Glombová, Marie ; Komárek, Vladimír (advisor) ; Nevšímalová, Soňa (referee) ; Starý, Jan (referee)
The aim of the study was to evaluate the importance of brain MRI's findings, and modify the criteria for brain MR imaging in NF1 patients according to this data, to improve the quality of life with early detection of important NF1 complications. Description of the whole cohort, with emphasise to possible cause of high range of sporadic NF1 cases. Evaluation the possibility diagnosis or follow up of brain gliomas by plasmatic values of neuron specific enolase (NSE) and S100B protein. Subjects and methods: I analysed data from 285 NF1 children followed up on our department from 1990 to 2010 by the same examination battery. I evaluated the incidence of brain MRI findings, clinical development, age at gliomas manifestation and necessity of treatment. I also described the whole cohort and made statistic analysis of plasmatic values of NSE and S100B protein in NF1 patients, with and without brain gliomas. Results: OPGs were found in 77/285 (27 %) children and GOOPs in 29/285 (10.2 %) of NF1 children, of who 19 had OPG and GOOP together, so the total number of brain glioma was 87/285 (30.5 %). Totally, 43/87 (49.4 %), respectively 43/285 (15.1 %) children with brain glioma were treated, and 4/285 (1.4 %) of this children died. Obstructive hydrocephalus was found in 22/285 (7.7 %) patients and was caused...
The role of main excitatory and inhibitory transmitters in epileptic seizures and the effect of antiepileptic drugs in the immature brain
Liptáková, Soňa ; Mareš, Pavel (advisor) ; Komárek, Vladimír (referee) ; Šlamberová, Romana (referee)
Epilepsy affects 50 million people worldwide. Approximately 50% of epilepsies start in the infancy and childhood. In addition, there are several age-bound epileptic syndromes which are difficult to treat. Due to these facts, developmental studies of epileptic seizures are of primary importance. Predominance of excitation over inhibition is a mechanism of nearly all types of epileptic seizures. Hypothesis that this compromised balance can be normalized by drugs affecting main excitatory and inhibitory system at different stages of brain development was tested in models with different pathogenesis. Our studies demonstrated that seizures induced by derrangement of GABAergic inhibition can be suppressed by antagonists of NMDA receptors (MK-801) and vice versa seizures with glutamatergic pathogenesis can be affected by drugs potentiating GABAergic system (valproate, ganaxolone) at all developmental stages in immature rats. Analysis of site of action of NMDA receptor antagonists demonstrated reciprocal function of anterior and posterior parts of substantia nigra, structure important for spread of epileptic activity to spinal cord. Special attention was given to an age-dependent model - flexion, emprosthotonic seizures elicited by administration of NMDA. Valproic acid, which potentiates GABAergic inhibition,...
Assessment of motor activity in children with autism by inertial sensors - Method of measurement and its utilization in diagnostic
Nývltová, Lucie ; Komárek, Vladimír (advisor) ; Zounková, Irena (referee)
Autism belongs to developmental pervasive disorders. Primary symptoms of autism include failure in communication, social interaction and imagination. Other symptoms are abnormalities in physical activity manifested by delays in motor development, stereotyped movements, postural instability, impaired in imitation and dyspraxia. These symptoms already come up in the first years of life. The first diagnosis of autism used to be made between 2nd and 3rd year of life, however if the diagnosis could be based on physical symptoms, it would be possible to establish the diagnosis earlier. The work presents a method of measurement by inertial sensors that could be used to assess physical activity of autistic children.
Alternative methods in the surgical treatment of mesial temporal lobe epilepsy
Vojtěch, Zdeněk ; Nevšímalová, Soňa (advisor) ; Šonka, Karel (referee) ; Komárek, Vladimír (referee)
Purpose: The aim of the study was to evaluate effectiveness and complications of alternative methods of stereotactic treatment (gamma knife radiosurgery and radiofrequency amygdalohippocampectomy) in mesial temporal lobe epilepsy due to mesial temporal sclerosis. Methods: Both patient groups underwent a standard preoperative epilepsy evaluation. Both procedures were planned according to the individual anatomy of each patient. Between November 1995 and May 1999, 14 patients underwent Leksell Gamma Knife radiosurgical amygdalohippocampectomy with a marginal dose of 18, 20, or 25 Gy to the 50% isodose following. Between April 2004 and October 2009 a group of 51 patients was treated using stereotactic thermo-lesion of the same mesiotemporal structures. Lesions were performed using a string electrode inserted through the occipital approach with a single trajectory. Results: In the radiosurgical group, one patient was classified as Engel Class Ib, three were Engel Class IIc, four were Engel Class IIIa, five were Engel Class IVb and one was Engel class IVc 39 months after treatment. One patient was classified as Engel Class Ib, three were Engel Class IIc, one was Engel Class IIIa, and two were Engel Class IVb in a subgroup of seven patients who were unoperated 2 years prior to the last visit and at least 8...
Hodnocení technologických postupů při pěstování kmínu kořenného s důrazem na sklizňový proces
Komárek, Vojtěch
This diploma thesis Evaluation of technological processes in the cultivation of caraway with an emphasis on the harvesting proces is focused on current situation and importance of caraway growing in Czech republic and in the world, on basic growth factors and work operations (mostly related to harvest) of common caraway. Main part is based on evaluation of two model technological processes. In first process were expenses of total 504,50 Kč.ha-1, in the second process were expenses bigger - 10 618 Kč.ha-1. Furthermore, this thesis was focused on enumerating the harvesting losses. For the purpose of comparability and objectification of the enumeration, two different methods of evaluation were used. By using the first method, 25,81 % losses were measured. Second method provided value of 22,09 %.

Assessment of notor stills in children with Aspuger syndrome and High - functioning autism using MABC-Z.
Nývltová, Lucie ; Komárek, Vladimír (advisor) ; Zounková, Irena (referee)
Asperger syndrome and high-functioning autism is a form of pervasive developmental disorder characterized by impaired social interaction and communication and restricted and stereotyped interests. These children are often clumsy and motor uncoordinated. They also performed worse in standardized tests of motor function. In this diploma thesis we used Movement Assessment Battery for Children 2 (MABC-2) for assessment motor skills in children with Asperger syndrome and high-functioning autism aged 7-10 years in comparison with the group of children developing typically. Our study included 9 children with Asperger syndrome, 3 children with high-functioning autism and 64 children with typical development. Evident motor deficit (below 5. percentile) was found in 2 children with Asperger syndrome and 3 children with high-functioning autism. 1 child with Asperger syndrome was in a risk of motor deficit (6.-16. percentile). Children with Asperger syndrome and high-functioning autism performed significantly worse than children developing typically in a total score of this test. Significant difference was found in two components of MABC-2, in Aiming and catching (AC) and Balance (BAL). Better results reached children with typical development. Results of this study indicate that children with Asperger syndrome...
