Encapsulation of some enzymes into organic particles

This bachelor thesis is focused on encapsulation of some enzymes into organic particles. The theoretical part is aimed to enzymes, their general characteristics, structure, activity and use, and to encapsulation techniques. Some methods used for analysis of particles, namely the determination of the distribution of particle size using dynamic light scattering, and determining the stability of the particles by the zeta potential, are described too. The experimental part describes methods of preparing particles, methods of determining the encapsulation efficiency and methods for determining particle size and stability. During the experiment part four types of enzymes were encapsulated, partly by manual preparation and partly by encapsulator. Encapsulation efficiency, size and stability of prepared particles were determined too. The particles were exposed to artificial intestinal, gastric and bile juices, as well as the effect of model foods. Subsequently, their long-term stability was observed.
Application of Zymomonas mobilis for bioethanol production from cellulose materials

The thesis deals with the use of Zymomonas mobilis to produce bioethanol from waste paper. There were used three types of paper made by various manufacturers to optimize the production of bioethanol. Individual papers were subjected to the same adjustments namely mechanical grinding and subsequent enzymatic hydrolysis. During cultivation of Zymomonas mobilis in different hydrolysates were taken samples for determination of bioethanol production. The analysis was carried out on high-performance liquid chromatography. Production of bioethanol and glucose consumption was evaluated as well as production of lactic acid since it was also noticeable on the chromatograms.
Effect of physical exercise on mood changes

Aim of paper is to find out what effects do physical exercises have on the psychical part of the person. Theoretical part speaks about psychology and emotions from the psychical view, and also from the biological view. It speaks about the effect of sport, group mentality and the way people in a group perceive the sport. It also mentions biological processes in the body and the substances that are made during the physical exertion. Methodical parts speaks about how we will put out hypothesis to the test. That is how, where, how long and under what circumstances will we do our experiement, and how will we evaluate it. Conclution is made of results, which containt tables and graphs with collected data and their explanation in text, evaluation and personal comments. Annexes contain the prototype of the used questionnaire. Results showed, that theoretical hypothesis, that physical activities are beneficial for a human, and statistical hypothesis both proved correct, and statistical one in 83,3% of the cases.
Porous layer open tubular columns with immobilized trypsin for protein digestion

We have developed a monolithic porous layer open tubular (PLOT) column with immobilized trypsin for protein digestion. The PLOT column was prepared in a 10 (mikro)m ID fused silica capillary. Trypsin was immobilized on the monolithic surface and the developed enzyme reactor was used for protein digestion followed by on-line ESI/MS analysis.
Utilization of the additives in farm animal nutrition

This thesis deals with the use of additives in livestock nutrition, the distribution of these substances as well as their characteristics. The work is focuses on characteristics of a selected group of zootechnical additives. In this group of substances each specific substance added to feed are presented and it also describes their impact and importance in livestock nutrition.
Hydrolysis of potatoes

This work compares acid, alkaline and enzymatic hydrolysis of potato starch with simultaneous application of various pressures (0MPa, 0,3MPa and 1MPa). Acid hydrolysis was made by sulphuric acid of intensity 3, 6, 9 and 12 %, alkaline hydrolysis was made by sodium hydroxide of intensity 3, 6, 9 and 12 % and four enzymatic preparations for starch hydrolysis were used for enzymatic hydrolysis. Hydrolysis effect was measured via GE and added by alcohol yield. There was positive influence of increased pressure 0,3MPa and 1Mpa on hydrolysis process visible in this experiment.
Hydrolysis of corn silage

Aim work is compare yield GE at acid, alkalic and enzymatic hydrolysis cellulose phytomass at different acidity (H2SO4, 3, 6, 9 and 12%), fundamentals (NaOH, 3, 6, 9 and 12%) and while using different enzymatic preparations (6 preparations to hydrolysis celluloses and hemicelluloses). Further is assessed positive influence compressive extrusion (0,3MPa and 1MPa).
Vybrané fyziologické parametry žížaly .i.Dendrobaena mrazeki./i. (Černosvitov, 1935)

.i.Dendrobaena mrazeki./i. shows preferences for xerothermic habitats, i.e. behaviour unusual for Central European earthworms. Several ecophysiological parameters of .i.D. mrazeki./i. were measured as a basis for further investigation of its environmental adaptations. pH about 7 prevailed in the gut of the species. The profile of digestive enzymes included all important saccharolytic enzymes and did not differ from those of other earthworms indicating no exclusivity in .i.D. mrazeki./i. feeding habits. Melting and supercooling points of body fluid were similar to those of relative species .i.D. octaedra./i.. The adaptation of .i.D. mrazeki./i. to xerothermic habitats may result from its desiccation tolerance, ability of quiescence and/or from its partly endogeic mode of life. Quiescent adult individuals were found to survive about 60 days at 15 % soil moisture.
