This thesis focuses on practical use of metallography in machine engineering. First part devotes to the technical material characteristics, their chemical composition and its application in machine engineering. This chapter also includes the issue of internal defects and practical demonstrations of internal microstructure. The following section describes the metallographic samples preparation. Phases are sorted chronologically according to their nature and technological process. Significant part of this chapter focuses on the final procedures of metallographic samples treatment, namely the polishing and etching. Major impact on phase structures image quality is the proper chemical composition of the etchant. Final part of the thesis describes the principle and nature of the light and electron microscopy. Based on the approach of image analysis the macroscopic but also lattice defects can be determined considering the selected magnification.
Research-like education in primary schools

In this thesis, we assume a pan-European issue of the decline of interest in science and science education. In connection with this issue we conducted a literature search. We have also mapped the literature related to the issue of education research in the Czech Republic and the rest of the world. In an effort to increase pupils' interest, increase enthusiasm for science, we deal with activation teaching methods that compare with frontal teaching. As a motivational method we chose a research method of teaching. We prepared a lesson, which we tested among pupils of secondary school. In this work we watched 16 hours in 6 classes. Those lectures were inquiry and frontal way led. The results are presented in the practical part of this thesis. During lectures we wanted to stimulate students' interest in science and science research. Testing the influence of newly created learning materials for the fluorescent microscope on students` knowledge

Among students, interest in Science is decreasing, despite the fact that knowledge of Science can be useful in many real life situations, such as medicine, conservation etc. Science education can also simply help to form a positive attitude towards nature. There are many possibilities of increasing students' interest in Science that could increase their motivation to study. Apart from Inquiry Based Education or Prescribed Practical Activities, working with devices used in research practice is one of the possibilities. If a school doesn't own such devices, there is a possibility to borrow them, or to work with them via the Internet. Possibilities of remote access to these devices are subject to research abroad, and this trend is also starting to appear in the Czech Republic. One example of a device that can be borrowed by schools is a fluorescence microscope at the Department of Didactics and Education of Biology, Faculty of Science, Charles University in Prague. In my thesis, I was investigating the influence of the presence or absence of this fluorescence microscope on the knowledge of students. Microscopic methods designed to morphological proof of selected parasite species

Parasites are a global health problem, and not only in developing countries. Therefore, it is important to ensure early prevention and correct diagnosis of parasitic diseases due to the rising incidence of imported diseases. This bachelor thesis was conducted as a literature review and dealt with microscopic methods which are mostly used in parasitological practice. On the basis of available resources, theoretical part is divided into two parts. The first dealt with the history, principles and preparation of samples for microscopy, the second was focused on the morphology of the seven selected parasitic species from three major groups of parasitology- protozoa, helmints and arthropods. Due to the large scale of parasites, there were chosen most important representatives affecting humans and with the imag on global health with possible zoonotic potential. In the experimental part were created photographs of selected species, using Bresser brand light microscope for it´s practical demonstration. Own part of the work dealt with the evaluation of photographs taken by light microscope and it´s comparison with electron microscopy in terms of demand and their use in practice.
Reconstruction of Data from Damaged CD/DVD

This bachelor thesis deals with the reconstruction of data from a damaged CD/DVD. The aim is to create an application which will be able to get a bitstream from an image (scanned fragments of a disc) acquired by an optical or electron microscope. This thesis also examines a data representation on the disc, process of preparation of specimens of CD and DVD and scanning of these specimens. The algorithm of the created software, its testing and results are described in this thesis as well.
The focus control unit for the optical microscope

The aim of bachelor thesis was to focus on the issue of creating photos with great depth of field, which are developed on an electronically controlled optical microscope with photographic equipment. Arduino module with a series ATMega (Atmel Corporation) microcontroller, that was selected for management control device has been described in detail in the next section. Work includes the design and implementation of the control unit, which automates the process of capturing digital images from the microscope. The control unit contains a module Arduino with ATmega328 processor, which controls the stepper motor connected to focus of the JENAVERT microscope (Carl Zeiss). Device communicates with the user by buttons and LCD display.
Realisation of USB microscope programmable movement in axis z

This bachalor thesis discuss about realization of USB microscope programmable movement in axis Z. The first chapter desribes solder joint and its quality requirements and inspection. Description of the z-axis movement and software is demonstrated in next parts. The last chapter is devoted to the use of programmable movement. Function, components and equipment are described in details.
The issue of revision of the electric sliding contact machines

This work is dealing with issue about revisions of electrical equipment with focused on sliding contact. The first time I will deal enumaration and simple desription the mainly machines which uses sliding contact. The next time i will deal legislation two of standard for revision and then I will write about basic procedures for work with microscope. The next time I will bring photographic documentation of samples of brushes and at the last the strengh test of brushes with results.
Microscope and it´s use in engineering practice

This work deals with the use of the light microscope in industry and its use within the automatization of the diagnostic method specialized on electrical machines with a sliding contact. The method is called the dust particles method and it is used to determine the loss of the brushes used in the machines. The current method involves a manual evaluation of an acquired data, which demands additional tests and the evaluation itself is very difficult. The objective of this work is to simplify and accelerate the method using the computer analysis. First, the usage of the light microscope, through which the samples are obtained, is showed. The method uses these samples as the input. Then, the principles of the dust particles method are demonstrated and the image analysis using the MATLAB program follows. The guide to the image analysis with the elimination of rising defects using the MATLAB program is available. Then, the volume of the intercepted dust particles is calculated using the MATLAB program. The dust particles were obtained from the LFC 55h brush using the Arsenal LPT 1513 – T light microscope.
Microscopic Analysis of Chips Security

The goal of this thesis is to work out an introduction to the chip packaging and decapsulation. Further can be found a description of a method leading to dacapsulate concrete chips. Final part is devoted to getting chip pictures using microscope and analysis of the pictures afterwards.
