Inhibitory effect of anthocyanidins on hepatic carbonyl reductase

Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Jitka Hlaváčová Supervisor: Prof. RNDr. Lenka Skálová, Ph.D. Title of diploma thesis: Inhibitory effect of anthocyanidins on hepatic carbonyl reductase Carbonyl reductase (CBR) is a reducing enzyme that plays a significant role in the first phase of biotransformation of both endogenous and exogenous substrates. It participates in detoxification of many xenobiotics, among them important drugs. Differences in expression and activity of hepatic biotransforming enzymes may affect efficiency and safety of treatment. The aim of this work was to compare the activity and expression of CBR in old and young individuals and to define an inhibitory effect of selected anthocyanidins on the activity of CBR in humans and rats and to determine a value of IC50 and a type of inhibition for the strongest inhibitors. Meaningly higher activity of CBR was detected in the senescent rats compared to the young ones and the old rats also showed larger interindividual variability. No significant age- related differences have been identified by measurement of activity CBR in human samples. Anthocyanidins showed a mild to moderate inhibitory effect on CBR. Eva Novotná, Ph.D. Title of diploma thesis: Cloning, expression and purification of selected mycobacterial enzyme Mycobacterium tuberculosis produces many enzymes which are nescessary for its growing and living. Inhibition of enzymes participating in biosynthesis are a potential target of new drugs in treatement of tuberculosis. Naphtoate synthase is an mycobacterial enzyme in biosynthetic pathway of menaquinone. It has a crucial meaning in electron transport chain under anaerobic conditions. Naphtoate synthase belongs to crotonase superfamily, catalyses a conversion of o-succinylbenzoyl-CoA (OSB-CoA) into 1,4-dihydroxy-2-naphtoyl-CoA (DHNA-CoA). Plasmid pET-28b(+) and a segment of DNA encoding the sequence of menB amplified by polymerase chain reaction was used for preparation of recombinant protein menB. Ligated plasmid was transformed into the cells E. coli HB101 by heat shock. Expression cells E. colli BL21 were used for the expression and the expression itself was started by the addition of IPTG. Prepared protein was isolated and purified by a machine Äkta which used the method of affinity chromatography.

Eva Novotná, Ph.D. Title of Diploma Thesis: Identification of new potent inhibitors of selected mycobacterial enzyme Mycobacterium tuberculosis remains one of the deadly pathogens, which are threat for people around the world. The combination of antituberculotic drugs is effective but the treatment is prolonged and sometimes it is not successful because of multiresistant forms of tuberculosis and also co-infection with HIV, that are the problems in last a few years. Therefore, it is necessity to find a new drug against tuberculosis, which will be effective against resistant forms of mycobacterium and can be used for HIV therapy. Enzyme isocitratelyase performs a potentially drug target against Mycobacterium tuberculosis. This enzyme plays a key role of glyoxylate pathway, where catalyzes the conversion of isocitrate to succinate and glyoxylate. This enzyme is necessary for mycobacterial survive during acute phase of the infection. Restoration of vanished roads and railways connetcting Moravia to Lower Austria

The project deals with the restoration of the railway connecting Hevlin and Laa an der Thaya. The renovation of the railway station in Hevlin is divided into individual stages. The first stage consists of conversion of the Art Nouveau expedition building in the museum. In the next stage a platform will be built and adjacent sites will be re-arranged. An important step at this phase is the roof construction for the future expedition building. The roofing will serve for summer cultural events or markets. This new utilization will benefit from free flat space of the platform. The last step is completion of perimeter walls and the building will serve as a railway station. After construction of bridge the railway connection between Hevlin and Laa an der Thaya will be reopened. Another railway between Brno and Vienna will be created as well. The aim of the study is to strengthen relation of neiborougs from South Moravia and LowerAustria.

Petra Matoušková, Ph.D. Title of diploma thesis: Reference gene selection for mRNA quantification in Haemonchus contortus Haemonchus contortus is a parasitic nematode, whose multiresistance to ant- helmintics means global problem, threatening mostly small ruminants farming. Genom and transcriptom have been published in 2013, allowing gene expression studies to be conducted. Reference genes for this studies have not been validated yet. Use of suitable reference genes is essential for accurate normalization of gene expression levels. Aim of this work was to identify and validate potential reference genes for gene expression studies in H. contortus adults. Eleven genes were chosen, stability of their expression was assessed in males and females of two H. contortus strains, one drug-susceptible (ISE) and one multi-drug-resistant (WR). Total RNA was extracted and reverse trancribed to cDNA. cDNA was diluted and analyzed using quantitative real-time PCR with SYBR Green I detection.
