The goal of my work was to introduce the fast pacing field of tissue engineering with focus on bone regeneration. Tissue engineering could be a future alternative to the currently used conventional approaches that suffer from healing failures. Due to increasing demand for bone tissue replacement damaged by degenerative diseases or injuries, many laboratories have attempted to come up with solutions in a form of artificial constructs. In the present light of interest are composite scaffolds usually made of polymer and ceramic combinations. Their main advantage is that they combine elasticity and tensile strength of a polymer with bioactivity and mechanical hardness of a ceramic, while removing drawbacks of each material. Trace elements and Pb isotopes in selected bone samples

The aim of this work is briefly summarize the current theoretical knowledge about trace elements and their inputs into calcified body tissues in a form of search. Here are described mechanisms of their inputs and depositing into human body and principles, which allows maintenance of their optimal levels. In this paper are also described the properties of bones and teeth and simply explained concepts such as an essential and toxic trace elements. The next part of this work is mainly about trace elements, which have the biggest potential to be used in an archeological research, particularly in paleodietary research. These elements are mainly Sr, Ba and Zn. Lead and its isotopes are discussed more here in relation to the effect of Pb to Romans in the past and to the identifying sources of pollution by this element. In the same chapter are also described principles of post-mortem alteration processes, to which are bones after burial exposed and which they have to face during analyzing and interpretations of data related to the amount of trace elements in origin bones.
Ancient-DNA analysis of teeth and skeletal remains with utilization of miniSTR loci.

During the last twenty years, immense progress occurred in the area of analysis of DNA extracted from historical material. Considering the common level of preservation of tissue material, this analysis is usually executed on samples procured from bones and teeth. The analysis of soft mummified tissue is possible only in rare cases. Limiting factor of these analyses is a high degree of degradation and small amount of DNA extractable from this kind of material. First researches concentrated only on short sections of mainly mitochondrial DNA. Today, the analysis of the complete mitochondrial genome of both contemporary and extinct organisms was made possible. In case of analyses conducted on human remains, sections of nuclear DNA are far more valuable, because they can reveal information including not only subject's sex, but also possible kinship between subjects found e.g. in the same grave. Fundamental component of the whole analysis is the process of extracting DNA from cells. Probably every laboratory working with historical DNA uses a differently modified extract protocol. The main requirement for methods of extraction is to secure enough DNA with such a level of purity that would allow its use for following steps of the analysis. Regulation of bone formation using osteogenic supplements in an osteoporotic model

Osteoporosis is a disease of the bone metabolism which is characterised with a decrease of bone substance. The cause of this disease is the imbalance between the creation of a new bone substance by osteoblasts and the resorption of a bone tissue by osteoclasts, in favour of the bone resorption. The risk group of the development of this disease are women after menopause, who naturally register a decline of the estrogen hormone. Estrogen operates as an inhibitor of proosteoclastic factors such as receptor activator of NFκB ligand (RANKL), interleukin (IL)-1, IL-6 or TNF-α. The imbalance of the bone metabolism can also be caused by a disbalance in the production of Prostaglandin E2 (PGE2) and 1α,25-dihydroxyvitamin D3. They are strong mediators which can both stimulate and inhibit an osteoclastogenesis in vitro in concordance with the conditions of the culture/co-culture. This thesis focuses on the examination of an influence of those mediators (PGE2 in the concentration of 10-6 M and 10-8 M; 1α,25-dihydroxyvitamin D3 in the concentration of 10-8 M and 10-9 M) on the osteoclastogenesis from the rat PBMC at the presence of osteoblasts, with or without the combination of proosteoclastic factors macrophage colony-stimulating factor (M-CSF) and RANKL. The diet calcium intake among adolescent girls in aerobic gymnastics

Nutrition with sufficient content of calcium is important for optimal growth and development of the skeleton. Insufficient intake of this mineral during adolescence can have negative effects on bone in the form of not reaching the peak bone mass, the amount of which formed at this time subsequently affects the health of bones during the rest of life. For girls doing aerobic gymnastics, bone health can also be compromised by other factors related to the risks of this aesthetic sport. The main goals of this thesis were to determine the daily dietary calcium intake and the awareness of the importance of calcium in the diet of girls doing aerobic gymnastics. For the research of the girls' knowledge an electronic questionnaire was used and it was completed by 92 respondents at the age of 12-18 years across performance categories of aerobic gymnastics. Based on the answered questions, it was found that the respondents' knowledge of this issue is unsatisfying. To determine the amount of calcium ingested by diet, 4-6 days diets were written by 11 members of aerobic gymnastics representation. After calculation of these diets, the insufficient daily intake of dietary calcium was found and it averaged about half of the recommended values. Biodegradable bones implants based on iron

This work deals with a current topic, which are biodegradable bone implants. These implants seem to be a suitable candidate for temporary fixations, with their gradual decomposition. An important parameter is not only the mechanical properties so that the material can withstand the load, but above all the corrosion properties and the degradation process. This work introduces the physiology of bone, trace elements and also describes current trends in the field of implants. The last part is a theoretical introduction to cross-stage processes. The practical part is focused on the preparation of iron-based samples with various dopants, subsequent annealing and analysis.

This thesis deals with the relationship between bone quality and lifestyle factors with a focus on nutrition, tobacco use, alcohol consumption and physical activity. The theoretical part of the thesis presents general knowledge about bones, their composition, structure, bone division, growth and development of bones. The next two chapters describe the process of bone remodeling and calcium- phosphate metabolism. The work describes in detail the factors influencing the quality of bone mass, especially those that can be affected, such as the use of tobacco products, alcohol, some drugs and sufficient physical activity. A separate chapter is devoted to the influence of diet on the quality of bone mass. The most common disorders of calcium-phosphate metabolism and their treatment are described as well. The last chapter of the practical part is devoted to the examination of bone metabolism. The aim of the practical part of the diploma thesis was to examine the eating habits and other components of the lifestyle (smoking, physical activity, etc.) of patients of Osteocentre, 3rd Medical Department, General Faculty Hospital in Prague and to find out how these components correlate with the results of densitometric examination of the patients.
