A study of the possibility of waste pastries using for the bioproduction of selected metabolites

The aim of this diploma thesis was to study the possibility of using waste bread to bioproduction of selected metabolites. As bakery waste was used waste bread that came from coffee-house “Zastávka”. Bread was pre-treated by grinding into small particles and then it was made to form 15% w/v suspension, which was subjected to enzymatic hydrolysis. For the hydrolysis has been used the -amylase for liquefaction of the substrate and that was followed by a glucoamylase which sacharificated the substrate. There have been several methods of hydrolysis from which was chosen the optimal method for pre-treatment of the substrate prior to fermentation. The effectivity and a process of hydrolysis were determined spectrophotometrically by Somogyi-Nelson method. Final yields of glucose from hydrolysis were determined by HPLC method. Enzymatic hydrolysis was followed by fermentation, which was carried out in two ways, namely by adjusting the pH of the hydrolyzate to pH 5, and without pH adjustment. During the fermentation was carried out sampling in which was determined the content of glucose, glycerol and ethanol by HPLC method. The yeasts Saccharomyces cerevisiae were used for the fermentation which was performed at 30 °C. High yield of glucose was achieved by hydrolysis in two steps (70,28 gl-1), but for the fermentation was used mixed hydrolysis (second method of mixed hydrolysis) with yield 67,94 gl-1. High ethanol yield was achieved during fermentation without treatment pH, namely 31,5 gl-1.
Characterization of selected yeast strains from the environment

The aim of this bachelor thesis deals with identification type-yeasts, which were obtained from the Culture Collection SAV in Bratislava. The purpose is extension of yeast database in laboratory of Faculty of Chemistry, which is used by identification yeasts in samples taken from food or environment. Identification of yeasts was implemented by method PCR-RFLP. Obtained segment is located in area 5,8S-ITS and is defined by using primers ITS1 and ITS4. Restriction analysis was carried out by restriction endonucleasis HaeIII, HinfI, HhaI and TaqI. Chosen species were also tested for presence of extracellular lipasis. In this part of characterisation was used SpiritBlue Agar. The presence of blue colour is lost by activity of lipasis.
Optimalization of PCR-RFLP method for taxonomy of yeasts

This thesis deal with optimalization method PCR – RFLP for taxonomy enlistment of yeasts. Conventional identification methods for yeasts are time-consuming. Molecular biological method based on PCR are instrumental towards fast and precise identification as compared to conventional phenotypic methods. In this thesis molecular biological method PCR – RFLP was used for identification and enlistment of yeasts. This metod follow repeating spacers of ribozomal DNA of yeast, characteristic for each species and strain. By the help of PCR were amplified specific partitions of DNA. These fragments of DNA were split by restriction endonucleases and identified by horizontal electroforesis. In background of this thesis there are information about yeasts, their taxonomy and molecular biological methods.
Comparison of pasta manufactoring in Europe and Asia

This bachelor thesis deals with differences between manufacturing technologies of European and Asian noodles. First part concentrates on wheat as a basic raw material for noodle production. Structure and chemical composition of wheat grain, growing conditions, Triticum durum are described. Next part considers wheat flour, the most important product made of wheat, its manufacturing, storage, defects, and kinds. Semolina made of Triticum durum wheat is mentioned. In more detail it concentrates on classification and manufacturing technology of pastes booth in Europe and Asia. Further it considers other raw materials used and also additives that are used in order to improve nutritional value, color, and taste of final products. In conclusion there is a summary of differences among raw material and the additives used in pasta and noodle production. Lastly the manufacturing technology comparison of European pastes and Asian noodles is done.
Importance of probiotics in child's nutrition

With great advances in science, medicine and knowledge of a man at the high level of treatment. However, even today the humanity seeking to return to its roots - the nature and use it to maintain its good health. The term probiotics is meant by living organisms that are capable of resettling in the human intestine and have a positive impact on maintaining healthy intestinal microorganisms, which also reflected the positive effects on human health in general. Company Valosun a.s. provided for testing probiotický food supplement for children, which was tested mothers with children preschool age and pregnant. The aim of our sociological survey was to determine what is the awareness of the issue of probiotics in this group, the perception of the queried the availability of probiotics and what are the most common reasons for their use.
Identification of yeasts from interspecific varieties of grapes

The main goal of my diploma thesis was to identify and characterize yeasts from must Hibernal and also collection yeasts by using methods called RFLP-PCR (Restriction Fragment Length Polymorphism - Polymerase Chain Reaction). Theory was the first part of my diploma thesis which dealt with wine, yeasts and molecular methods. Theory section was followed by experimental section divided into two parts. The main goal of the first part was to characterize and identify yeasts from must Hibernal by using PCR and RFLP-PCR methods. In the samples there were found yeasts Saccharomyces and Pichia. The second experimental part of my diploma thesis had a goal to extend the database of new yeasts using the same methods mentioned in the first part of experimental section.
Grape must and its changes during the fermentation process

This bachelor thesis deals with chemical changes of grape must during fermentation process. The theoretical part describes history of viticulture, characteristics of vine and varieties analyzed in this work. There are discussed the morphological and chemical composition of grapes in the theoretical part and also basic technological steps of wine production. Another chapter is devoted to the principles of methods for chemical analysis of wine. In the experimental part, monitoring results of changes in chemical parameters of musts during fermentation are discussed. The measured parameters in this work were reducing sugars, pH, titratable acid and ethanol. The analyzed musts were obtained from Sauvignon and Pinot Noir varieties, which originated from integrated and organic vineyards.
Occurance of urinary stones in the CR and simulation of conditions of their rise.

The aim of this diploma thesis was identification of urinary concrements by the infrared spectroscopy and study of conditions of formation of the most frequent urinary stones (calcium oxalates) by the thermodynamic simulation. The theoretical part of this study includes basic information about problems with urolithiasis and technique of identification of urinary concrements. Important part has been devoted to the thermodynamic simulation of formation of calcium oxalate concrements and on this simulation were determinated the conditions for formation of urinary stones. The experimental part contains analysis of real urinary concrements by the infrared spectroscopy and statistical evaluation of the file of 50 149 samples, which were analysed in years 1994 - 2010.
Dietary fiber in food and possibilities of its determination

The work deals with the definition of dietary fiber, its division and basic describtion of building dietary fiber components. Finally, it focuses on basic methods and ways of determining of dietary fiber. The practical part compares the obtained amount of insoluble dietary fibre in different kinds of gluten-free and gluten bread. The results were obtained by using the method according to Henneberg and Stohmann.
Monitoring of the influence of using indigenous yeasts for wine production in the conditions of winery

This thesis deals with identification of yeasts by applying the RFLP-PCR method. Objective of the thesis was to identify the yeasts present in wine from Grüner Veltliner during fermentation. Identification was made by amplification of 5,8S-ITS sequences of DNA by the polymerase chain reaction with primers ITS1 and ITS4. Amplified DNA was submitted to the restriction analysis by restriction endonuclease HaeIII, HinfI and HhaI. By restriction analysis with a specific enzyme, the amplified DNA is chopped into the specific fragments which are characteristic for given kind of yeasts. In the analysed wine, the dominance of autochthonal yeast Saccharomyces cerevisiae was confirmed throughout fermentation. The other identified yeasts in the wine were of kind Pichia. The second part of the thesis was to expand the database by characterization of 28 type-yeasts, using RFLP-PCR analysis. To compare the genetic similarity, program BioNumerics was used, which processed the results of UPGMA cluster analysis using Jaccard´s coefficients.
