Miroslava Mayerová Title of the Bachelor's Thesis: Art group 12/15 and its place in the second half of the 20th century Czech art Abstract This thesis focuses on the evolution and influence of art group 12/15 on socio-cultural development of society in the second half of the 20th century. The theoretical part encompasses the socio-cultural environment and historical situation before and during the formation of the group, describes its formation, and presents the work of individual group members until their second joint exhibition. The didactic part teaches students the group's activities by utilising concepts used by 12/15. The practical part of the work carries out didactic task and by form of self-assess evaluates its feasibility and usefulness. The work includes historical documents and articles about group.
Study of effect of flavonoids on biotransformation enzymes expression

Flavonoids belong to the group of phenolic compounds included in the secondary metabolites of plants, which are represented mainly in fruits and vegetables. These compounds have antioxidant and anti-carcinogenic effects. Some of them can affect biotransformation enzymes, which include the cytochrome P450 and can interfere with the metabolism of xenobiotics. In the present work we studied effect of dihydromyricetine on the gene expression of cytochrome P450 1A1, 1A2 and cytochrome b5 in livers, lungs, kidneys, colon and small intestine of laboratory rats. The small intestine was divided into three parts namely a proximal, middle and distal. At first, isolation of total RNA was made using commercial reagents and isolated RNA was converted to cDNA by reverse transcription. Finally, using polymerase chain reaction in real time (RT-PCR), the relatively gene expression of genes observed in the organs of laboratory rats pretreated dihydromyricetine and control laboratory rats (untreated) to a reference gene β-actin was determined. It was found that dihydromyricetine did not significantly affect the gene expression of studied genes in most organs. Theatre as a Window to the World of Mental Disability

The thesis provides a qualitative analysis of an autobiographical play based on journals written by a mentally challenged woman. The analysis is founded on authentic personal experience, the script of the play and a video recording of its performance. The empirical section is preceded by a theoretical introduction focused on mental disability in relation to theatre. The empirical section begins with the question: What can we learn about a mentally disabled person through theatre? The analytical process consists of three levels: a content analysis of the performance, an analysis of the author's perspective and the viewpoint of an implied spectator. The analysis shows that the story contains common autobiographical topics and the author barely reflects her disability. The creative perspective consists of the process of adapting the story into a play. Theatre also plays a therapeutic role and raises awareness. There is an imaginary window on the boundary between the stage and the audience, one that the author opens from within and the viewer from without. During the course of the play, the implied spectator becomes confronted with their prejudice and an accumulation of ambivalent feelings. This process ends in a catharsis and a genuine encounter with the actor's world and her spontaneity. KEY WORDS...
The use virus derived vectors for the expression of proteins in plants

The tobacco mosaic virus (TMV) is one of the most studied virus. Its properties (simplicity, self assembly of viral capsid, the ability to proliferate in a large variety of hosts) offers a lot of applications in various scientific or biological fields. Gene vectors based on the tobacco mosaic virus represent a very good way for expressing proteins in plants. Because the tobacco mosaic virus is well researched and a large number of his properties have already been discovered, it is used for nanotechnologic sectors in this period. Not only for the formation of nanoparticles, but also for example for a potential production of vaccines in plants. In this paper I summarize the known information about the process of self assembly of tobacco mosaic virus and the possibility of using this virus in the emerging fields of biology. (eng): Over the past few decades, plant viruses and vectors derived from them have become not only an integral part of the work in laboratories, but also began to be used commercially for the production of vaccines, drugs, enzymes and other proteins / peptides. One of the most commonly used viruses for the expression of heterologous proteins is tobacco mosaic virus. In this work, I would like to focus on plant viruses and vectors derived therefrom. Compare it with other expression systems used in plants and describe how their benefits could be further used and combined with modern methods of cloning. The expression profile of cathepsin L in developmental stages of Fascioloides magna

Our experimental organism Fascioloides magna is a digenetic liver fluke from Fasciolidae family which parasitizes in domestic and free-living ruminants of North America and Central Europe including Czech Republic. In Czech Republic this highly pathogenic worm causes a severe liver damage to cervids and bovids and the prevalence locally reaches up to 95%. The biology of F. magna including e.g. the characteristics of host-parasite molecular interaction and the functions of particular molecules produced by the parasite are not fully understood. According to results of our previous research the excretory-secretory products of F. magna adults contain number of molecules which play the crucial role in host tissue invasion, digestion and evasion of the host immune response. One of the most abundant is cysteine peptidase cathepsin L (FmCL). FmCL is supposed to play various key roles in biological processes of all stages during a life cycle and therefore we can suppose its different expression level in particular life stages. In order to define the expression level of FmCL we performed the pilot study with miracidia and adults where the qPCR method was applied. Eva Novotná, Ph.D. Title of diploma thesis: Cloning, expression and purification of mycobacterial dihydrofolate reductase Dihydrofolate reductase is an enzyme essential for the metabolism of folic acid - it catalyzes the reduction of dihydrofolate to tetrahydrofolate. Tetrahydrofolate is an important cofactor involved in one-cabron transfer reactions. Dihydrofolate reductase plays a key role in the synthesis of DNA, RNA and proteins. Dihydrofolate reductase was also found in M. tuberculosis. This bacterium is the most common causative agent of tuberculosis in humans. Thus dihydrofolate reductase could be a potential target for the design of new antituberculotics. The recombinant protein dihydrofolate reductase was prepared in several steps. The coding sequence of the protein was first amplified by polymerase chain reaction. A recombinant plasmid, obtained by the ligation of an amplified segment of DNA with plasmid pET-28b(+), was transformed into competent cells E. coli strain BH101 by the heat shock method. Stability of the catalytic domain of neutral trehalase studied by differential scanning fluorimetry

This bachelor thesis is part of a project aiming for the crystallization of trehalase domain of neutral trehalase Nth1 from Saccharomyces cerevisiae. The main goal of this thesis is to optimize the purification protocol of two different constructs of yeast Nth1 and determine the optimal buffer for crystallography of these constructs using the differential scanning fluorimetry DSF. Trehalases are important, highly conserved enzymes found in many organisms. These enzymes belong to the class of hydrolases, subgroup of glycosidases and their common feature is hydrolytic cleavage of trehalose molecule into two glucose subunits. Trehalose is a naturally occurring non-reducing disaccharide which serve in yeast cells as storage carbohydrate and energy and stress metabolite. Trehalose and its hydrolysis is very important in the life cycle of the insect, as it is present as the main sugar component of insect hemolymph, so some trehalase inhibitors could be used as insecticides. By this time, however, only one structure of trehalase from a prokaryotic organism Escherichia coli was solved. For this reason it is very important to solve the structure of trehalase from a eukaryotic organism. Aesthetic Factor in John Dewey's Conception of an Experience Aesthetic Relevance demonstrated by Example of Snowbording

This diploma thesis examines John Dewey's concept of experience, focusing especially on his notion that an aesthetic factor is present in every complete and consummatory experience. It deals with the differences between a common experience and "an experience" and examines how an aesthetic factor works in them. The indispensable role of the aesthetic factor is intentionally demonstrated on the non-artistic example of experiences of snowboarding. The most important findings are developed through the approach of Richard Shusterman's somaesthetics. The theories of these two philosophers support the main idea of this thesis, namely that dynamic movements of the body can bring aesthetic experiences to their actor. Thus, some of the reasons that explain the attractiveness of this modern sport come to light that are not apparent at first glance.
