The Influencing of Anticholinesterase Activity of 7-methoxytacrine
Novotná, Petra ; Herink, Josef (advisor) ; Vopršalová, Marie (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of biological and medical sciences Candidate: Mgr. Petra Novotná Consultant: doc. MUDr. J. Herink, DrSc. Title of Thesis: The influencing of anticholinesterase activity of 7-methoxytacrine and galantamine by L-carnitine The aim of this work was to test the ability of L-carnitine (KAR) to increase the permeation of indirect parasympatomimetic 7-methoxytacrine (MEOTA) and galantamine (GAL) through the blood-brain barrier. The monitoring of acetylcholinesterase activity (AChE) in the parts of the brain chosen served as marker of changes presumed. All experiments were performed in laboratory rats, drugs tested were given systematically. The modificated Ellman's method for the determination of the AChE activity was used. The principle of this method is based on hydrolysis of acetylthiocholine iodide. The previous administration of KAR to MEOTA augmented the inhibition of AChE more in the frontal cortex, septum and basal ganglias and less in the hipocampus. Intraperitoneal administration of KAR was somewhat stronger in comparison with peroral one. Activity of AChE was more reduced after administration of MEOTA than after administration of GAL. Contemporary administration of KAR with MEOTA reduced AChE activity in the selected...

Eating gabits, body image in younger and middle-school age girl. The contribution to the issue of primary prevention of eating disorders
Novotná, Petra ; Fraňková, Slávka (advisor) ; Niederlová, Markéta (referee) ; Marádová, Eva (referee)
The aims of this study are dietary habits, eating preferenees and aversions, body image and self-esteem, the faetors whieh in its pathologieal form belong to one of the major risk faetors and are eonsidered as a key eomponent of prevention of eating disorders in ehildhood. The analysis of interviews and questionnaire methods for the younger group of girls and middle sehool age girls (N = 10 1) showed that girls enter the 1 st c1asses with relatively eonsistent system of eating preferenees and aversions. The ehanges are in middle sehool age when the range of experienee with meals extends, preferenees are redueed and aversions to eertain food and dishes are growing. Preferenees of slirnness appeared in the area of body image, though it dec1ined with age and moved to a more realistie proportions. Pronouneed pathologieal eating behavior was not eonfirmed in our reference file, we have seen only some trend s that might result in the eating problems in later life. In a group of girls of middle sehool age, we found inereased negative self-esteem and evaluation of own skills. At the end of the work we gaye some suggestions for the prevention of eating disorders.
Creating Memory in Public Broadcasting
Novotná, Petra ; Abu Ghosh, Yasar (advisor) ; Zandlová, Markéta (referee)
In my diploma thesis I describe the process, in which some episodes of the radio program Příběhy 20. století are made and with this example I explain, which factors can influence remembering in a medium of the public sphere and how "lieux de mémoire" occur in its broadcasting. I examine the recording of some of the narratives that can possibly be presented in the radio program, how the interviewers handle the interviewees and what motivation do they have to work in a project like this. I focus on how the stories are picked for the radio program, how are they edited, who takes part in the editing and how is it possible that they are in charge of it. My diploma thesis also includes a detailed description of the social position of Post Bellum organisation and its fundamental project Paměť národa, which are both closely connected to radio program Příběhy 20. století. In the conclusion of my thesis I deal with the fact, that this social position is one of the main elements in the process of memory making in public sphere medium, because it has an indirect impact on it. In my thesis I try to answer, what influence can the examined radio program (as part of institutionalized remembering and media communication) have on social memory.
The Role of the Public Prosecutor in Criminal Proceedings
Novotná, Petra ; Tejnská, Katarína (advisor) ; Šelleng, Dalibor (referee)
The Role of the Public Prosecutor in Criminal Proceedings The diploma thesis deals with the role of the public prosecutor in criminal proceedings. The public prosecutor has an irreplaceable role in criminal proceedings, and he has statutory powers in all stages of criminal proceedings. The aim of this thesis is to provide an overview of the activities of the public prosecutor in criminal proceedings and to make an analysis of two selected areas. This thesis consists of five parts, which are further divided into chapters. The first part deals with the historical development of public prosecution in our territory between 1918 and 1993. It provides a brief overview of the fundamental change that public prosecution has undergone in this period. The second part is dedicated to the general characteristics of the Public Prosecutor's Office. The first chapter focuses on the constitutional definition of the Public Prosecutor's Office. The next chapter deals with the description of the Public Prosecutor's Office and the method of filling the position of the public prosecutor, part of the chapter describes the issue of supervision within the Public Prosecutor's Office. The last chapter provides an overview of the public prosecutor's powers outside the area of criminal proceedings. The third part focuses on...
Multiple Instance Learning Framework Used For Ecg Premature Contraction Localization
Novotna, Petra
We propose the model combining convolutional neural network with multiple instancelearning in order to localize the premature atrial contraction and premature ventricular contraction. The model is based on ResNet architecture modified for 1D signal processing. Model was trainedon China Physiological Signal Challenge 2018 database extended by manually labeled ground truthpositions of premature complexes. The presented method did not reach satisfying results in PAClocalization (with dice = 0.127 for avg-pooling implementation). On the other hand, results of localizationof PVCs were comparable with other published studies (with dice = 0.952 for avg-poolingimplementation).
Atrial Fibrillation Classification Using Deep Convolution Networks
Novotna, Petra
We propose the usage of three deep convolutional neural networks architectures for classification of a single lead electrocardiogram (ECG) recordings and evaluate them on the atrial fibrillation (AFIB) classification, for which data set was provided by the Department of Biomedical Engineering, BUT. The compared networks are based on ResNet, VGG net and AlexNet. Single lead signals are transformed into the form of spectrogram. AFIB data was augmented for the purpose of similar size of both respected classes and for successful classification. The most successful architecture, based on AlexNet, was found to perform obtaining an accuracy of 92 % and F1 score of 56 % on the hidden testing set.
The role of PKN family kinases in cancer
Novotná, Petra ; Rösel, Daniel (advisor) ; Ramaniuk, Volha (referee)
This bachelor thesis is focused on the PKN family of Ser/Thr kinases. This family includes three isoforms PKN1, PKN2 and PKN3. Especially it deals with the kinase PKN3 in more detail. These are kinases related to protein kinase C, belonging to the AGC superfamily. PKN kinases are activated via small G proteins of the Rho GTPase family or unsaturated fatty acids. PKN kinases are involved in many cellular processes, such as the regulation of cytoskeletal rearrangements, affect cell adhesion, cell movement, embryonic development and the cell cycle. Expression of PKN3 is particularly increased in cancer cells but is only present in small amounts in normal body cells. Therefore, PKN3 appears to be a very interesting therapeutic target for the treatment of cancer. Studies have shown that PKN3 has a significant effect on the motility of cancer cells, thus contributing to their migration and ability to form metastases.
