The mechanism of cell death induction in human malignant melanoma in vitro

Melanoma is a complex malignant disease characterized by increasing incidence and mortality. Melanoma biology is complicated and comprises many aberrant genes and signaling pathways. Treatment of melanoma relies on surgery, the role of radiation therapy and chemotherapy are limited. In advanced disseminating and metastasizing melanoma chemotherapy largely fails and that is why new approaches are being sought including identification of deregulated molecules or processes which could be used as targets. Some of these targets center on frequently aberrant cell death regulating mechanisms. Some traditional cytostatic drugs such as inhibitors of topoisomerases although useful in other types tumors so far proved useless in treatment of melanoma. Nevertheless, expression of topoisomerases in melanoma is often high, thereby enabling at least theoretically their use as targets for chemotherapeutical intervention. Aim of this study was to assess the cytoxicity and proapoptotic effect of campthotecin (topoisomerase I inhibitor), etoposide (topoisomerase IIα inhibitor) and their combination in selected human melanoma cell lines. Comparison of driving comfort between Cars of Professional Racing Drivers and Normal Drivers

Work title: The discomfort experienced by racing drivers when driving in comparison to the discomfort experienced by ordinary drivers ofpassenger cars. Work objectives: The objective of this theses is actual detection of discomfort and comparison of the reaction time of professional racing drivers with drivers of passenger vehicles when driving and mutual comparison of their results. By using subjective and objective methods we will monitor and record how discomfort is manifested by individuals, how it influences their complexly dealing with movement manoeuvres and how it changes in time during the course of driving.. Method: Three professional racing drivers and a sample of randomly chosen subjects ranging in age from 20-28 years were tested in a reaction and decision making test, requiring reactions to visual impulses from the upper and lower limbs. Additional tests evaluated the discomfort ofthe test subjects and the current mental state ofthe individua!. Intensive Care of the Paediatric Invasive Meningococal Infection

The thesis deals with the issue of the pediatrie invasive meningococcal infection. Theoretical part is focused on the deseription of meningococcal infection, its incidence, epidemiology, clinical picture, complication and consequences. diagnosis, therapeutic and preventions procedures. Also there is an emphasis on early detection and prompt treatment of sepsis shock. The nursing section of the thesis includes comprehensive care of a seven-month-oldinfant. This is a deseriptive report which deseribes the dynamic course of the disease, aggressive treatment including antibiotics, steroids, respiratory support, circulation support, renal replacement therapy and overall therapy of disseminative intravascular coagulation syndrome. Through that care the child survived without any mutilation. The case report focused on the needs of the child and its mother in nursing care. It elaborates the main nursing diagnoses, their targets, plans and realizations.

Title: The use of altitude training in Olympic cycle 2004-2008 (in the example of rowing) Aim: The operational objective of this work is to provide a summary of selected published information on the issue of altitude training, and its use in rowing. The main aim of this work will be the practical verification of the theory of recovery training at higher altitude, focusing on the example of rowing through the evaluation of biochemical analysis. Another aim will be to determine whether training at higher altitude might have positively affected the performance of rowers in the Olympic cycle 2004-2008. Methodology of work: Based on the knowledge gained from a literature review on the matter we are planning to organize training camps at altitude in the four-year Olympic cycle. According to those tasks and objectives each year in the annual cycle, we chose the suitability of training camp at altitude. During the training camp at altitude as we are use biochemical analysis to test the use of training at altitude in practice. The results: The results of monitoring the internal environment and the hematology using biochemici analysis showed that altitude is limiting for training.
