The influence of essential oils on nematode Heterodera schachtii
Rejchrtová, Pavlína ; Zouhar, Miloslav (advisor) ; Mazáková, Jana (referee)
The sugarbeet cyst nematode (Heterodera schachtii) belong to one of the most importatnt pests of sugar beet in our country and also worldwide. Its occurrence cause economic losses on growing sugar beet. High contamination of fields can lead to interruption of cultivation of sugar beet. The purpose of my thesis was make searches on topic of the sugarbeet cyst nematode (BCN), its occurrence, life cycle, the symptoms of attack and preventive and other protection options. Next target was examinete the impact of selected essential oils on mortality of H. schachtii and their direct or indirect effects on BCN under controlled greenhouse conditions in pot experiment. For testing of dirrect effect of essential oils 23 various essential oils from two companies (Salus and Sigma) were taken. Firstly 0,5 % concentration of essential oils was tested. For each variant was established three replicates, and two control variant. The sugarbeet cyst nematodes were exposed to the direct action of the essential oils for 24 hours, then the experiment was evaluated. In the case of essences FV, FEO, ANO, PA and TAO were further tested concentrations of 0,4 %, 0,3 %, 0,2 % and 0,1 %. For testing of indirrect effect of essences were ranked five essential oils (FV, FEO, PA, ANO, TAO) showing in the direct effect testing experiment 100% efficiency on nematode mortality. After 24 hours of incubation, the fitrate papers with nematodes were transfered into Baerman funnels and living nematode were extracted and counted. For testing the efficiency of essential oils under in vivo conditions were established pot tests under controlled greenhouse condition. It was founded 5 variants (FV, FEO, PA, ANO, TAO) and control variant (K), each of the five repetions. After 90 days was experiment evaluated by extraction of nematode cysts by Fenwick can. From the experimental results I can conclude, that different essential oils used in this study showed various effectiveness on mortality of sugar beet cyst nematode. Results of pot experiment suggest statisticaly significant difference of the number of cysts. Results of pot tests also showed, that essential oils had also some influence on the growth characteristics of sugar beet, namely ground biomass and root system weight.

Thermal analysis of interaction of epidermal lipids VI.
Gillová, Martina ; Doležal, Pavel (referee) ; Musilová, Marie (advisor)
Thermal analysis of interaction of epidermal lipids VI. Martina Gillová, degree work, June 2010 Abstract A skin separates the organism from the external environment, but at the same time it enables the contact with surroundings too. During transdermal aplication a drug has to get over a skin barrier, which is represented especially by outer skin layer, stratum corneum. This layer is composed of keratinized cells embedded in lipid matrix. Currently scientists are searching new substances, which will temporarily be able to increase permeability of the skin and facilitate penetrating of the drug through the skin barrier. We call these substances permeation enhancers. In this work, we concentrated on the evaluation of interactions of cholesterol (one of the components of lipid matrix of stratum corneum) with permeation enhancer ETH5-10 by using DSC (differential scanning calorimetry). Then, we observed interactions of mixtures of these two substances with water. Every time we added such amount of water to our sample, that it made approximately 75% of the sample weight. We did the evaluation of individual components too. The sample with the composition of cca 50% cholesterol and 50% ETH5-10 standed out markedly from the range of another samples.

Design of strain gauge devices for safety belts load measuring
Lontras, Martin ; Jebáček, Ivo (referee) ; Mališ, Michal (advisor)
The aim of this thesis is to design, produce and experimentally verified measuring device for measuring force in seatbelt, by using strain gages. First are discussed possibilities of measuring force by using device placed in between two parts of seat belt or in anchor point. These suggestions were not accepted for various reasons. Therefore is further described girder, which belt slightly deflected so that part of the load is transferred to the strain gauge measuring element. As default concept is choosen aluminum U-profile, in which is placed measuring mechanism. There are performed calculations of basic geometry and stress control. Then is described manufacturing and calibration on universal testing machine. In conclusion is realized measuring of the force in seat belt, which is loaded by shock force, occasioned by dropping weights from a certain height, which simulates the behavior of the belt and gauge during a crash test.

Analysis of beef cattle herd
BRŮHA, Karel
The purpose of this thesis was to evaluate the level of performance of Aberdeen Angus beef cattle breed bred in Less-favoured areas in southern Bohemia. This evaluation was carried out on Mr. Lepša?s farm in the years 2010 and 2011. 74 basic herd cows and 108 purebred calves born during this period were selected for this purpose. The date of birth, number of births, age at first calving and the average time interval between calving were checked on cows and the date of birth, the weight at birth and the weight in the age of 120 and 210 days were checked on calves. Average daily increments in the interval from birth to 120 days of age, from 120 to 210 days of age and from the day of birth to 210 days were calculated. The group of calves was sorted out by following parameters. Year of birth; sex and year of birth; month of birth; order of calving of the mother. Microsoft Excel was used for processing the statistic data. Basic statistical characteristics for the monitored parameters were calculated and differences between groups were verified by single-factor analysis of variance (F-test) and paired t-tests for the corresponding levels of significance. Breeding cows were evaluated by age and number of births. Cows with 1 (23%) or 2 (17.6%) births are represented in the largest number in the herd including even the cows with 15 births. Cows at first calving reached an average age of 813 days and average length of the interval between calving was 393 days. Calves regardless of sex reached the average birth weight of 33.72 kg in the year 2010, 33.31 kg in 2011, in 120 days of age the average live weight reached 207.95 kg in 2010, 202.94 kg in 2011 and calves in 210 days of age reached an average weight of 309.61 kg in 2010 and in 2011 306.09 kg. Average daily gain of calves from birth to 120 days reached 1451 g in 2010, 1.413 g in 2011, between 120 and 210 days of age the average daily gain was 1413 g in 2010, 1169 in 2011 and in the period from birth to 210 days of age it was 1314 g in 2010 and 1299 g in 2011. Analysis of breeding from economic perspective by comparing revenues (including subventions) and expenditures in each year resulted in findings that the breeding is moderately profitable and it follows that without subventions which constitute an important part of the revenue would be difficult to realize this breeding.

The gelation of hydroxyethylcellulose and hyaluronan using citric acid
Martinková, Martina ; Krouská, Jitka (referee) ; Hurčíková, Andrea (advisor)
This master's thesis deals with gelation of hydroxyethylcellulose and hyaluronan crosslinked with nontoxic crosslinking agent – citric acid. First of all, the optimalization of gelation process of hydroxyethylcellulose took place. After obtaining the product insolube in the ultrapure deionized water, a reaction mixture of hyaluronan and hydroxyethylcellulose in two different weight ratios were prepared. There was prepared solution containing only hyaluronan as the polymeric part too. Citric acid was used in concentrations of 5 %, 10 % and 20 % (w/w of polymer). These solutions of polymers and citric acid were gelated under the same conditions and the products were compared to each other using Differential Scanning Calorimetry, Thermogravimetry and Fourier Transform Infrared Spectroscopy. The products containing hydroxyethylcellulose and hyaluronan with hydroxyethylcellulose in weight ratio equal to 3 : 7 were considered crosslinked.
