Development of peptidomimetics on alpha(v)beta(3) integrin receptor for tumor imaging

The integrins are family of heterodimeric receptors with high importance in many cell processes. They are expressed by all multicellular animals. Mainly alpha(v)beta(3) subset plays important role in cells adhesion with surroundings. Adhesion decrease by occupation of these receptors is used as restriction for tumor metastasis or for early tumor imaging. Description of structure and three-dimensional orientation of binding place of this subset were set RGD (Arg-Gly-Asp) peptide as high affinite ligands. According to computing accounted models of interaction between RGD peptide and binding place were also nonpeptide ligands prepared. In this publication we tried to prepare potentionally usable molecules for tumor imaging on PET scanners. For the base we chose already known molecules with high affinity to alpha(v)beta(3) integrins and with iodine substitution we got possibility for radioactive labelling. Unfortunately, this incorporation of iodine into the molecule decreased IC50 to value preclusive practical using.

School social worker - Attitude of Pedagogical workers in Elementary schools

In this Bachelor Thesis I am dealing with the topic of school social work. Its main objective was to determine the elementary school teachers' attitude towards school social workers. A partial objective was to determine the pedagogues' opinion on whether the function of a school social worker could have a positive impact on functioning of schools and a healthy social climate. Up until now, the terms School social work and School social worker have not been very well-known in the Czech Republic. They are neither well-established nor do they have any legislative backing. In isolated cases a similar position exists in some community type elementary schools. In recent years the topic of introducing school social workers has gained momentum, however, up until now, mostly on academic level. The theoretical part of my bachelor thesis is devoted to mapping the school social work throughout the world, where school social workers have been active for many years. The U.S. have the longest history of school social work, that is why most of the first chapter is devoted to the history and present of the American school social work. In the U.S. school social work dates back to 1877 when the Child Labour Act has been adopted followed later by the Compulsory Education Act. At that period the social workers' main job was to ensure that children attend schools. Later, once school social work had been officially introduced, their authorities have gradually broadened. At present their school and out-of-school activities are very extensive and according to the School Social Work Association of America they provide services to three main target groups: students, parents and family, and school staff. Besides the U.S., school social work has also evolved in Europe. The next section of the first chapter is devoted to England, Germany and Slovakia. The history of school social work in Slovakia is the shortest, however, considering our common history and similar educational systems, provides the most relevant comparison and experience. In the theory oriented chapter two I describe the Czech educational system and analyse the current situation at elementary schools. I outline how counselling services have evolved, how new working positions are being created, such as the school prevention methodologist or the education counsellor. In this chapter I also strive to describe specific activities by the Ministry of Education, Youth and Sports implemented in form of national projects the cost of which is partially being covered by the ESF. The projects aim to improve the educational and preventive function of schools, develop counselling services, and improve the know-how and competences of educational workers. In the practical section of my thesis I strive to uncover opinions of educational workers on establishing a new position of a school social worker. To carry out the survey I have used a qualitative research strategy and the semi-controlled interview technique. The research sample included elementary school teachers from the Vysočina region. I was able to interview a total of 21 respondents. When evaluating the interviews, I have used the tuft formation method. The results of my survey suggest that a school social worker would be a welcome addition to the team, however, it is not entirely clear whether he/she is currently needed. Considering the fact that the research was done while the "Common education" concept was being introduced at schools, the teachers did perceive the necessity of introducing other positions. To answer the partial question of my thesis, we could say, that if we had school social workers at schools, their work would have a positive impact on the overall school climate.

Non-canonical Bioenergetics of the Cell

Cancer cells generally present abnormal bioenergetic properties including an elevated glucose uptake, a high glycolysis and a poorly efficient oxidative phosphorylation system. However, the determinants of cancer cells metabolic reprogramming remain unknown. The main question in this project was how environmental conditions in vivo can influence functioning of mitochondrial OXPHOS, because details of mitochondrial bioenergetics of cancer cells is poorly documented. We have combined two conditions, namely glucose and oxygen deprivation, to measure their potential interaction. We examined the impact of glucose deprivation and oxygen deprivation on cell survival, overall bioenergetics and OXPHOS protein expression. As a model, we have chosen a human breast carcinoma (HTB-126) and appropriate control (HTB-125) cultured cells, as large fraction of breast malignancies exhibit hypoxic tumor regions with low oxygen concentrations and poor glucose delivery. Heraldic and prosopographical analysis of two armorials from the family archives of Vratislavs from Mitrovice. With edition

This thesis focuses on scientifical elaboration of two heraldical manuscripts from the Vratislavs from Mirtrovice family archives in the village Dírná (Soběslav region). According to the village name, the manuscrips are known as the Armorials from Dírná in the profesional literature. They were laid aside a little bit because their existence was known only to a relatively narrow circle of specialists as they were kept far away from the research centres. Both books had never been observed in ligt of authorship, datation or analysis of their content. We are speaking about two paper manuscripts, inventary No. 56, sign. I B 9 and No. 58 sign. I B 10. They comprise 984 and 1358 painted coats-of-arms including name of the family beneath each picture. The codicological analysis enabled to identify authors of both armorials, i. e. Kryštof Vilém Harant from Polžice and Bezdružice (1617-1690) and Václav Ignác Vratislav from Mitrovice (1645-1728) and the epoque of their creation in the 1670's and 1680's. Further research proved the necessity of creating an electronical databases that enabled to work effectively with immense volume of data. Analysis of the content of armorials showed that they contain 961 family coats-of-arms. Those represent from the two thirds Czech noble families, the rest is aproximately in the same...

The Sibling Constellation

The topic of this thesis is the sibling constellation theory. This theory says that the order of birth can affect many areas of life. The aim of this thesis is to verify this theory. The theoretical part of the thesis defines the term sibling constellation and briefly describes the history of research connected with this theory along with information about differences between certain author´s interpretations. The thesis also deals with the upbringing stereotypes that cause differences between individuals born in a certain order and with the description of their typical personality traits. The practical part of the thesis concentrates on verifying of sibling constellation theory through a questionnaire. The questionnaire is based on facts about each sibling position that were learned from literature and deals with the relationship between respondent´s parents and siblings in order to reveal possible rivalry and jealousy in their family environment.

Realisation of Foster Care in Austria

The graduation theses considers the solutions to foster care in Austria. It describes the con- ditions for realisation of foster care in a given country, explains the preventive importance of ambulatory services for children, youth and their families and presents available forms of substantional care in institutions and family foster care. Practical part of my work demon- strates one of the available forms on an example of a particular institution. The aim of this work is a description of current system and presentation of principles of foster care in Austria, so that it could serve as a possible inspiration for development of foster care in the Czech Republic.

Lesbian families in a context of Czech society

Nukleární rodina složená ze dvou biologických rodičů a jejich dětí, v níž muž je v první řadě "živitelem" a žena pečuje o děti a domácnost, je modelem charakteristickým zejména pro 19. století. Emil Durkheim ukázal, že tento typ rodinného uspořádání již sám o sobě je jedním z výsledků postupující individualizace, o níž se v poslední době hovoří v souvislosti s tzv. krizí rodiny. Uvádí se, že rodina tuto nukleární formu nabyla v první polovině 20. století, kdy se poprvé silně prosadil požadavek lásky mezi manželi jako jedna z důležitých podmínek k jejímu založení. Pozice muže a ženy se začala zrovnoprávňovat především kvůli vstupu žen do zaměstnání přibližně v 60. letech minulého století. Cílem tohoto typu rodiny se stalo dosažení individualizace všech jejích členům, což však bylo vyváženo její větší nestabilitou. Současná rodina již není jednotkou ekonomickou, nýbrž je založena na sdílení emocí. Je svazkem mezi dvěma svobodnými individui, kteří se dohadují na konkrétní podobě rodiny a vztahu. Chybí zde předem daný vzor či model určující, co je to rodina, mateřství, otcovství a jaké role má kdo zastávat. Současní lidé usilují o autonomii a nezávislost, ale přitom si dále přejí žít společně s ostatními, protože je potřebují k tomu, aby mohli být sami sebou, chtějí být společně svobodní (Dudová, Vohlídalová,...

Continuing Professional Education Planning in Social Service Organisations

The goal of this dissertation was to compile a questionnaire for social workers and heads of social services that could be used to draw up educational plans for their organisations. The theoretical part deals with the continuing professional education in organisations and describes general principles of learning needs analysis. It also describes the legal basis of continuing education of social workers and concerns itself with theoretical principles of continuing professional education planning. The practical part contains the data from the questionnaires filled in by social workers from the social assistance service for families with children of Charity Děčín and openIaccess club of Charity Beroun, and educational plans of these organisations for the next two years. The first draft of questionnaire was changed in accordance with the critical comments of social workers. The final version of the questionnaire is at the end of the dissertation.
