Characterization of pore opening relevant residues in TM3 and TM4 domains of Orai1

Calcium (Ca2+) ions play a crucial role in almost every aspect of cellular life. The most prominent calcium entry pathway into the cell is the calcium release-activated calcium (CRAC) channel, composed of the Orai1 protein, and the stromal interaction molecule STIM1. The channel is activated through conformational changes upon STIM1 coupling to the C-terminus of Orai1 protein following store depletion, which in turn allows Ca2+ influx into the cell. The abnormal function of the CRAC channel caused by mutations gives rise to distinct pathologies. Since it has not yet been elucidated how the signal propagation moves to the pore upon coupling, this thesis dives into its investigation by focusing on characterizing the TM3 and TM4 domains and their importance in leading to an open permissive conformation of the channel. The pivotal foundation for the creation of novel strategies in the modulation of the Orai1 function lies with the understanding of the dynamics of the Orai1 pore opening.
UAV photogrammetry as tool for evaluation of fluvio-morphological changes of streams

This thesis is focused on a use of a technology that is relatively new and popular in geoscience applications, including the fluvial dynamics research. Unmanned aerial vehicles are becoming widely used in numerous geographical subdisciplines due to their capabilities and advantages of UAV imaging in comparison with other remote sensing techniques. These advantages and also limitations together with general characterization of UAVs are summarized in the secondchapter of the thesis. The following theoretical chapters deal with basics of UAV photogrammetry and also evaluate the potential of UAV imaging in fluvial geomorphology. The fifth chapter of the thesis comprises a case study that evaluates the use of an UAV equipped with a non-metric camera for fluvial environment reconstruction. Images of Javoří potok have been processed in Agisoft PhotoScan and ArcGIS to indicate geomorphological changes of the stream channel that happened during a 5 month period from November 2014 to April 2015. A method of programming the TV channel ČT1 in connection with its focus groups

This thesis deals with programming of the TV channel ČT1 in connection with its audience. The first part of this thesis is a theoretical explanation of the television programming. It clarifies reasons and methods of the channel structure, then it introduces the research of viewer ratings. It concerns the techniques of the television programming whose goal is to gain sufficient ratings of the stated programme. However, it also determines the Czech Television as a public broadcaster with respect to the Czech law. It tries to explain how this fact affects programming of the Czech Television's channels. The second part of this thesis consists of practical research of programme ČT1. It analyses four weeks in 2015, it clarifies their main features. It takes knowledge from three interviews with the key operatives who are responsible for the programming at the Czech Television. It also devotes to the fact how the target audience forms placing of programmes sociologically. It is known that the association of human life with television broadcasting is necessary and strong. Besides, it identifies the channel ČT1 as it is, it compares its target audience with other channels of Czech television. Renovation of a water reservoir in the village of Žďárná

The theme of this diploma thesis is to restore the existing small reservoir in the village Žďárná. A new safety spillway and bottom outlet will be built, the flood will be shaped according to the cross-sections and the dam of the water reservoir will be fortified. The designed objects will be hydraulically verified. Using the HEC-HMS program, a hydrological model of the river basin is created and subsequently a derived flood wave PV20.
Channel Merging Techniques For Improving Dynamic Range Of ± 10v Signal Chain

This paper presents the channel merging technique that is used in the Analog Devices part AD7606C. In more detail, it describes the current dynamic range (DR) and total harmonic distortion (THD) performance that has been achieved on the first AD7606C silicon. Then, it presents options to achieve higher values of the DR and also options to mitigate the undesirable non-linear current which decreases the THD performance.
Molecular mechanisms of polymodal regulation of TRPA1 receptor

The TRPA1 channel is a universal, nociception-mediating cellular sensor activated by various environmental irritants, potentially harmful physical modalities and endogenous mediators of pathophysiological processes. The polymodality of TRPA1 channel allows the activation stimuli to further enhance or suppress each other's effect. While this modulation effect has its physiological importance in promoting the protective cellular and behavioral mechanisms, it may result into the unpleasant pain-related effects accompanying the chronical pain caused by aberrant TRPA1 channel activity. In order to effectively and selectively target the synergic properties of TRPA1 modulators, while preserving the sensitivity to the environmental threads, the knowledge of the mechanisms of polymodal regulation at the molecular level are required. This doctoral thesis aims at the elucidation of three main mechanisms of TRPA1 regulation: 1) the regulation via intracellular signaling cascades and phosphorylation, 2) the interaction with membrane phospholipids and 3) the temperature-driven gating. The results presented in the thesis show that the effects of the inflammatory mediator bradykinin are decreased by the low-frequency high-induction electromagnetic field used in magnetotherapy. Assessment of anthropogenic modification of the river network using hydromorphological survey as an example of the upper Metuje River

Human modifications and related pressure on watercourses of its ecosystem became greatly discussed problematic in the latest dry years. The translated bachelor's thesis deals with these encroachments on watercourses as well as with its impact on hydromorphological quality. In recherché there are discussed basic hydromorphological knowledge and modifications of watercourses with its negative and positive effects on the watercourse along with its surrounding. For the evaluation of the hydromorphological condition was used HEM methodology (Langhammer 2014) applied on upper course of the Metuje River. Overall, based on the outcome of the HEM methodology the upper stream of the Metuje River is evaluated as a weakly modified. Less disturbed sections can be found in the area of the National Nature Reserve Adršpašsko-Teplické skály. The largest modifications can be seen in the build-up areas of Teplice nad Metují, where these modifications come particularly from the period of the First Republic. The area of the floodplain, is the most affected area. Trace of the rivercourse was overall shorted by 1 569 meters, therefore 7,5 % since mid-19th century, chiefly in the surrounding of Teplice nad Metují. Environmental education on children's channel ČT:D Ty Brďo! and S Jakubem v přírodě

This bachelor thesis is focused on the environmental education in the children's shows at the public channel ČT:D. Specifically this is the shows Ty Brďo! and S Jakubem v přírodě. The main intention is the qualitative analysis of these shows and using of them in the education. To find out what these shows can offer to the children from the perspective of the media studies. The thesis is divided into three parts. The first part emphasis the theory. Primarily the channel ČT:D, it's origin and evolution, program scheme, web sites and education. Besides there's summary of the Framework educational program (RPV) and the Recommended expected outputs of the environmental education (DOV). These materials are then compared with information gained by qualitative analysis. The second part of the thesis is dedicated mainly qualitative content analysis of individual parts and comparison of acquired data with the Framework educational program and the Recommended expected outputs. The last part of the thesis is dedicated to the focus group, which was carried out with eight children from the target group of analysed shows, and summary of the obtained information.
Technical infrastructure in cities

This bachelor's thesis consists of three parts: Theory, calculation and project. Theory deals with possibilities of laying utilities into the ground, collectors, technical corridors, technical cannals, basement manifolds and to the overhead lines. Furthermore, the issue of coordination between utilities and greenery both in the Czech Republic and abroad is also addresed. The calculation part includes water demand, rainwater quantity and waste water balance calculations. Project contains the accompanying report, technical report and drawings. It focuses on the Jircháře street located in Staré Brno, where a new apartment building has already been built, for which new relocation of utilities from Jircháře street to Kopečná street had to be made.
Testing of the LTE basic parameters

This thesis deals with brief introduction to LTE system and processing of signal in uplink and downlink. There is also presented some basic parameters that must be met for correctly working equipment in system. These parameters are divided for transmitter and receiver. From these parameters were chosen those, which can be measured in school labs with available measurement devices. For measuring these parameters were created instructions. For easier record of measured data was prepared protocol with blank tables.
