The Effect of EGF - like Peptides on Cumulus-Oocyte Complexes
Blaha, Milan ; Němcová, Lucie (advisor) ; Krylov, Vladimír (referee)
In mammals, the ovarian cycle is controlled by follicle-stimulating hormone (FSH) and luteinizing hormone (LH), secreted by the anterior lobe of the pituitary gland. FSH is assumed to be essential for follicle growth. LH is required for the terminal phases of follicle growth, estrogen synthesis, ovulation, development of the corpus luteum and progesterone synthesis. Although cumulus cells and oocytes lack LH-receptors, LH causes cumulus expansion and meiosis resumption. The effect of LH is mediated via EGF-like peptides - amphiregulin (AREG), epiregulin (EREG) and betacellulin (BTC). The precursors of these peptides are present in the mural granulosa cells and EGF-like peptides are released by activation of metalloproteases in response to LH-stimulus. The aim of this thesis is to present EGF-like peptides in the wide context of the molecular mechanisms regulating the late phases of folliculogenesis and meiotic maturation of oocytes.
Signaling pathways and genes regulating gonadotropin-induced maturation of porcine oocytes and cumulus expansion
Blaha, Milan ; Němcová, Lucie (advisor) ; Král, Jiří (referee)
In vitro, meotic maturation of porcine oocytes and cumulus expansion are induced by FSH and EGF-like peptides AREG and EREG. FSH and EGF-like peptides induce expression of cumulus expansion-related genes (HAS2, PTGS2 and TNFAIP6). To define signaling pathways that control FSH- and AREG-induced cumulus expansion, porcine cumulus-oocyte complexes were treated with specific protein kinase inhibitors. Inhibitors of MAPK3/1, MAPK14 and ERBB1 significantly reduced both FSH- and AREG-induced expression of HAS2, PTGS2 and TNFAIP6. These inhibitors decreased FSH/LH-induced expression of AREG and EREG in mural granulosa cells. Surprisingly, inhibitor of PKA had no effect on AREG expression in cumulus-oocyte complexes but the inhibitor decreased expression of TNFAIP6 induced by AREG. Inhibitor of PI3K increased expression levels of AREG and PTGS2 but EREG, HAS2 and TNFAIP6 were reduced. Expression levels of the cumulus expansion-related genes were not affected by an analog of cGMP (8-CPT-cGMP). However, 8-CPT-cGMP blocked spontaneous in vitro meiotic maturation of porcine oocytes and its effect was abolished by FSH. Key words: cumulus expansion, cumulus expansion-related genes, meotic maturation, FSH, amphiregulin, cGMP
The process of addiction treatment in the fellowship of Alcoholics Anonymous
Němcová, Lucie ; Grygar, Jakub (advisor) ; Hájek, Martin (referee)
This thesis concerns an alternative approach to the alcohol-addiction treatment within the fellowship of Alcoholics Anonymous and highlights the importance of self-help groups in the contemporary society. This worldwide biggest self-help group uses sociologically important processes to achieve a new way of life connected with sobriety. I have studied these processes through the participant observation. Alcoholics Anonymous influences its members through its specific culture which reflects in group rituals, high level of group affiliation, spirituality and co-constructed community story, which bears the member's knowledge. This thesis looks on the transition to identity of the sober alcoholic and the new way of living according to Twelve-Step program of Alcoholics Anonymous through the perspective of drift model of conversion, transition rituals or principles used in the narrative therapy. Within these processes there occurs radical reconstruction of perceptual, cognitive and behavioral schemes which influence the meanings in the interpretation of everyday life and consecutively change the identity of the individual and his social action. These changes of the individual are strengthened by the group members and then gradually spread out of the group boundaries too.
Transformation of the Public Discourse on Euthanasia between 2008 and 2020
Žáková, Klára ; Tučková, Anna (advisor) ; Němcová, Lucie (referee)
Klára Žáková, Proměna veřejného diskurzu o eutanázii mezi lety 2008 a 2020 The theoretical part of this work will be an interpretation of used terms and a presentation of arguments for and against euthanasia with the help of research of academic publications and articles. The analytical part of the work will be qualitative research, where I will perform a discursive analysis of the legalization of euthanasia in the Czech Republic. It will focus on the change of approaches during the years 2008-2020, when the draft law on legalization took place and their subsequent nonacceptance. The analysis will focus on the media image of this situation, the actors who enter into this issue and their attitudes. The primary source will be the media archive Newton Media, which will help map the media space on this topic in the Czech Republic.
Freedom or safety on the internet? Framing analysis of two social movements.
Lacina, Ondřej ; Frič, Pavol (advisor) ; Němcová, Lucie (referee)
The aim of this bachelor's thesis is to describe the mobilization processes of two social movements that arise in response to the current state of the Czech cyberspace. The theoretical part of the work presents the conceptualization of the mobilization processes of social movements. It also builds on the definition of the social movement from the Italian sociologist Mario Diani (1992). Finally, it presents key framing tasks based on framing theory from David A. Snow and Robert D. Benford et al. The methodological part of the work presents an introduction to the research method which is a qualitative content analysis of media documents produced by the movements. The analytical part describes each framing process that was used by the social movements. Keywords: mobilization, social movement, counter-movement, framing analysis, framing processes
Optimization Models in Energy Production
Němcová, Lucie ; Bednář, Josef (referee) ; Popela, Pavel (advisor)
This bachelor's thesis focuses on optimization of a boiler's size of a waste incineration plant from an economic point of view. The problem was approximated by discretization because random parameters with normal distribution occured in the incineration plant model. To evaluate the quality of such approximation there were created confidence intervals using random sample information, independent random number streams method and common random number streams method. Used methods were compared and the smallest number of scenarios that aproximates the problem "well enough" was proposed in the last part of this thesis.
The development of the discourse of right-wing populism in the Czech Republic
Němcová, Lucie ; Císař, Ondřej (advisor) ; Čada, Karel (referee)
The thesis examines the development of right-wing populist discourse in the Czech Republic and aims to define the forms and possible changes in populist discourse with a special emphasis on argumentation, then to interpret the changes in a broader social and political context. The research focuses on three organizational incarnations of right-wing populism: Public Affairs, Dawn of Direct Democracy - Tomio Okamura and Freedom and Direct Democracy - Tomio Okamura. The research method is a discursive-historical approach, which draws on the analysis of topoi. An analysis of the programs of the two movements and the party showed that the right-wing populist discourse has shifted from the original themes that the Public Affairs party has raised. These are included in the other two movements, but each adds something extra. From the initial anti-elite attitudes, social chauvinism and emphasis on direct democracy in the case of Public Affairs to nationalism and the promotion of national pride in the case of the Dawn, to the harsh euroscepticism, Islamophobia and anti-immigration appeals in the case of the SPD. The topoi used in the VV program is primarily topos of abuse, but the Dawn, with its specific appeal to national awareness, also introduces topos of people or topos of advantage, and the SPD clearly...
Corporate culture as a factor of motivation
NĚMCOVÁ, Lucie
The main aim of this thesis is to specify issues of a corporate culture, while focusing on its cultural dimensions and a possible motivation factor in the direction of the knowledge economy. This thesis compares the current situation and the motivational potential of corporate culture in two selected companies. The theoretical part describes basic concepts, procedures and opinions of individual authors on corporate culture, motivation and knowledge economy issues. For research of corporate culture in chosen companies two surveys are used: The Value Survey Module 94 & 2013 (VSM) and Color test of semantic differential (TBSD). The research has identified which dominant cultural dimensions are motivating in selected organizations. Proposed measures that can change current state are based on the own analysis and they are given in conclusion.
The Importance of the Social Therapeutic Workshop for Integrating Clients in their Future Activity in the Sheltered Workshop
NĚMCOVÁ, Lucie
This Bachelor´s thesis deals with the importance of the social therapeutic workshop for integrating clients in their future activity in the sheltered workshop (sheltered workplace). The main objective is to map necessary work skills of clients for their transfer to the sheltered workshop (sheltered workplace). The thesis consists of the theoretical part, which contains literary information, and the research part where the established objectives are being evaluated. The chapter named "Current State" is focussed on the target group of persons in the social therapeutic workshop and the sheltered workshop (sheltered workplace), and further there is mentioned the importance of work and self-fulfilment for people with the health handicap. There is described the concept of social services, their characteristics and the proper definition of the social rehabilitation and the social therapeutic workshop. Next chapter deals with the work rehabilitation directed at the sheltered workshop (sheltered workplace). And consecutively, the attention is aimed at the social worker and workers in the sphere of social services. The last chapter is focussed on the family in the context of their care for the child with the health handicap from the point of view of the family, and the cooperation with them from the point of view of professional workers. The research part established the following objective: To identify necessary work habits of clients in the process of the social rehabilitation which they obtain in the social therapeutic workshop. The main research question "What are the necessary work skills of clients for their transfer to the activity in the sheltered workshop?" was further divided into 5 partial research questions. The qualitative research strategy was used for obtaining information. Data were collected by means of the questionnaire, in particular the technique of the semistructured interview. The research sample was made up with the method of the intentional (purpose) selection which defined particular criteria for integration in the research sample. There were made up two research samples involving clients of the social therapeutic workshop and professional leading workers of the social therapeutic workshop and the sheltered workshop (sheltered workplace). The obtained information was processed with the method of coding and consequent categorization. The results are mentioned in the subheads corresponding with the partial research questions. The research shows that clients come to the social therapeutic workshop in order to use their time meaningfully and to train new skills which they can use both at work and in their daily life. The big motivation is also the team and related social integration. The family represents irreplaceable role which includes support in everyday activities and the feeling of security. The professional leading workers consider the social therapeutic workshop to be the very important first stage of the job. They see the advantage of the social therapeutic workshop in providing possibility to map knowledge and skills of individuals who try to move from the social service to the sheltered workshop.
