Vlčková, Hana ; Solich, Petr (advisor) ; Nobilis, Milan (referee) ; Šimek, Petr (referee)
5 ABSTRACT Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of analytical chemistry Candidate: Mgr. Hana Vlčková Supervisor: Prof. RNDr. Petr Solich, CSc. Supervisor specialist: Assoc. Prof. PharmDr. Lucie Nováková, Ph.D. Title of Doctoral Thesis: Analysis of statins in biological samples The doctoral thesis deals with analysis of statins in biological samples. Statins are the most widely used drugs for the treatment of Familiar hypercholesterolemia. Due to very low concentration of statins (ng/ml) in human plasma a choice of suitable analytical methods is limited. Liquid chromatography with mass spectrometry detection is the most suitable technique due to high selectivity and sensitivity. Sample preparation step has to be used before chromatographic determination of analytes in complex matrices, such as biological samples. Primarily theoretical part of doctoral thesis summarizes individual pharmacological properties and effects of statins. Another part is devoted to theoretical knowledge of UHPLC-MS/MS. The widest section deals with description of the individual sample preparation techniques. The practical part contends with development of UHPLC-MS/MS methods for the determination of statins in biological samples and selection and development of suitable sample preparation...

Vácha, Miroslav ; Weinzettl, Vladimír (advisor) ; Piffl, Vojtěch (referee)
Title: Detection systems for measurements of high-temperature plasma radiation on the COMPASS tokamak by fast bolometers and soft X-ray detectors Author: Miroslav Vácha Department: Institute of Particle and Nuclear Physics Supervisor: Mgr. Vladimír Weinzettl, Ph.D., Institute of Plasma Physics AS CR, v.v.i. Supervisor's e-mail address: vwei@ipp.cas.cz Abstract: Multi-channel spectroscopic diagnostics on the COMPASS tokamak com- prising AXUV-based bolometers, soft X-ray (SXR) and visible light detectors will cover a wide spectral range of the core and edge plasma emission, aiming to realize a fast tomography at microsecond time scales. This thesis reports on a development of the bolometric and SXR diagnostics. First, procedures of the detector selection and their tests are described. There the design and tests of the special complex port plug, com- bining both detection systems, taken into account a strongly limited available space, a heat protection and a shielding during a cleaning glow discharge, is also summarized. Development of new electronic amplifiers and their connection with the data acquisition system is documented. In the last chapters, basic methods for analysis of data collected using these diagnostic systems are introduced. Keywords: tokamak, plasma radiation, photodiode, tomography

Artificial neural networks and their application in the assessment of insurance risks
Macek, Karel ; Jiroutek, Pavel (referee) ; Mrázová, Iveta (advisor)
The present work studies applicability of artificial neural networks in the assessment of insurance risk. Structure and function of multilayer perceptrons, Kohonen maps, Fuzzy ART networks, and Fuzzy ARTMAP networks are described. Concept of insurance risk is defined and the ratemaking by generalized linear models is introduced. Neural networks' methods for reduction of input space's dimension, knowledge extraction, and visualization are summed up. Data describing traffic accidents are acquainted and results achieved on them are presented. The work successfully demonstrates theoretically and experimentally that multilayer perceptrons approximate better than generalized linear models. Modification of multilayer perceptron estimated distribution function of total claim. Analysis performed by Kohonen map and by subsequent visualization detected two significant clusters. Analysis by Fuzzy ART network is presented as a part of new algorithm for reduction of input space's dimension. New algorithm inspired by Fuzzy ARTMAP network discovered a group of accidents where the claim is above average. This group is delimited by interpretable rules. Attached CD contains scripts for Matlab and MySQL that was used for mentioned analyses.
