Virus interaction with the cellular DNA damage response

Maintenance of genomic integrity is an essential mechanism for every cell. Genomic integrity is disturbed by diverse exogenous or endogenous effects influencing the cell and causing damage of its DNA. Cellular mechanisms capable of fixing these disturbances in structure or sequence are indispensable because damaged genetic information can later cause expression of damaged proteins or inaccurate segregation of chromosomes to daughter cells. Therefore, many effective mechanisms for fixing wide range of types of DNA damage have evolved. This thesis focuses mainly on the eucaryotic MRN complex, which plays an important role in detection and repair of double strand breaks. Many viral families try to block these cellular repair mechanisms because they are activated soon after viral infection. One of the reasons for their activation is the resemblance of some viral genomes to the cellular DNA with double strand breaks. Thus, in many cases, the cell ends up inhibiting the life cycle of the virus by attempting to repair viral genomes. The role of the minor patient in the decision-making process regarding the provided health care

The bachelor's thesis entitled "The role of the minor patient in the decision-making process about the provided health care" deals with the issue of the rights of child patients and their respect. The work tries to outline the issue of providing health care to minor patients, its pitfalls and possible solutions to related problems. The aim of the work was to map the knowledge of minor patients in the area of their rights in the provision of health care and to evaluate their respect by health workers. The theoretical part of the thesis provides a basic overview of legal sources that regulate the role of minor patients in the provision of health care. This part of the work further outlines selected topics of the given issue from the point of view of valid legislation and points out possible pitfalls in practice. The empirical part presents the results of own work and their comparison with the results of other researches. The research method was a questionnaire of our own construction, which was distributed in the outpatient clinic of a general practitioner for children and adolescents. In the study itself were included 92 duly completed questionnaires. Legal presumption of paternity determination vs. the material truth Abstract This work discusses the relationship between paternity determined by one of the legal presumptions of paternity determination, as defined by the Civil Code, and the material truth, or the possible contradiction between them, which the nature of the legal regulation allows. In the Czech Republic, the paternity determination is governed by a total of five legal presumptions, which are based on the probability of paternity determined by them - a first presumption testifying to the mother's husband ex lege, its conversion in favor of the second presumption in the case of divorcing marriage, a one-and-a-half presumption consisting of the father's consent to the mother's artificial insemination, a second presumption based on the parent's consent statement before the relevant public authority, and finally, a third presumption based on the fact of the intercourse between the father and the mother at a decisive time, when paternity is determined by a court decision based on the proposal of some of the concerned persons. However, its rapid growth brings regulatory challenges. This paper examines blockchain regulation, focusing on the European Union (EU) framework and comparative analysis with selected jurisdictions around the world. At the outset, the thesis seeks to clarify the basic concepts of blockchain and its regulatory implications. It tries to introduce the issues and frame the most important terms in a non- technical way. The thesis then examines EU legislation, including contemporary and forthcoming secondary regulations such as the Markets in Cryptoassets Regulation (MiCAR), and explores their objectives, challenges and implications for innovation and market participants. Ability of plants to adapt to anthropogenic pollution

Due to global climate change and human interventions, the environment is changing rapidly. This brings many stress factors that organisms must adapt to. One of the adaptive mechanisms currently widely discussed is rapid evolution, i.e., hereditary change based on natural selection occurring at an ecologically relevant pace. This work deals with the synthesis of knowledge about rapid evolution, the methodology of its research, and its role in the adaptive processes of plants to anthropogenic pollution, focusing on heavy metals in soil and artificial light at night. The collected studies indicate that heavy metals exert strong selective pressure that can lead to the separation of a tolerant population from the original within a few generations. However, the significance of artificial light at night as a selective factor for plant evolution has not yet been studied. Although individual studies deal with adaptive evolution in phototactic insects in response to artificial light at night, extensive research has not been conducted here either. Circadian regulation of the glymphatic system and its connection with the development of neurodegenerative diseases

Due to the absence of an active lymphatic system of the brain and the responsible nodal system, it has long been unclear how interstitial metabolic waste products, in particular amyloid-β, tau- protein, α-synuclein and other biomarkers associated with the development of neurodegenerative diseases, are eliminated. A key role in this process is played by the glymphatic system, a complex network of tunnel-shaped perivascular spaces (PVSs) that conduct the flow of cerebrospinal fluid. These spaces are surrounded by astrocytes, responsible for the transport of fluids and the removal of metabolites. Studies suggest that the glymphatic system is subject to an endogenous circadian clock, and there is an established correlation between the volume of perivascular spaces and, with it, the rate of cerebrospinal fluid flow and sleep. Factors affecting the function of the glymphatic system include pulsation of blood vessels, spatial orientation of the body and functionality of aquaporin-4 (AQP4). Employment experiences among people with schizophrenia

This master's thesis deals with the issue of work experiences and employment of individuals with schizophrenia. Research indicates that engagement in work activities can have a positive impact on the course of the illness, but employment is significantly more challenging for individuals with schizophrenia compared to the general population. Although most individuals with schizophrenia are interested in being employed, only a small proportion of them succeed in the long term. In the Czech context, this is a relatively unexplored issue, therefore there is a lack of knowledge about how people with schizophrenia experience their employment and what difficulties they face. The aim of this master's thesis is to further explore the work experiences of individuals with schizophrenia and to determine what specific factors they perceive as supportive or limiting for their involvement in paid employment. The theoretical part of the thesis is divided into two main chapters, with the first one focusing on schizophrenia itself including its symptoms, prognosis, diagnosis, and the impacts of stigma. Updated summary of Tertiary relics in the vicinity of Pilsen

The aim of the bachelor thesis is to consolidate the knowledge from previous research on Tertiary relics in the vicinity of Pilsen, especially in the districts of Pilsen-City and Pilsen-North. Emphasis is placed mainly on sites that are important in terms of their extent or have provided identifiable samples of plant macrofossils or pollen in the past. In particular, these are the sites Dobříč/Jarov, Ejpovice/Kyšice and Horní Bříza - Modrý kříž. To a lesser extent, these are summaries of the manifestations of volcanism in the defined area. The thesis will be a possible starting point for a follow-up thesis, in which the student will process new findings of plant macrofossils from his own field research. Its further content will be a critical comparison of the individual reconstructions of the palaeochannel network and the determination of the approximate flow direction both for individual relics and the determination of the dominant drainage direction of the entire area.
Synthesis and evaluation of potential antitubercular agents based on nitro aromates

The goal of the master's thesis was the synthesis and assessment of potential molecules against tuberculosis based on aromatic compounds containing nitro groups. The thesis reviews tuberculosis and its therapy, in the theoretical part, followed by well-know and new nitro group containing antibiotics. The experimental part reports multitep synthesis from parent compounds - substituted benzoic acid, pyruvic acid and variously substituted anilines, which were evaluated for their antimycobacterial activity. The core structure of the prepared compound molecules is N-phenyl-2-[2-(3,5- dinitrobenzoyl)hydrazinylidene]propanamide, which was variously substituted on the benzene ring of N-phenylpropanamide. In case of four compounds, one nitro group was switched to the trifluoromethyl group. Yields varied from 7 to 72 % and their minimum inhibitory concentrations (MIC) ranged from 4 to more than 1000 µmol/l. N-(4-Bromphenyl)-2-[2-(3,5- dinitrobenzoyl)hydrazinylidene]propanamide proved to be the most effective derivate. Nursing Care of High Flow Oxygen Therapy Patients

The objective of the bachelor's thesis is to describe the nursing care and its specifics in patients receiving high flow oxygen therapy (HFNO). The theoretical section of the thesis focuses on the general oxygen therapy and explores its application options and usage principles. This section discusses indications and contraindications of the oxygen therapy and its basic types. Specifically, it focuses on the high flow oxygen therapy, including its mechanism or therapeutical advantages and disadvantages. Moreover, it discusses the indications and contraindications of the high flow nasal oxygen treatment (HFNO) and describes the conditions in which patients received oxygen therapy using the high flow nasal oxygen device. The final part of the theoretical section deals with the most prevalent shared symptom, its diagnostics and treatment. The practical section of the thesis presents several case studies, using specific data relevant to each case. As a result, nursing anamneses and nursing diagnoses were formulated.
