Biotechnological production of polyhydroxyalkanoates using wastes of coffee production

This bachelor thesis is focused on study of production of polyhydroxyalkanoates (PHA) by bacteria Burkholderia cepacia and Burkholderia sacchari. Production of PHA has been studied using spent coffee grounds hydrolysates which have been prepared by different aproaches. In the thesis is compared standard method of coffee ground hydrolysis by mineral acid followed by enzymatic hydrolysis. Hydrolysates which have been produced were analyzed in terms of concentration of saccharides and efficiency of hydrolysis. The best producer of PHA (based on results) was bacterium Burkholderia cepacia, in the medium which has been prepared by hydrolysis of spent coffee grounds extracted by 60% solution of ethanol. Biomass yield of this cultivation was 3,553 g/l with 32,472% PHA content. This PHA contained 6,09% 3-hydroxyvalerate. In the other experiment, we verified alternative hydrolysis of spent coffee grounds – by commercially available enzymes. We used cellulase, hemicellulase and the enzymatic cocktail which has been produced by mould. Hemicellulase was the most effective hydrolytic enzyme and its application resulted in production of the highest amount of biomass – 5,708 g/l. In this cultivation, only homopolymer PHB has been which is probably caused by the fact, that during the enzymatic hydrolysis levulinic acid or any other potential precursor of 3HV is formed.

Comparison of healthcare systems in the Great Britain and the Federal Republic of Germany in years 2000 - 2014

This bachelor thesis is focused on a comparison of health systems of two states which are using completely different ways of financing based on divergent models of heatlhcare system. The thesis describes the period 2000 - 2014. Theoretical part of thesis is devoted to description of basic terms of the healthcare and functioning of basic healthcare models. In practical part of thesis are analysed and compared healthcare systems of Great Britain and Germany. They are compared in economic point of view where the main point is percentage share of healthcare expenditure on GDP. They are also compared in terms of quality of healthcare where is used for example number of doctors or infant mortality. The aim of thesis is to compare those healthcare systems and to find which is better in terms of public finance and provided quality of health care.

Analysis of the Czech-Chinese cooperation focusing on foreign trade in mechanical and power engineering

The aim of this diploma thesis is to highlight in theoretical and practical level the possibilities and opportunities as well as risks and threats to enter the Chinese market for business or commercial enterprises. Framework of the first part - SWOT analysis of the Chinese market - was designed according to information provided by individual companies actually exporting to the Chinese market. This section is devoted to monetary and fiscal policy and analysis of consumer demand, the tax system in China, protection of intellectual property rights or the issue of corruption, bureaucracy and the Chinese state-owned enterprises. Subsequently there are examples of three companies - Skoda Power and Skoda Transportation and Skoda JS - operating for several years on the Chinese market. Each company chose different way of entering this market and the aim was to analyze these approaches in terms of financial and legislative expediency. The last part of the paper is devoted to vision and analysis of emerging perspective industries and energy.

RING! - Completion of the Ring Road in Brno

Diploma thesis deals with the completion of the urban fabric in Brno in the area bounded by streets Cejl, Koliště, Křenová a Vlhká. The location is very close to the historical center of the city , but only for the original walls. This site was created as a suburb outside the walls , which was unlike any other inner city type structures with smaller houses with courtyards and gardens. In the 19th century this area avoid the great industrial boom and there were factories built Jewish industrialists. Also this way since the mid-19th century through the railway . After the war in 1945, factories were combined into complex textile manufacturing Mosilana . Development of this area is very mixed , different mass proportions and functional use and is not as good as it close to the historic cores could be . With the considered plan to move the main train station in the displaced position established in this study . This study proposes the redevelopment and extension of the site . Below we deal with historical context , spatial , functional and traffic analyzes and proposals that I worked for two semesters . Also in this study, I deal in detail southern part of the area where I was during one semester proposed two corner houses at the intersection of Křenová and Koliště street. In the last semester, I examined a draft of the Museum of Jewish Culture in Skořepka.

Nicolas Sarkozy s reflection in the of ČTK in a period of Czech European Union Presidency

Bachelor's thesis Nicolas Sarkozy's reflection in the news of ČTK in a period of Czech European Union Presidency pursues French president Nicolas Sarkozy's activities in a period of Czech Presidency from 1st January to 30th June 2009. The Czech Republic was the second post-communist country after Slovenia which was the leader of the European Union so our Presidency received much interest. At the beginning, the Czech Republic as well as its Presidency was in a shadow of France which led the European Union before and its President Nicolas Sarkozy whose Presidency was very ambitious and dynamic. The aim of this thesis was to find out, how the news corresponded with Sarkozy's qualitites. Whether the news corresponded and Nicolas Sarkozy was really portrayed as the hyperactive and tireless politician, or his picture was the different, and the news of ČTK did not correspond with his characteristic properties. The second part of the thesis was a quantitative analysis of all news which in the text news of ČTK about Nicolas Sarkozy issued in the pursued period, and the detailed analysis of their metadata. These are always for clarity put and summarized in the tables. In some cases, I used graphs for better visibility. More detailed graphs of the quantity of news issued for each month are listed in the annexe.

Secondary prepositions (in Russian-Czech-Ukrainian context)

The prepositions, as a word class, is not uniquely determined. Some linguists consider the preposition as a particular morpheme, some others as an auxiliary, not fully significant word class, some linguists do not regard the preposition as a word at all. The prepositions, from the formal point of view, can be divided into primary and secondary and, depending on the number of their components, into one-verbal and multi-verbal prepositions. The secondary prepositions are further divided by the majority of linguists into adnominal, adverbal and adverbial, however some linguists speak of adadjectival instead of adverbial prepositions. The lexical meaning of prepositions is analysed in different theories, we are of the opinion that the preposition has a lexical meaning. This fact has been proved by I. K. Kutcherenko (10), and we have verified his theory in the practical part of our dissertation. We have focused on the semantic nuances of chosen causal secondary prepositions, collocates, which are the most frequently used with prepositions, and also the frequency of causal prepositions. Depending on a given context, prepositions can change their lexical meaninig and can be replaced by synonymous equivalent terms. Markéta Kalinová Abstract The thesis titled The possibilities of individualisation, differentiation and personalization in teaching and learning strategies deals with teaching and learning strategies, forms and methods suitable for teacher's work in a mixed ability classroom. Theoretical basis results from the presupposition that every human being and naturally also every pupil is different and has their own educational needs. The thesis tries to find opportunities how to fulfil these needs within the framework of collective schooling. It also concerns the development of differentiated teaching in the history of education. The subject matter is also described in connection with the recent Czech educational reform. The empirical research describes a situation in one Czech basic school. The aim was to find out elements of differentiated instruction. Accounting for Construction Contracts

A construction contract is a contract specifically negotiated for the construction of an asset or a combination of assets that are closely interrelated or interdependent in terms of their design, technology and function or their ultimate purpose or use. The primary issue in accounting for construction contracts is the allocation of contract revenues and contract costs to the periods in which the construction work is performed. Four approaches to accounting for construction contracts have developed over time: the completed-contract method, recognition based on cost incurred, the percentage-of-completion method and recognition based on invoicing. The basic principle of the completed-contract method is to recognize revenue only when it is certain, i.e. in the period when the contract is completed. During the period of performance no revenue is recognised and costs are accumulated in the balance sheet. The main disadvantage of this method is the lack of comparability of the presented amounts between single periods. On the other hand the method is based on the finally determined results, which means that the reported values are certain and cannot be manipulated by the company. Recognition based on costs incurred has the basic concept of recognizing profit only when it is certain. Thus the profit is recognised when the contract is finished. During the realization of the project revenue is recognised only to the extent of contract costs incurred. Contract costs are recognised as an expenses in the period in which they are incurred. The amounts of expense presented in the income statement are fully comparable between accounting periods. The reported revenue has much lower volatility than when using the completed-contract method. This approach provides the users of financial statement with an indication of the volume of a company's business. Under the percentage-of-completion method contract revenue is matched with the contract costs incurred in reaching a stage of completion, resulting in a reported revenue, expenses and profit which can be attributed to the proportion of work completed. Financial statements based on this method present more accurately the relationships between the gross profit from contracts and related period costs. The main disadvantage is the dependence on estimates of contract revenue, contract costs and percentage of completion. The estimates can be made intentionally or unintentionally inaccurate. In contrast to the other approaches recognition based on invoicing has no theoretical concept. Contract revenue is recognized according to the invoiced amounts in the appropriate period. An expense is recognised in the amount of costs allocated to parts of the project, which are the object of invoicing. The fundamental disadvantage of this approach is the possibility of manipulation of the amounts of recognized revenue and expenses. The presented amounts can be very different in similar economic situations. For the users of financial statements, the percentage-of-completion method is preferable, because this method provides useful information on the extent of contract activity and performance during a period. The presented amounts are fully comparable between accounting periods. On 24 June 2010 the IASB and FASB published an exposure draft Revenue from Contracts with Customers. This new standard should supercede IAS11. It has another approach to revenue recognition, but it doesn't mean that the presented values of construction contracts will differ significantly.

Truth and Meaning: The Dialectics of Theory and Practice

Tarski's semantic conception of truth is arguably the most influential - certainly, most discussed - modern conception of truth. It has provoked many different interpretations and reactions, some thinkers celebrating it for successfully explicating the notion of truth, whereas others have argued that it is no good as a philosophical account of truth. The aim of the thesis is to offer a systematic and critical investigation of its nature and significance, based on the thorough explanation of its conceptual, technical as well as historical underpinnings. The methodological strategy adopted in the thesis reflects the author's belief that in order to evaluate the import of Tarski's conception we need to understand what logical, mathematical and philosophical aspects it has, what role they play in his project of theoretical semantics, which of them hang in together, and which should be kept separate. Chapter 2 therefore starts with a detailed exposition of the conceptual and historical background of Tarski's semantic conception of truth and his method of truth definition for formalized languages, situating it within his project of theoretical semantics, and Chapter 3 explains the formal machinery of Tarski's truth definitions for increasingly more complex languages. In both variations, part is machined by turning. Progressive variation can produce the part for approximately one third of the time.
