Significance of detection of human papillomaviruses in draining lymph nodes in cervical cancer paients
Sláma, Jiří ; Dundr, Pavel (advisor) ; Svoboda, Bohuslav (referee) ; Špaček, Jiří (referee)
Background: Metastatic involvement of pelvic nodes is the most important prognostic parameter in early-stage cervical cancer. Still, approximately 15% of patients with negative pelvic nodes experience recurrence, majority of them in the pelvis. The presence of HPV DNA in histology-negative pelvic nodes is considered a subclinical metastatic spread. Methods: Patients with early-stage cervical cancer referred for surgical treatment were enrolled in the study. Cytobrush technique was used for sample collection from the fresh tissue to avoid any loss of material for histology. Results: Altogether, 49 patients were enrolled in the study. High-risk (HR) HPV was identified in the tumor in 91.8% patients, and in the sentinel node or other pelvic nodes in 49.9% patients. Among the 10 HR HPV genotypes detected, HPV 16 was the most frequently represented in both the tumor and the lymph nodes (66.7% and 71.4%, respectively). All metastatic lymph nodes were HPV positive. Conclusion: The presence of HR HPV DNA in a sentinel node had a positive predictive value for metastatic involvement of pelvic lymph nodes in our study. This could be considered a sign of an early subclinical metastatic spread; however, the prognostic value has to be evaluated through a longer follow-up.

Filippi, Tomáš ; Bruckner, Tomáš (advisor) ; Palovská, Helena (referee)
This study deals with the value of ICT services. Enterprise Informatics is a widespread line of business and it is part of every company. There are not only software and hardware resources in enterprise informatics, but also people belong into and work with informatics every day. Currently, there are new trends how to manage enterprise informatics. Based on the same principles as for the business. A prerequisite for effective management is accurate overview of the costs and benefits. For this purpose there is a group of metrics which ensure measuring the amount of inputs, outputs and quality of services. The value of ICT services should be positive. Benefits should be greater than costs. The aim of this work is to create a model for monitoring the value of enterprise Informatics. Model simulate the dynamics of enterprise informatics and describe all important parameters in terms of value. Model should also find out if there are some external or internal factors, which could bring on some random element complicating the process of effective management.

Further differences were found in the hurdles run technique of each other probands. Keywords: Hurdle, run, technique, kinematic analysis

Photochromic effect in nanoporous titanium dioxide film with Ag nanocrystals
Galář, Pavel ; Malý, Petr (advisor) ; Dohnalová, Kateřina (referee)
The diploma thesis is devoted to multicolour photochromic effect in nanocomposite material Ag-TiO2, which is caused by a spectrally selective photoinduced decrease in extinction. The Thesis introduces briefly the theory of particle plasmons which are of fundamental significance for the investigated phenomenon. A short summary of the models and results published on photochromic effect in Ag-TiO2 is than given. The core of thesis deals with the optimization of this nanocomposite material towards a high photochromatic response. The influence of concentration of the TiO2 solution and of the speed of its spin coating deposition on a substrate was measured. Also the effect of AgNO3 quality used for sample preparation was monitored. A change in the extinction was found to depend on parameters of the excitation laser beam too. As a part of the optimization process, also the alternative TiO2 matrixes produced on basis of lamellar micelles were tested. The photochromic effect was monitored by measurements of the light extinction and scattering in dependence on the sample photoexposition. The properties of a set of the high-quality samples were studied using photoluminescence spectroscopy. Measurements of the photoluminescence decay with subpicosecond time resolution by up-conversion technique as well as of the...

Salabová, Markéta ; Šatínský, Dalibor (advisor) ; Chocholouš, Petr (referee)
SEPARATION AND DETERMINATION OF BETAMETHASONE AND CHLORAMPHENICOL IN MULTICOMPONENT PHARMACEUTICAL PREPARATIONS USING A MONOLITHIC COLUMNS COUPLED TO SEQUENTIAL INJECTION SYSTEM Markéta Salabová Abstract A new separation method based on a reversed-phase sequential injection chromatography (SIC) technique was used for simultaneous determination of chloramphenicol and betamethasone in pharmaceutical eye drops in this contribution. Short monolithic column coupled with a sequential injection analysis (SIA) system enabled separation of two compounds in one step. A Chromolith Flash RP-18e, 25-4.6 mm column with 5 mm precolumn (Merck, Germany) and a FIAlab 3000 system (USA) with a 6-port selection valve and 5 ml syringe were used for sequential injection chromatographic separations in study. The mobile phase used was acetonitrile-water (30:80, v/v), flow rate 0.48 ml min-1 ; UV detection was at two wavelengths 241 nm and 278 nm, (absorption maximum of betamethasone and chloramphenicol). The basic validation parameters showed good results: linearity of determination for both compounds including internal standard (propylparaben) > 0.999; repeatability of determination (RSD) in the range 0.8 - 1.7% at two different concentration levels, and detection limits in the range 0.5 - 1.0 µg ml-1 . The chromatographic...

Detection of Differential Item Functioning with Non-Linear Regression: Non-IRT Approach Accounting for Guessing
Drabinová, Adéla ; Martinková, Patrícia
In this article, we present a new method for estimation of Item Response Function and for detection of uniform and non-uniform Differential Item Functioning (DIF) in dichotomous items based on Non-Linear Regression (NLR). Proposed method extends Logistic Regression (LR) procedure by including pseudoguessing parameter. NLR technique is compared to LR procedure and Lord’s and Raju’s statistics for three-parameter Item Response Theory (IRT) models in simulation study based on Graduate Management Admission Test. NLR shows superiority in power at low rejection rate over IRT methods and outperforms LR procedure in power for case of uniform DIF detection. Our research suggests that the newly proposed non-IRT procedure is an attractive and user friendly approach to DIF detection.
