Heat Shock Proteins - Focus on Therapeutic Approaches in Oncology

Since the discovery of heat shock proteins in 1960s and their immunogenic properties 20 years later in 1980s they have been under rigorous investigation with great hopes for advent of immunotherapy in oncology as well as other pathologies. So are the HSPs a breakthrough in therapy of cancers or will they be just another form of palliative treatment of these notorious and deadly diseases? HSPs is a family of proteins expressed virtually by all living organisms. These proteins have a number of functions that are crucial to organism's intracellular and extracellular homeostasis. Due to the significance of HSPs, over the eons of the evolution, they have been highly conserved in our genetic code. The human HSPs shares almost 50 percent homology with simple unicellular prokaryotes to well over 95 percent homology with higher species of living organisms. Despite of the fact that HSPs have been knows for approximately 50 years and despite of the intensive resears in the field, currently Russia is the only place where HSP immunotherapy has been approved as of 2008 for treatment in renal cell carcinoma. The HSPs immunotherapy has proven benefitial as an adjuvant treatment in some stages of a selected cancers. The Possibilities of solving of unemployment in the Czech Republic and in France

My thesis ,,Possibilities of solving of unemployment in the Czech Republic and in France" has the objective of insight to the issue of solving of unemployment from the perspective of the state and its employment policy, especially to instruments of active employment policy in our republic and in France. My thesis is divided into nine chapters. The research part of the thesis deals with theoretical knowledge from areas as labour market, employment, unemployment and employment policies on the level of European Union and on the level of given surveyed states. At its end I made a short preview to the issue of macroeconomical indicators and national economy that I later used in the practical part for brief characteristics of economical situation of the states and their chosen parts. As I have mentioned above, the practical part of my thesis deals with economical characteristics of both surveyed states and analysis of unemployment in them. Its main part is formed by my focusing on the comparison of possibilities of solving of unemployment in selected higher territorial self-governing units of the Czech Republic and France. Here I perform analysis of the labour market according to different criteria in the Czech region Highlands and in the French region Limousin. Then I define and create a comparison of the particular ways of solving of unemployment in these areas. In my thesis I further express my own assessment of effectiveness of the Active employment policy operating both in the Czech Republic specifically in the Highland region and in France in mentioned Limousin region. The very end contains mainly summary of knowledge gained at the practical part and statement how the set target was achieved.

Inhibiting properties of the inhibitory protein TbIF1 to \kur{T. brucei} F1-ATPase.

To map and define the minimal inhibiting sequence of the inhibitory peptide TbIF1 and to improve its inhibiting property, a series of the N- and C- terminal truncations of TbIF1 was created. The mutated recombinant TbIF1 his-tagged proteins were over-expressed and purified from the E.coli cells and their binding and inhibiting properties were tested using purified F1-ATPase complex.

The Classical Nucleation Theory (CNT) is modified to treat a special case of admixtures - electrolytes. Thermodynamic features of aqueous electrolytic solutions are described in a slightly different way from that for ordinary liquid mixtures. Namely, the excess Gibbs energy is expressed in terms of the osmotic coefficient and the mean stochiometric activity coefficient instead of the activity coefficients.

Petra Dvořáková SUMMARY The aim of this thesis was the study of adhesive properties of branched terpolymers of D,L-lactic acid, glycolic acid and dipentaerythritol, and release of fluconazole and aciclovir from these carriers. All tested carriers were plasticized with 30% of triethyl citrate (TEC). In the theoretical part of this thesis mechanisms of bioadhesion, biological substrates, polymers used in drug delivery systems and mechanisms of drug release are described. In the experimental part adhesivity of plasticized oligoesters was measured. Hydrated mucin isolated from porcine stomach was used as a model substrate for bioadhesion. The rate of adhesivity is the maximal force Fmax [N], which is required for the detachment of tested samples from the substrate. This force was related to the size of the contact surface and expressed in units mN/mm2 . The highest adhesivity was measured in oligoesters labelled 5D and 8D, which had low dynamic viscosity. Petr Fiala The study of endoglin expression in normocholesterolemic and hypercholesterolemic mice Diploma thesis Charles University in Prague, Faculty of Pharmacy in Hradec Králové Pharmacy Background: We studied the changes of expression of endoglin in normocholesterolemic and hypercholesterolemic mice. The changes of endoglin expression were detected in aorta by means of imunohistochemical methods and followed by quantification by Western blot analysis. Methods: We used 8 female normocholesterolemic mice C57BL/6J and 8 female ApoE deficient mice C57BL/6J. ApoE deficient mice were fed during a 8 weeks by chow diet with 1% of cholesterol. Biochemical analysis of blood samples was performed. Then we prepared cuts of aorta with segment of semilunar valve for immunohistochemical analysis. Detection of expression of the endoglin was performed by Avidin- Biotin method (ABC) with detection by DAB. Results: The expression of endoglin was detected only on endothelium of normocholesterolemic mice. The expression of endoglin in apo-E deficent mice was detected on endothelium of atherosclerosis plaque, aortic semilunar valves and also in endothelium with no qtherosclerosis and in capillaries in myocardium. The Six Sigma methodology

Bachelor thesis deals with the desription of the statistical foundations and individual steps of Six Sigma, which are descibed by the DMAIC model. The thesis focuses on statistical process control and control charts. The main goal is to evaluate the possible use of control charts for attributes, namely the control chart for fraction nonconforming or p-diagram and to describe its properties expressed by the operating characteristic (OC). The influence of the sample size on the OC curve is examined. The last part of this theses deals with the desription of the xi-diagram as an alternative of the p-diagram. The properties of this alternative and classical p-diagram are compared.

Gene expression profiling after experimental perinatal asphyxia and the effect of complement-derived anaphylatoxin C3a

Background: The complement system is involved in neuroprotection and brain repair after brain damage. To understand the molecular mechanisms of these processes, we performed gene expression profiling using quantitative real-time polymerase chain reaction (qPCR), which is the most accurate modern strategy for gene expression analysis. Project: Our project was directly aimed at expression profiling of selected genes potentially involved in loss and rescue of neural tissue during three weeks after hypoxic-ischemic brain injury, an experimental model of perinatal asphyxia. Recent experiments have shown that over-expression of C3a under the control of the GFAP promoter (C3a/GFAP) reduced hippocampal injury after left common carotid artery ligation in neonatal mice by 50%, compared to wild type mice. Here, we assessed how the local expression of C3a/GFAP transgene affects gene expression profiles. Gene expression was measured on samples from hippocampus ipsilateral and contralateral to the injury and ipsilateral part of cortex, taken at the time of injury, 6 and 24 hours; 3, 7 and 21 days after the injury. Specificity of interrogation of children

RESUMÉ: The target of the thesis is to point out the specialities that differ the questioning of children and children's testimonies from those of adults in criminal procedure. These specialities are based on the unique reception, memory, imagination, portion of sugestibility and also the ability to express of chidren, which are of deeply different quality to adults. Variations are to be found as well between children depending on their age. It is necessary to provide special approach to questioning of children and evaluation of their testimonies. Person who questions needs to operate with high quality of profesional abilities and knowledge and needs to undergo proper preparations before each action. Mentioned above is focused, whether the child under the questioning is the victim of crime. Inappropriate questioning increases chance of secondary victimization. Criminal Procedure Code deals with questiong of children in its provisions. The age of questioned child does not automatically decrease valuation of the testimony. During questioning of children the profesional teacher should be present while presence of the parents of questioned child is welcomed just under the condition that it is assumed to make the questioning easier or possible. As it is obvious from the title, the discourse deals with the legal adaptation of property relations between husbands in connection with undertaking no matter whtether carry on business one's or both. Simultaneously it deals with the problems of the protection of the party that is weaker economically or in other way. No less target of importace is protection the third party as well.
