Problems of determination of non-traditional organochloric compounds in environmental compartments

The baccalaureate theoretical thesis resolved the current issue of the presence of xenobiotics based on organohalogenated compounds in environmental elements. The focus was on congeners of polychlorinated biphenyls (PCB), polychlorinated diphenilyethers (PBDE), and polychlorinated dibenzodioxines and dibenzofuranes (PCDD/DF). The physical and chemical characteristics of these organically pollutants were described, as well as its toxicological and environmental aspects. The possibilities of its analytical determination were also described. The complete analytical procedure was described for indicator congeners PCB.
Hormones in sewage sludge

Currently there is a spate of interest in the presence of pharmacologically active substances in the environment. These substances are excreted in active or metabolized form and with wastewater pass the wastewater treatment plant. Current treatment technologies do not always eliminate all pharmaceuticals effectively and therefore they enter the environment. One of these active groups is the group of steroid hormones. Steroid hormones belong to the group of endocrine disruptors and they are considered to be dangerous for the ecosystems. Due to the hydrophobic character of steroid hormones they undergo partial or total sorption from wastewater to sludge. Sewage sludge is the by product of wastewater treatment and contains heavy metals, organic contaminants and pathogenic bacteria. A part of produced sewage sludge is used as a fertilizer in the Czech Republic. This issue follows the regulation 347/2016 – conditions of agricultural use of sewage sludge. Waste policy of EU will alter the conditions of sludge disposal, so it is necessary to obtain data about the concentration levels, fate and behaviour of those pollutants. This thesis was focused on five natural female hormones and four synthetic, which are used as a part of contraceptive pills and substitutional hormonal therapy. Determination of hormones was performed in four steps including ultrasonic assisted extraction, clean up by solid phase extraction, derivatization and final analysis by gas chromatography with mass spectrometry on triple quadrupole in MS/MS mode.
Influence of technological processes on the content of priority contaminants in wastewater treatment plants

Water is one of the most abundant substances on Earth and it is an essential part of our lives. Its pollution has an impact on the whole ecosystem. Polybrominated diphenyl ethersare synthetic persistent organic pollutants used as flame retardants in various commercial and household products. This thesis is focused on assesing the degree of elimination of PBDEs in wastewater treatment plants with different technological processes of purification. The samples were collected in the three wastewater treatment plants in the south-moravian region, that means WTP Brno – Modřice, WTP Mikulov and WTP in VFU Brno.
Bisphenol A in water ecosystem

This diploma thesis deals with Bisphenol A, especially with its impact on the aquatic ecosystem. Information about its properties, production and current use are summarized here. Its harmful impact on the environment, especially on the aquatic ecosystem and the human body is characterized. Also the methods of its degradation within the aquatic environment have been described. A summary of the options of a determination of Bisphenol A in water samples is incorporated and the method of gas chromatography with mass spectrometry (GC/TOF MS) and comprehensive two-dimensional gas chromatography with mass spectrometry (GCxGC/TOF-MS) is compared in the experimental part. Analytical determination precedes the isolation of the analyte from the water samples by solid phase extraction (SPE) using SupelcleanTM ENVITM - 18 and derivatization using the silylation reagent, N,O-bis(trimethylsilyl)trifluoroacetamide (BSTFA). The result of this work is the analysis of a series of real samples from wastewater treatment plants Brno Modřice and Luhačovice by a two-dimensional gas chromatography with mass spectrometry (GCxGC/TOF-MS).
Passive sampling of polar organic pollutants from water

This thesis deals with the passive sampling of polar compounds from water using passive samplers POCIS (Polar Organic Chemical Integrative Sampler). These devices were exposed for 4 weeks in the waste water at the inflow of municipal waste water treatment plant in Brno – Modřice, and also at the outflow of the treated water. After exposition sequestered compounds were released by extraction with a mixture of methanol, toluene and dichlormethane. Extracts were analysed by liquid chromatography/mass spectrometry with electrospray ionization. Further, methylated and trimethylsilylated extracts were analysed by orthogonal comprehensive two-dimensional gas chromatography with mass spectrometric detection. Separated compounds were identified on the base of their retention and mass spectra.
Analysis of biochar aqueous extracts by separation methods

This master's thesis deals with the analysis of biochar extracts by separation methods. All analyzed biochar was produced from waterworks sludge by microwave pyrolysis. The aim of the thesis is the optimization of the sample preparation method and its analysis in a laboratory environment. With the ever-growing world population, the problem of a sustainable economy in both agriculture and waste management is becoming increasingly urgent. This fact has led most countries to consider promoting the so-called Circular Economy. The use of sewage sludge as a feedstock for biochar production is perfectly in line with this strategy. So far, however, the short term and the long term benefits and risks of using biochar have not been sufficiently described. Potentially hazardous organic substances were extracted from the biochar extracts by three techniques: liquid-liquid extraction, solid-phase extraction, and solid- phase micro-extraction. The obtained samples were analyzed using the GS-MS/TOF method. The individual methods were compared. Several biochar samples from WWTP Brno and WWTP Drahovice were selected and analyzed to verify the suitability of the selected sample analysis method.

Mycotoxins in Brewing Materials and Beer

The presented thesis deals with the issue of mycotoxins in brewing materials and beer. Attention was devoted mainly to the selected fusarium mycotoxins (deoxynivalenol, zearalenol, T-2 toxin, and HT-2 toxin) ochratoxin A and aflatoxins B1, B2, G1, and G2. The aim of the thesis was to optimize and validate analytical methods for the determination of the above mentioned mycotoxins in the brewing materials and beer. Analytes were separated using high-performance liquid chromatography with mass – spectrometric detection (HPLC-MS/MS) and ultra-performance liquid chromatography with fluorescence detection (UPLC/FLR). These analytical methods were then applied for mapping the occurrence of fusarium mycotoxins in malting barley crops in the Czech Republic and monitoring the level of contamination with mycotoxins in malting and brewing industries. In addition, experiments studying over-foaming of beer were conducted as primary gushing – over-foaming of beer – is connected, similarly as mycotoxins, with the presence of microscopic filamentous fungi in the raw materials for beer production. Studies describing in detail these methods are part of this thesis (Annex I – V). From all published results, it is evident that the occurrence of mycotoxins in cereals including barley is natural and cannot be completely prevented, not even if all conditions of correct agricultural practice are observed. It is known that some mycotoxins present in contaminated malting barley pass to the final product – beer due to their chemical and physical properties. However, the mycotoxin concentrations found do not mean any significant health risk for consumers.
Degradation of Heat Transfer Fluids in Thermal Solar Systems and Propane-1,3-diol as a New Option

Stárnutí teplonosných kapalin na organické bázi je dlouhodobým problémem, který je znám od počátku jejich používání. První část této disertační práce je tak věnována případové studii funkčního experimentálního systému, který byl jako nový naplněn teplonosnou kapalinou na bázi propan-1,2-diol a pozorován po období 7 let. Pro analýzu stárnutí kapaliny v tomto systému byly sledovány základní provozní vlastnosti kapaliny jako jsou hustota, viskozita, teplota tuhnutí, pH a obsah kovů. Skrze tyto vlastnosti tak bylo sledováno stárnutí kapaliny nepřímo. Přímé sledování stárnutí bylo posléze provedeno analýzou degradačních produktů, jako jsou organické kyseliny a změny ve složení směsi pomocí izotachoforézy a hmotnostní spektrometrie. Pro srovnání byly taktéž analyzovány vybrané vzorky z několik dalších systémů plněných identickou kapalinou s prokazatelně pokročilou formou degradace. V druhé části práce jsou představeny základní fyzikálně-chemické vlastnosti směsí propan-1,3-diolu s vodou a jejich analytické hodnocení a matematické modelování pro universální použití jakožto nového základu pro nemrznoucí teplonosné kapaliny. Na základě dostupných informací je pak hodnocena použitelnost této směsi. Výhoda propan-1,3-diolu je spatřována především ve výrobě z obnovitelných zdrojů a v některých fyzikálních a chemických vlastnostech, které dle dosavadních poznatků předčívají doposud používané glykolové směsi.
