SUMMARY Studying the pathogenesis of insulin resistance and the role of PKC in insulin resistance In HHTg rats, elevated serum triglycerides and FFA were associated with the ectopic accumulation of triglycerides in tissues and reduced insulin sensitivity of peripheral tissues. Impaired glucose utilization in the peripheral tissues was associated with the reduced activity of GS in skeletal muscle. Decreased GS activity and glucose utilization in peripheral tissues indicate a possible defect in insulin signal transduction. In line with this, our results show that skeletal muscle IR was associated with the increased activation and translocation of PKC θ. Nutritionally induced obesity of HHTg rats resulted, in many cases, in the further deterioration of metabolic abnormalities associated with IR. We found that PKC θ, in particular, could contribute to the metabolic abnormalities associated with IR and obesity. The age-related increase in IR and deterioration of some parametres of carbohydrate and lipid metabolism, were not associated, in HHTg rats, with obesity but with increased serum levels of triglycerides and FFA. Title: Study of inclusive electron-positron pair production in collisions of Ar + KC1 at 1.756 AGeV Author: Mgr. Filip Kfi'zek, Ph.D. Abstract: Spectroscopy of di-leptons emerging in decays of the light vector mesons př, CD, and ^ is a promising way how to investigate changes of properties of these hadrons in the surrounding nuclear environment. This thesis deals with an analysis of inclusive e"V pair emission from. Ar+KCl collisions at a kinetic beam energy of 1.756 AGeV. The measurement was carried out using the High Acceptance Di-Electron Spectrometer (HADES). HADES is a unique apparatus dedicated to study e+e~ pair production in hot and dense hadronic matter. After an introduction to the field, the HADES spectrometer is briefly described. Then we focus on the analysis of the Ar+KCl run. We describe and discuss event selection performed by the on-line trigger, lepton identification, and the subsequent pair analysis. Efficiency corrected spectra of parrs are compared with predictions of a thermal model based Monte Carlo event generator (Pluto) and, further, with forecasts of a microscopic transport code (HSD). Forbidden Oxygen line emission from the traces of meteors

The aim of this diploma thesis was to study forbidden green oxygen line emission. It was found out that green line emit mainly in heights 100-110 kilometers, exceptionally below 90 kilometers. Time evolution of green line intensity with respect of heights was investigated in detail. There was suggested theoretical dependency and agreement with observed data was fulfilled. Dependency of integral intensity was investigated as well. Mechanisms of excitation and quenching of green line in emission was proposed in previous studies in 70. of previous century. This work discuss with its results. Efficiency of transformation of kinetic energy of meteoroid to forbidden green line's emission was discussed. The concentrations of atomic oxygen was estimated by several way in heights of green line emission.

Utilization of molecular markers in oil seed rape breeding programmes

Current breeding of oilseed rape is focused on breeding of F1 hybrids and the male sterility and self-incompatibility could play the significant role in hybrid breeding programmes. Plant breeders also more widely utilize molecular genetic techniques for selection of desirable plants/genotypes. Molecular methods are faster, more reliable and more specific than conventional ones, which are based mainly on morphological descriptors. The aim of my thesis was to optimize PCR analysis methods and to develop specific molecular markers for target plant selection in oilseed rape hybrid breeding programmes. The new marker for detection of fertility restoration gene (Rf) in CMS Ogu-INRA plants was tested. New primer pair RsPPRF2/RsPPRR2 was designed in the coding region of PPR-B protein, which participates in the restoration of fertility in CMS Ogu-INRA. Also newly designed primers BrSLGIIF/BrSLGIIR were tested in SI plants. The optimal annealing temperaturse of these primers was 58 °C. But amplification in some SC plants was also observed. The optimization of the PCR reaction was performed for all designed primers. The set of F1 SI hybrids created by crossing of two lines with different S II haplotypes was tested by using of the PCR-RFLP technique for detection of polymorphism in amplified fragments.

Coeliac disease and gluten-free diet

Co liac disease is an auto-immune disease, which means that the body produces antibodies that attack its own tissues. For people with coeliac disease this attack is triggered by gluten, a protein found in wheat, rye and barley. This gluten-intolerance causes an inflammatory response that damages the gut. When the gut lining is damaged, the body cannot absorb a11 the n cessary nutrients from food. Because of this lack of nutrients, people with undiagnosed coeliac disease can have a wide range of symptoms and can suffer from nu tritional deficiencies. The symptoms are v ry individual but can include stomach and bowel symptoms, tiredness, anaemia, diarrhoea, abdominal discomfort, weight loss and vomiting, so it can take some hrne before an accu rate diagnosis is sought. Both adults and children can have this disease. There is evidence to suggest that the p revalence of coeliac disease is 1 in 200-300 people in The Czech republic. The most people with this disease is found in Europe and USA, in Africa and A ia the disease occurs very far ly. The only treatment is to be on a gluten-free diet for alllife. Software jako služba na českém trhu

Tato diplomová práce pojednává o tématice poskytování aplikací formou Software as a Service, neboli software jako služba. Mezi hlavní cíle tohoto textu patří především vymezení tématiky SaaS, popis současného stavu a předpokládaných trendů a v neposlední řadě též poskytnutí návodu, jak postupovat při vstupu na trh SaaS s jednoduchou aplikací. Přidruženým cílem je snaha ukázat, že i nerozsáhlá on-line aplikace může odpovědět na aktuální poptávku. Metodou pro splnění stanovených cílů bylo především detailní studium současných trendů a získání praktických zkušeností ze samotné problematiky SaaS. Celou praktickou část této diplomové práce považuji za klíčovou, neboť předkládá postup, jakým jsem já (s několika společníky) postupoval při přípravách na poskytování on-line aplikace formou SaaS. Správnost tohoto postupu ovšem doopravdy ověří až čas. Přínos celé diplomové práce vidím především v tom, že se nevěnuje poskytování robustních informačních systémů. Naopak je zaměřena na poskytování jednoduché on-line aplikace, která může odpovědět na aktuální poptávku. O této tématice jsem nenašel téměř žádnou kvalitní literaturu. Struktura diplomové práce je transparentní ? v teoretické části podávám výklad o problematice SaaS a souvisejících tématech. V praktické části se potom věnuji právě dokončovanému podnikatelskému záměru, který spočívá v poskytování on-line aplikace v modelu software jako služba. V závěru diplomové práce hodnotím dosažení stanovených cílů a předkládám návrhy na praktické využití tohoto textu.

Oriented immobilization of PNgase F on a porous polymer monolith for rapid N-glycan release

We demonstrate a simple and rapid method for the oriented immobilization of peptide-N4-(Nacetyl- glucosaminyl) asparagine amidase F (PNGase F) on a porous polymer monolith. The oriented immobilization is based on the affinity of glutathione-S-transferase (GST) tagged PNGase F towards glutathione modified monolith prepared in the capillary format. This approach allows the oriented and easily replaceable immobilization of PNGase F for rapid and efficient release of N-linked glycans. The reactor was tested by deglycosylation of several native glycoproteins such as ribonuclease B, fetuin or human immunoglobulin G. The proteins were effectively deglycosylated in several minutes and the released N-linked glycans were analyzed using off-line MALDI/MS or CE/LIF.

Be stars are a group of hot, massive, emission-line stars characterized by intense radiatively-driven stellar wind and extremely rapid rotation. The typical emission in the Balmer series arises in a thin equatorial disc around the star. The detailed structure and most of all origin of the disc are still unknown. The aim of this thesis is to give a thorough review of the main directions of research into the eld. We describe observational features that set Be stars apart from the rest of the B spectral class: Balmer emission, linear polarization, infrared excess and superionized resonance lines in the far UV range; also main physical characteristics of the underlying star, and variability. We discuss the so-called Be phenomenon - the unknown mechanism creating the emission-producing circumstellar disc in Be stars, and providine an overview of the most successful models attempting to explain the Be phenomenon. Finally we discuss the rotation law of the disc and its importance in placing a physical constraint on possible models for Be stars, and apply one of the examined rotation parameter nding methods.

The ubiquitin–proteasome system is involved in the regulation of activity of spermadhesin aqn1 and acrosin inhibitor, the two sperm surface proteins, during porcine fertilization

The spermadhesin AQN1and acrosin inhibitor (AI/SPINK2) proteins bind to the sperm plasma membrane at ejaculation. The AQN1 has been implicated in sperm binding to zona pellucida (ZP) of the oocyte as well as in sperm interactions with the epithelium of the oviductal sperm reservoir. The SPINK2 protects spermatozoa from proteolytic degradation during their trip up the female genital tract toward the oocyte. This study examined the role of two components of the 19S proteasome regulatory complex, the ubiquitin C-terminal hydrolase UCHL3 and PSMD8 in the AQN1-mediated boar sperm binding to zona pellucida. Interaction of PSMD4 subunit with the acrosomal surface-associated acrosin inhibitor AI/SPINK2 provided another line of evidence for the presence of 26S proteasomes on the sperm surface. Detection of the ubiquitinated forms of SPINK2 supports the hypothesis that SPINK2 activity is controlled by ubiquitin-proteasome system (UPS). The activity of the porcine AQN1, and thus the efficiency of sperm-oocyte recognition/binding, may be controlled by elements of the sperm surface-bound UPS, in particular by UCHL3, and by proteasomal regulatory complex subunit PSMD8. Ubiquitinated isoforms of AQN1 were also detected in boar sperm extracts. The UCHL inhibitor ubiquitin aldehyde and the antibodies against UCHL3 or PSMD8 increased the rate of sperm-ZP penetration and polyspermy during porcine in vitro fertilization (IVF). In contrast, the addition of recombinant UCHL3 to fertilization medium significantly reduced polyspermy rates, while maintaining satisfactory rate of monospermic fertilization (~50%). These results are significant for production agriculture. The high level of polyspermy that hinders porcine IVF for commercial embryo transfer could be mitigated by the modulation of the UCHL3 and/or PSMD8 activity.
