A comparison of central and peripheral effect of galantamine.
Galusová, Petra ; Vopršalová, Marie (referee) ; Herink, Josef (advisor)
Galantamine is a reversible inhibitor of acety1cholinesterase (AchE). It is recommended for the treatment of rnild to moderate Alzheimer's dementia, vascular dementia and dementia with Lewy bodies. Galantamine is characterized as cognitive enhancer with dual mechanism ofaction (it has also shown activity in modulating the nicotinic cholinergic receptor). The aim of this work is to determine the effective inhibitory dose of galantamine on the selected route of administration and comparing its central and peripheral effects of galantamine. Determination to proceed on the basis of the fmdings of AchE activity in the central nervous system (in the frontal cortex, hippocampus, septum, basal ganglia and pituitary gland) and on the contrary peripheral nervous system activity of butyry1cholinesterase has been detected (specifically in the liver and plasma). The results obtained by the Ellman's method showed that galantamine is generally a weak inhibitor and statistically significant effect is found only at the highest dose used. At the highest dose used can be expected adverse effects of peripheral type.

Study of the relations between the structure and function of C - terminal vanilloid receptor TRPV1
Gryčová, Lenka ; Obšil, Tomáš (advisor) ; Heřman, Petr (referee) ; Urbánková, Eva (referee) ; Krůšek, Jan (referee)
Transient receptor potential channel vanilloid receptor subunit 1 (TRPV1) is a thermosensitive cation channel activated by noxious heat as well as a wide range of chemical stimuli. Although ATP by itself does not directly activate TRPV1, it was shown that intracellular ATP increases its activity by directly interacting with the Walker A motif residing on the C-terminus of TRPV1. In order to identify the amino acid residues that are essential for the binding of ATP to the TRPV1 channel, we performed the following point mutations of the Walker A motif: P732A, D733A, G734A, K735A, D736A, and D737A. Employing bulk fluorescence measurements, namely a TNP-ATP competition assay and FITC labeling and quenching experiments, we identified the key role of the K735 residue in the binding of the nucleotide. Experimental data was interpreted according to our molecular modelling simulations. Calmodulin (CaM) is known to play an important role in the regulation of TRP channels activity. Although it has been reported that CaM binds to the Cterminus of TRPV1 (TRPV1-CT), no classic CaM-binding motif was found in this region. In this work, we explored this unusual TRPV1 CaM-binding motif in detail and found that five residues from a putative CaM-binding motif are important for TRPV1-CT's binding to CaM, with arginine R785...

Horse in the frame of human culture
Reinbergerová, Anna ; Opletalová, Lenka (referee) ; Czumalo, Vladimír (advisor)
The dissertation "Horse in the history of human culture" presents a summary of a history of horse in the human culture. Horses have played a significant role in homo sapiensis evolution ever since it came to Eurasia. From beeing hunted for food, horses soon turned in the process of domestication into working, riding and driving animals of significant importance. In different cultures and epochs, different types of horses of different purposes have been emerging. But since its domestication, horses have allways been tellig a story of culture in its time and space. This is why this topic is a part of the wide array of interest of the therory of culture. In this dissertation, I chose to catch a relation of horses and culture in the historic circumstancies. I hoped to connect my experience with horses with scientific literature on this topic, historical facts and presently existing historical artefacts related to horses. In most of scientific and historical literature, many notes on horses may be find, but mostly they are interpreting the facts wrogly, and sometimes they are even wery far from the truth. the historicians usualy lack the needed knowledge of hipology, etology and zoology. It is common that mainly the horse illustrations are incorrectly described and understood. In the hipological literature, on...

The causes of poor learning results in the subject 6MI211 - Data Analysis
Ryšavá, Iveta ; Komárková, Lenka (advisor) ; Voráček, Jan (referee)
The aim of this bachelor thesis is to statistically process data related to the failure of full time students in the subject Data Analysis and to appropriately interpret the acquired results. Resources to our work are the academic results based on Data Analysis and Math for economists from the winter semester of the academic year 2006/2007 and 2007/2008 and, in particular, a questionnaire formulated by ourselves. The nature of the data collected leads to the analysis of qualitative variables. First single data are described using a table of relative and absolute frequencies for each variable. In addition, we have worked with selected pairs of variables, namely the two-data which are processed in the form of contingency tables and these tables are then further analyzed. There are, inter alia, used tests of independence and homogeneity. For better understanding we have used the tables and the related tests are completed by graphs. From our bachelor thesis, we expect that we find the views of students on this subject and find out, why this subject causes so many problems for most of them.
