Preparation and characterization of GlnB22-insulin, a naturally occurring mutant in diabetic patients

Diabetes mellitus is a widespread disease which can be diagnosed in all age groups from newborns to seniors. Diabetes has multiple causes. The final disorder of blood glucose regulation is caused by a complex of factors and agents, in many cases by genetic defects connected with an unhealthy lifestyle. Moreover, a recently discovered form of diabetes, referred to as the monogenic diabetes, results from a gene mutation whereby only a single gene defect causes the onset of disease. The gene encoding the insulin primary structure belongs to several fundamental genes. Each change in a peptide chain is unique and leads to variation in this disease. Diagnosing patients with this mutation results in more detailed specification of their treatment and often also in the improvement of their living conditions. Mutations of the insulin gene have various symptoms. Some of the changes lead to the neonatal diabetes and patients are fully addicted to insulin since their birth. Other variations will never break out at their carriers. Seriousness of the disease course depends on characteristics of an original amino acid, its function in the insulin biosynthesis, its interaction with the insulin receptor, and on characteristics of a newly inserted amino acid and its influence on natural processes. Music as an Effective Means in Teaching English Pronunciation

The aim of this thesis is to point out the beneficial effect music has on learning English pronunciation. The theoretical part describes how music can positively influence the process of learning and helps to create an effective educational environment. This work further focuses on the motivational effect of music and on its ability to increase student interest. The practical part provides the reader with concrete examples of the methodology used in singing classes at the basic art schools with focus on selected features of English pronunciation. Synthesis of Trichloroacetyl Esters of Aldols

This diploma thesis describes synthesis of trichloroacetyl esters of aldols which were obtained by a reaction of trichloroacetic acid with formaldehyde and another comercial aldehyde. Used were hydratopaldehyde, isobutyraldehyde, 2-methylpentanal, 2-ethylhexanal and 2-ethylbutanal. Some of these esters were described in the literature but only in part, several of them are new compounds. Excellent yields and clean products prove that the applied method is a good way to achieve these compounds that can be easily used for further processes.
The Journuy Toward Freedom. Inspiration by Saint John of the Cross.

The Bachelor thesis with the title The journey toward freedom: Inspired by Saint John of the Cross sets oneself a goal to inform readers about spirituality and teaching of Saint John of the Cross. It tries to interdigitate two the most famous writings of Saint John of the Cross - it is the book The Ascent of the Mount Carmel and the book The Dark Night - with the Bible. The topic of the freedom is the main topic of the Bachelor thesis, especially the process of liberation the man from something and toward something. In the first part of the Bachelor thesis the reader gets to know the basic informations about life of Saint John of the Cross; about roots of Saint John ́s teaching; what is the foundation of the man and at the same time what is the aim of the one' s life and for which he is calling. Next what is the cause for necesarry free oneself. In the next chapters the reader learns about the important symbols, counsels and rules of Saint John' s teaching and toward which he is inviting the man with his teaching. The second part of the Bachelor thesis deals in detail with individual parts of the process of liberation leads through the "dark night" all the way to the "transcendental union" the soul with the God, ergo to the "absolute freedom". The aim of this work is to analyse the current system of motivation of employees in the chosen company following suggestions of changes and recommendations for improving management in this area. The theoretical part contents basic ideas of motivation and employee benefits. The practical part is based on an analysis of the company. The purpose of changes should be a more sophisticated and long-term motivation of employees. Proposals for better solutions in the field of employee motivation should eliminate mistakes of managers which they commit on their employees.

The financial crisis is a phenomenon that affected almost the whole world in 2008. This thesis deals with issues of the financial crisis in the Czech Republic. The thesis is divided into several parts. In the first part the financial crisis, its causes, and its course till affecting the Czech Republic is characterized. Furthermore, I assess the impact of the crises on the Czech economy and its development in 2008. In the next part I evaluate the impact of the crisis on the Czech investors in 2008 and I focus on the scope of the Czech National Bank. In the last part I think about solving financial crisis and I think about it, whether the financial crisis has a positive impact on the economy of the State.

Significant peak broadening was observed for some N-methylated peptides as a consequence of cis/trans isomerism of the peptide bond. Chromatographic behavior of the N-methylated peptides was compared with data obtained by NMR.
