CÍL PRÁCE Cílem této diplomové práce bylo detekovat a kvantifikovat změny endoteliální exprese VCAM-1 a endoglinu ve stěně cévy u apoE deficientních myší, kterým byla podávána standardní laboratorní dieta. Dále byl sledován vliv podávaného hypolipidemika atorvastatinu na změnu v expresi těchto endoteliálních markerů. Pro zobrazení exprese VCAM-1 a endoglinu byly využity imunohistochemické metody a ke kvantifikaci jejich exprese stereologické metody.
Clinical diagnosis of benign and malign prostate diseases.

Increased incidence of benign prostatic hyperplasia (BPH) has been associated with age of men. The quality of lives gets worse in men with BPH. Adenocarcinoma of the prostate (CaP) is the most common malignant disease in aged men, younger men as well. In 2005 the incidence of prostate cancer has exceeded the number of occurrence of lung cancer in the Czech Republic. Prostate specific antigen (PSA) has found widespread usefulness in diagnosis of prostate cancer and BPH, too. PSA is organ-specific marker only, therefore, new options (modify PSA, new markers) have been searched to improve CaP detection. Histological estimation of prostate biopsy specimens validates the diagnosis of CaP. Gleason score (GS) determining histology of biopsy tissue affect treatment strategy. Frequently GS of biopsy can be varying GS of final preparation after radical prostatectomy. Therefore, we analyzed the correlation between the prostate biopsy GS and GS of final preparation in patients undergoing radical prostatectomy. Almost half of cases in this group of 88 patients treated in The Clinic of urology in Faculty Hospital of Hradec Králové was identical to GS of prostatectomy specimens and same part of cases was undergraded. Wound repair and factors influencing healing in veterinary clinical medicine II.

Healing of skin injuries is a complex physiological process that involves interaction of large numbers of activated cellular and enzyme systems. It follows a relatively simple scheme; however, the system control is complicated. The length and quality of healing is influenced by outer and inner factors resulting from the knowledge of physiology and pathology of injury healing. Healing of skin injuries is divided into stages that often time overlap. However, at certain stages the healing processes reach their tops and they become for that period characteristic. MDOCTM (polyanhydroglucoronic acid) is used in the clinical practice mainly to speed up the haemostasis. This diploma work is concerned with its effect in two types of remedial products, i.e. gel and nano textile prepared in accordance with the original patented method. The work focuses on how the products influence healing of injuries with 29 animals (1 cat, 21 dogs and 4 horses). In detail it describes the effect on healing processes of horses. Effect of intravenously applicated lactoferine on early faze; of myocardial ischemia II. Biochemical examinations

Lactoferrin (Lf) is an 80-kDa iron-binding glykoprotein. Lf is present physiologically in exocrine secretions, eg. tears, saliva, milk, sinovial fluid, seminal fluid and in the secondary granules of neutophils. The precise function of Lf in organism is considered to be very complex and it is still a hot subject of scientific disputation. Lf was documented to act as antimicrobial, antiinflamatory and antitumoral agent. Another of its properties is ability to inhibit hydroxyl radical formation via Fenton reaction. Afinity of Lf to iron is about 260times higher than that of transferrin. Isoproterenol (ISO) has been used as a model compound to induce infarct-like lesions in the rat and various other animal species. Administration of necrotic dose of ISO caused ischaemia followed by damage of the myocardium. Ischaemia alters iron homeostasis and redox-active free iron, which catalyses ROS-generation. The only possibility for myocardial tissue recovery in acute moycardial infarction represents the reperfusion of ischemic myocardium. But the whole process is associated with damage of myocardium due to burst of hydroxyl radical catalyzed by free iron (Fenton reaction). Transgenic mouse: Experimental model for study of acute lymphoblastic leukemia.

The TEL and AML1 genes occur in a markedly high numer of different aberrations in heamatological malignancies. We studied TEL/AML hybride gene in transgenic mice model, translocation t(12;21)(p13;q22). This hybride gene was searched by PCR methode in 76 individuals. After, they were examined for presence of mRNA of fusion gen TEL/AML1 using RQ-RT-PCR. Compared the blood cell analysis of transgenic and non-transgenic mice, we searched difference in the erytrocyte, lymphocyte, trombocyte count and lymphocyte ratio in peripheral blood. The TEL/AML1 positive Acute lymphoblastic leukemia is B cell precursor leukemia, therefor we used cytofloumetric analysis for study the ratio of the B lymphocytes of transgenic and non-transgenic mice.Then, we observed the leukemic cell marker of pre-B lymphocytes (B220+ /CD43+ ). Presence of the pre-B lymphocytes in peripheral blood show incomplete differenciation and proliferation of B lymphocytes, cosequently leucemogenesis.
Epidemiological trends of treatment benign prostate hyperplasia.

Epidemiological trends of treatment benign prostate hyperplasia Background: The purpose of the work was to follow up trends of treatment benign prostate hyperplasia (BPH) in Czech Republic. The aim of second part was to review the registered alpha-blockers and finasterides in Czech Republic from 1997 to 2008. The last part compares the expenses for transurethral resection of the prostate (TURP), transvesical prostatectomy (TVPE) and the annual treatment with tamsulosin in Slovakia, Poland, Germany, Austria and the United States of America. Methods: The first part is based on data provided by the Urological Clinic of Faculty hospital in Hradec Králové for the decade 1997 - 2007. An increase was observed of the number of patients and the medical therapy and a fall of the surgical therapy of TURP and TVPE. Financial charges were based on informations from foreign clinics and articles. Results: The number of patients with BPH increased from 1624 in 2003 to 2140 in 2007. There were on average 2491 prescriptions per year and the number of patient's visits was 2,12. TURP was practised more on the clinic. The percentage of patients with TURP decreased from 11,76 % to 5,47 % in years 2003 to 2007. TVPE fell from 1,24 % in 2006 to 0,56 % in 2007. The influence of intravenously administeres lactoferrin in early phase of coronnary thrombosis - histopathological analysis

Lactoferin (La) is an 80-kDa iron-binding glykoprotein. It shares a high degree of homology at amino acid sequence level and also the three dimensional conformation level with transferin. Afinity of Lf to iron is about 260times higher than that of transferin. Lf is present physiologically in exocrine secretions, eg.: tears, saliva, milk, sinovial fluid, seminal fluid and in the secondary granules of neutophils. The precise function of Lf in organism is considered to be very complex and is still a hot subject of scientific disputation. Lf was documented to act as antimicrobial, antiinflamatory and antitumoral agent. These effects are probably based on its iron chelating properties. Similarly La inhibits hydroxyl radical formation via Fenton reaction due to chelation of free iron. The catecholamine model of myocardial injury, represented by administration of synthetic catecholamine isoprenaline (ISO), posseses many pathophysiological similarities with acute myocardial infarction (AMI). The only possibility for myocardial tissue recovery in AMI represents the reperfusion of ischaemic myocardium. But this process is associated with tissue derangement due to burst of hydroxyl radical catalyzed by free iron (Fenton reaction).
