktická část se pak věnuje kompletnímu návrhu celého systému, to znamená od měřicího zařízení přes koncentrátor a gateway až po backend odesílání dat do cloudového úložiště. Výsledky tohoto komplexního problému jsou prezentovány průběžně a přílohy obsahují detailnější a názornější ukázky.
Effects of electrical discharges on the Mars´ atmosphere

The aim of this thesis was to study the effects of electric discharge on the atmosphere simu-lating conditions on Mars. This bachelor thesis is focused on simulation of Mars atmosphere at atmospheric pressure and normal laboratory temperature. Mars's atmosphere was more like Earth in the past. Mars is mostly composed of carbon dioxide, which accounts for more than 90% of Mars' atmosphere. To study the Mars atmosphere simulation, glow discharge generated in the reactor at atmospheric pressure was used at a CO2 flow (100 sccm) and dis-charge current of 20, 30 and 40 mA. A part of the measurements was focused on the influ-ence of hydrogen addition at different flow rates. The measurement time course with differ-ent measurement changes was also recorded. Reaction products were analysed using a pro-ton ion mass spectrometer and a flight time analyser. A high amount of aliphatic hydrocar-bons and their derivates, especially alcohols, was detected as well as several cyclic struc-tures. The main products identified in all experiments with a substantial concentration were molecules of ethen, ethane, ethanol, acetone, pentane, benzene and others.
Application of tissue culture test plates for production of recombinant protein in HEK293 cells; determination of optimal conditions

Efficient production of the recombinant proteins (r-proteins) must be based on previous testing of an expression of a small amount of the r-proteins. This work focuses on optimizing the expression of the r-proteins in 12-well plates. It includes testing of an appropriate speed of shaking, production and transfection volume. It compares all the current testing vessels (it compares a 50-ml centrifugation tube to new tested plates that can substitute the unsuitable tubes). It also compares these new tested plates to production square bottles in order to compare the r-protein expression in the plates to the r-protein expression in the bottles. It monitors effects of carbon dioxide on a number of vital cells, their viability, a relative frequency of positive cells on GFP in various cultivation vessels (plates, tubes, bottles), and pH of HEK 293 cellular cultivation during the 4-day cultivation process as well. On the basis of the results and statistical processing of the results, we have set the optimal agitation speed of 230 rpm for the 12-well plates. We have also set the appropriate production and transfection volume of 2 and 0.5 ml for the 12-well plates. In order to evaluate variables and compare cultivations in all the vessels, the tubes could be substituted by the plates. There is a statistically significant impact of carbon dioxide on the number of cells, their variability, relative frequency of cells (positive on GFP) and pH of the cellular HEK 293 cultivation in the cultivation vessels. There is the strongest r-protein expression in carbon dioxide conditions. The results of this work allow to employ the 12-well plates when we aim to test the expression of the r-proteins in a small amount and in carbon dioxide conditions. On the basis of the findings, the expression of the r-proteins in the 12-well plates and carbon dioxide conditions can substitute the expression of the r-proteins in the production bottle and in carbon dioxide free conditions.

The first part of the thesis provides basic information about carbon dioxide and its production. The second part summarizes sectors that emit the most carbon dioxide. The last part describes the international legislation related to emission allowances and carbon trading.
Diagnostics of plasma generated in the atmosphere simulating Mars

The aim of the diploma thesis was the diagnostics of plasma generated in the atmosphere simulating conditions on Mars. This diploma thesis is focused on the simulation of Mars’ atmosphere at atmospheric pressure and normal laboratory temperature. Due to the similar conditions of Mars' atmosphere with Earth, this planet has been explored in the past as well as up to now. Mars' atmosphere is composed mostly of carbon dioxide, which makes up more than 90 % of Mars' atmosphere. A glow discharge generated in a special reactor at atmospheric pressure at a flow of pure CO2 was used to simulate the atmosphere of Mars. Part of the measurement was performed only in pure CO2 with changing current of 20, 25, 30, 35 and 40 mA. Part of the measurements was focused on the study of the effect of the addition of various gases, such as nitrogen, hydrogen and methane, at changes in their flow rates of 1, 2, 3, 4 and 5 sccm. The products formed in the special reactor were analysed using a mass spectrometer with proton ionization and with a flight time analyser. Optical emission spectrometry was used for plasma diagnostics and composition. Mainly simple aliphatic hydrocarbons, alcohols, aldehydes, and ketones were detected. With increasing flow rates of the individual gases, more complex aromatic compounds with higher molecular weights were formed. Corresponding mass and optical emission spectra were measured simultaneously.
System for Opening a Window Based on the Temperature and CO2 Concentration in the Room

The purpose of this work is to enable an automated maintenance of fresh air in a room. Using automated ventilation, so the smallest possible heat losses occur. The data from sensors of indoor and outdoor temperature, CO2, humidity, and window opening status have been collected and analyzed. Then a suitable algorithm has been selected to accomplish the required values. The program also contains an algorithm for predicting the system's behavior so the controlled appliances (window and heating) work together appropriately.

Determination of emissions and harmful particles of exhaust gases of CNG, petrol and diesel vehicles

The thesis deals with an analyses of emission and harmful particles of exhaust fumes of automobiles with an environmental impact. More precisely, it describes the basic characteristics of current fuel types and emission produced by internal combustion engines. Next, it deals with the measures and possibilities of decreasing car traffic emission with respect to environmental impact. It also describes the current European standards for emission and the systems of the cars that are able to decrease emission. The work also deals with the measuring of emission and the methods that are used. According to the measuring methods, the analysis of exhalation is done and it is based on the data of the real car fleet. Values of the emission analyses are compared. The financial aspect is a part of the analyses.
Determination of CO2 exhaust emissions for CNG, petrol and diesel vehicles

The thesis deals with the topic of Carbon dioxide emissions that are created by Petrol, Diesel and CNG vehicles. In the first part of the thesis, the results of theoretical calculations are compared with the values of the manufacturer of the specific vehicle. In the second part, a specific method has been selected for calculations with real data from transport. The savings of CO2 produced by the specific vehicle are expressed in percentage – depending on the fuel used. The operating expenses from the real data are calculated and compared with hybrid and electric vehicles. In the final part of the thesis, the ratio of fleet type vehicles to the financial/ecological aspect was proposed.
