Effect of carbamazepine on thyroid hormone homeostasis
Šimko, Julius ; Šonka, Karel (advisor) ; Mareš, Jan (referee) ; Zamrazil, Václav (referee)
8. Summary Introduction: While carbamazepine (CBZ) treatment may affect serum thyroid hormone concentrations it rarely leads to clinically important hypothyroidism. This study was aimed to evaluate an early effect of CBZ on thyroid status in hypothyroid patients with thyroid hormone replacement, as compared with patients without a thyroid disorder. Methods: Twenty-nine patients indicated for CBZ treatment were followed prospectively. Their thyrotropin (TSH), total thyroxine (TT4), free thyroxine (FT4) and antibodies against thyreoperoxidase (TPO- ab) and thyreoglobulin (TG-ab) serum levels were assayed before the start of CBZ medication (150 mg/d increasing to 450 mg/d), and then their TSH, TT4 and FT4 serum levels were assayed at week intervals for 7 weeks. Nineteen patients had no thyroid disorder before CBZ treatment (control group A), whereas 10 patients were treated with L-thyroxine (median 100μg/d ) for hypothyroidism and were stable before CBZ treatment (group B). The fluctuations of thyroid status after the start of CBZ treatment were compared between the groups. Results: In the control group, TT4 was significantly decreased by ca. 15 to 25 %, starting from the 1st week of treatment (Friedman, p<0.001), while FT4 was decreased by only ca. 10 to 15 %, and the significance (p<0.001) was delayed till...

The effects of dexrazoxane on ischemia-reperfusion injury in rat heart
Boudíková, Adéla ; Neckář, Jan (advisor) ; Hloušková, Patricie (referee)
Dexrazoxane (DEX) is clinically used to reduce cardiotoxic efects of anthracycline cytostatics. Its cardioprotective efect is caused by chelatation of free iron and defends myocard against dangerous hydroxyl radicals. This research finds out how dexrazoxane works in ischemic-reperfusion damages of rat's heart. Each rat was infused by DEX (50, 150, 450 mg/kg) or by control solution. Isolated perfused rat's hearts were exposed to local ischemia for 30 minutes than 10 minutes of reperfusion for studing ischemic arrhythmias followed by 15 minutes of local ischemia and 10 minutes of reperfusion to examine reperfusion arrhythmias. For evaluation of EKG (ventricular arrhythmias) was used software CAR and Lambeth convention. Global ischemias (15 min.) were induced in rat's hearts (DEX 150 mg/kg) and left ventricules were used for HPLC to determinate concentration of glutathion. In vivo experiments rats were infused by DEX 50, 150 mg/kg or control solution and were exposed for 20 minutes to local ischemia and for 3 hours to reperfusion. Infarct size was evaluated based on the cross section of heart (GIMP, Ellipse). Maximum total number of ischemic arrhytmias decreased by DEX 150 mg/kg (64% comparing to controls). Reperfusion score was reduced by DEX 150 to 48% and percents of ventricular fibrilation was...

Dvořák, Aleš ; Coufal, Pavel (referee) ; Čabala, Radomír (advisor)
Chiral separation of omeprazole was the subject of this work. Five diferent monosubstituted derivatives of α- and β-cyclodextrins, two of them cationic and three anionics, were used as chiral selectors in various buffers of different pH. The influence of CD and buffer concentrations on chiral separation was studied. Voltage of 25 kV, UV detection in the range from 202 to 303 nm and constant temperature of 20 řC was used for all measurements. Concentration of CDs between 0 and 7 mmol.l-1 was used. Fused silica capillary of 50 µm ID and 375 µm OD, 48,5 cm total and 40 cm effective lenght was used as separation capillary. Dimethylformamide was used in stock solution of omeprazole as solvent, and was simultaneously applied as EOF marker. Succesfull chiral separation of omeprazole was achieved under the application of 2'-O-carboxymethyl-β-cyclodextrin (2'-CM-β-CD) and mono-6-deoxy-6-N,N,N',N',N'-pentamethylethylendiammonio- cyclomaltoheptaose dichloride (PEMEDA-β-CD) where the resolution of 1,42 (pH = 5,1) and 4,29 (pH = 11,1) was measured, respectively.

Ecotoxicological evaluation of contaminated soil and water and use of bioassays for bioremediation
Hubálek, Tomáš
The aim of this thesis is to identify the ecotoxicity of contaminated soil and water during bioremediation. Different test organisms were selected for the battery of ecotoxicity tests such as the bacterium Vibrio fischeri, the terrestrial plants Sinapis alba, Lactuca sativa, and Hordeum vulgare, the water plant Lemna minor, the earthworm Eisenia fetida, and the crustaceans Daphnia magna and Heterocypris incongruens. A comparison of individual biotests with various organisms and endpoints should demonstrate their suitability and sensitivity for the ecotoxicity evaluation of contaminated soil and water. The bioassays were used for the evaluation of the bioremediation of hydrocarbon-contaminated soil, composting of PAH-contaminated soil and remediation of contaminated groundwater. The ecotoxicity of hydrocarbon-contaminated soil originating from a brownfield was evaluated during a 17-month biodegradation pilot test (experiment I). The initial concentration of total petroleum hydrocarbons (TPH) in the soil was 6380 µg.g-1 dry weight; the concentration of TPH in the soil decreased by 65.5 % after bioremediation. The highest toxicity was detected in the first period of bioremediation; however, certain toxic effects were detectable during the whole bioremediation process. The contact tests were the most...

Tomalová, Pavla ; Nachtigal, Petr (advisor) ; Kopecký, Martin (referee)
Influence of MDOCTM Administration on Atherogenic Process in the Experimental Model of Atherosclerosis. Mgr. Pavla Tomalová In this study we investigated prospective hypolipidemic and anti - inflammatory effects of modified oxidized cellulose (MDOC). In the therapy of hyperlipidemia and blood - vessel complications (atherosclerosis) there are used the most considerable substances - statins, today. However, it was also documented, that feed dietary roughage has positive influence on cardiovascular system, inclusive of cholesterol level. MDOC (polyanhydroglucuronic acid), because of it's properties can be classified probably as the soluble fibre. We used apoE deficient mice as the model of atherosclerosis, fed by atherogenic diet. We analyzed levels of total cholesterol and other lipoprotein fractions and serum concentrations of inflammatory markers (IL-6 and VCAM-1) in blood. Total cholesterol concentrations were assessed enzymatically by conventional diagnostic methods and spectrophotometric analyses. For the detection of VCAM-1 expression in aortic sinus and part of aortic arch we used imunohistochemical methods and for quantification of VCAM-1 expression we used stereological methods. Biochemical analyses results showed that MDOC administration did not affect levels of total cholesterol, VLDL cholesterol,...

Czech protectorate press in Reinhard Heydrich era
Renčínová, Pavla ; Bednařík, Petr (advisor) ; Kryšpínová, Jitka (referee)
A total power over the Czech official print became an integral part of the Nazis' policy during the Second World War. The print in all occupied countries should have served as one of the most important means by which the Nazis wanted to promote their ideology. Reinhard Heydrich, who came to the Protectorate Bohemia and Moravia in autumn 1941, at first needed Czech people to cooperate for war industry. He was ready doing anything to achieve it. There were some journalists who /for various reasons/ helped him a lot. They did not only administratively execute his instructions but also took part in many fascist agitations. They had close connections with Gestapo and occupation authorities. On regular press conferences they were given instructions how to inform about what 's happening in the protectorate as well as on the battle field. As the result, they were responsible for many people's lives and fates and meant a great danger for whole nation. Besides these loyal quislings there were a number of such journalists who cooperated with the Czech resistance and exile in London. More than a hundred of them were executed or died in concentrate camps. London radio broadcasting showed how little people believed in the official protectorate print. Although the Nazis were quite successful in threatening the Czech...

Mareček, Vojtěch ; Dokoupil, Vladimír (referee) ; Foller, Bronislav (advisor)
This diploma thesis deals with projection and general design of rotary indexing table for dedicated machine tools. The major attention is given to substitution of hydraulic drive table by table driven by servomotor, namely double axis its positions in vertical and horizontal directions. The total configuration of rotary table is solved as a construction kit, user can used any servomotor and additive gearbox. The work is concentrated to the optimization of table construction for this new designed conception. The positioning and fixing of table plate is solved by couple of Hirth couplings with central hydraulic motor. The computational part of work includes calculations of dynamic of rotary motion of the table and hydraulic system. All important screw connection, gear wheels and Hirth couplings were verified by calculations. Also working lifetime of bearings and strength deformation of major constructional sections of rotary table verification were controlled. Inherent part of work is security analysis, economics analysis, patent search and analysis of construction identity. Presented work contains three-dimensional model, drawing documentation of both variants of configurations and production drawing of major section parts of rotary table.
