Termální analýza interakcí epidermálních lipidů Ι Veronika Kafková, diplomová práce, červen 2007 Abstrakt Fyziologickou funkcí kůže je chránit organismus proti vnějším vlivům prostředí. Stratum corneum (rohová vrstva) je nejsvrchnější vrstvou kůže a je zodpovědná za její bariérovou funkci. Podstatou chování rohové vrstvy je její jedinečné chemické složení a uspořádání jednotlivých složek. Rohová vrstva je tvořená plochými, do sebe zapadajícími buňkami vyplněnými keratinem (korneocyty) a intercelulární lipidovou matrix. Hlavními složkami lipidové matrix jsou ceramidy (50%), mastné kyseliny (10%) a cholesterol (25%). Cílem této diplomové práce je experimentální ověření možnosti využití diferenční skenovací kalorimetrie (DSC), jako vhodné a sensitivní metody pro měření in vitro směsí lipidů obsažených ve stratum corneum. Změřili jsme a vyhodnotili DSC záznamy vzorků mastných kyselin (kyseliny olejové, linolové, myristové, palmitové) a cholesterolu, směsí mastné kyseliny a cholesterolu (1:1,m/m), dvousložkových směsí kyseliny olejové a cholesterolu (v poměrech od 1:9 do 9:1) a směsí kyseliny olejové, cholesterolu a vody. Metabolism carcinogens and drugs by the system of monooxygenases

Ellipticine, an alkaloid isolated from Apocynaceae plants, exhibits significant antitumor and HIV activities. Ellipticine is a pro-drug, whose pharmacological and genotoxic effects depend on activation by cytochromes P450 (CYP) and peroxidases (Px) to a reactive species generating DNA adducts. To elucidate contribution of CYPs (and which of them) and Px to ellipticine activation, we used rat and mouse models, mice with deleted gene of NADPH:CYP reductase in the liver, thus absenting this enzyme in the liver (HRNTM ) and a control mouse line (WT), rats treated with ellipticine, and microsomal systems isolated from the liver of mouse lines and from the liver, kidney and lung of rats. The purified enzymes, CYP1A1 and 3A4, reconstituted with NADPH:CYP reductase were also used. The effect of cytochrome b5, a facultative component of the mixed function monooxygenase system, on ellipticine oxidation by CYP1A1 and 3A4 was also investigated. Carcinogenic benzo(a)pyrene (BaP), known to covalently bind to DNA after its activation with CYPs, was investigated for its potential to generate DNA adducts and to induce CYP and NADPH:CYP reductase enzymes in mouse livers. Verification of Rhythmic Gymnastics Programmes in Physical Education Curricula for Special Elementary Schools

Název: Ověřování programů rytmické gymnastiky v učebních osnovách TV ZŠ speciálních Verification of rhythmic gymnastics' programmes in classes of psychical education at special eleiJlantary schools Cíle práce: Cílem diplomové práce bylo vytvořit a aplikovat modelové jednotky v hodinách tělesné výchovy s postiženými žáky. Metoda: Pro vypracování diplomové práce bylo použito metody pozorování. Na základě této metody jsme získali písemný materiál sestavený ze zjištěných poznatků vypozorovaných průběžným sledováním respondentů při průzkumu pohybových dovedností při cvičení s náčiním a při aplikaci modelových jednotek. Dále bylo použito metody srovnávání pohybových dovedností na začátku sledovaného období a na konci sledovaného období. Posledním úkolem bylo vypracovat anketu, kterou bude zjištěn zájem respondentů o rytmickou gymnastiku v hodinách TV. Výsledky: Výsledkem diplomové práce bylo zařazení pohybových cvičení s náčiním a hudebním doprovodem do hodin TV. Hudba tvořila funkci motivační a regulační. Náčiní zde přispívalo, nejen k pestrosti cvičení, ale také k podněcování a zlepšování motorických dovedností. Z vyhodnocení ankety je patrné, že všech 12 respondentů cvičilo při hodinách TV rytmickou gymnastiku velmi rádo a u všech došlo k posílení zájmu o hodiny TV. Klíčová slova: pohybová aktivita,...

Methodics of investigation, documentation and care of oriental rags and carpets

Zpracovaná metodika si klade za cíl především zlepšit v procesu správy a provozu veřejnosti zpřístupněných památek informovanost a odtud povědomí o hodnotě orientálních koberců. Tím vytvořit základní předpoklad pro zlepšení jejich inventarizace, péče i prezentace. Text je rozdělen na 7 kapitol. Jejich autoři podávají přehled o historické i současné funkci koberců v dějinách evropské obytné kultury, popis jejich výroby, materiálů a barev a následně specifikaci charakteru jednotlivých významných výrobních oblastí. V dalších kapitolách pak uvádějí pravidla průzkumu, evidence a klasifikace koberců, profesionální principy jejich fotografické dokumentace a především dnes tolik potřebné rady a pokyny k průběžné kontrole jejich stavu a preventivní péči o ně. Závěrečná kapitola posléze přináší rámcová pravidla pro presentaci orientálních koberců v zámeckých instalacích. Vzhledem ke specifičnosti a obecně nedostatečným znalostem předmětu metodiky jsme se rozhodli připojit pro čtenáře kromě výběrové bibliografie i výkladový slovník nejčastěji užívaných odborných pojmů.

Thermal analysis of interaction of epidermal lipids III Lenka Dvořáková, diploma paper, June 2008 Abstract The skin is the organ protecting the human body from unwanted influences from the environment. Physical chemical properties, structure and ordering of the epidermal lipids, mainly in the stratum corneum, play a key role in the barrier function of the skin. The major lipids are ceramides, cholesterol and free fatty acids in this layer. We are used the samples mimic such the most frequently referred composition of the epidermal lipids, contained pseudo-ceramid, oleic acid, cholesterol and cholesterol sulfate. Because the samples with oleic acids can at the higher temperatures influence the behavior of the particular constituents of the mixture, we are also performed the evaluation of the contained simpler samples which compound cholesterol and pseudo-ceramid. We are tried hydrated the mixtures at in vitro environment by various routes. And we are observed their characteristics and interactions by means of the differential scanning calorimetry. We are also deal with the evaluation of the particular raw materials and the double-barrelled mixtures. We are verified and in detail elaborated the operating sequences which are not completely unambiguously referred in the literature by means of these studies.

The parameters of maximum breathing gas exchange depending on diaphragmactivation

The alm of the work was to evaluate the impact of facilitation of physiologic coordination of respiratory and postural functions of diaphragm by Vojta-therapy on gas exchange. This thesis summarizes available knowledge about anatomical background of diaphragm functions, together with other trunk muscles known as deep stabilizing system. Moreover, it describes coordination of breathing and postural functions. We tried to objectify impact of diaphragm activation on gas exchange in healthy adults. Two following spiroergometries were compared aťt:er Vojta-therapy stimulation and with placebo. We evaluated minute ventilation, breath frequency, tidal volume, oxygen consumption, rate oť elimination of carbon dioxide and respiratory quotient. There were no statistical differences between placebo and Vojta- therapy group at exercise intensity levels 1 W /kg, 2 W /kg and in maximal exercise. Powered by TCPDF (www.tcpdf.org)

The specialization of the final master thesis is theory and a practical view of Location Intelligence. The author is writing about specific parts of development of the application which is focused on analysis of consumer's behaviour for managers of retail businesses focused on customer's satisfaction. The application is a new way of researching consumer's behaviour. Spatial data is connected with the subject of research -- analysis of the movement of consumers in retail stores. The application is a practical tool for better targeting for managers or marketing specialists. The app was developed by four researcher Martin Matějka, Tomáš Rosenberger, Vilém Kalus (three diploma researchers) and Ondřej Funiok. The Main goal of the final thesis was to describe the application from the business point of view and to design layout of the user interface. The business model helped us to specify the application from the business point of view. We reached the main goal with research articles of Location Intelligence. The thesis contains theory basis of informatics and geo-informatics field, Business Intelligence, Location Intelligence and selected business models. The practical part of the thesis is composed of integrating application to the business model, functionality description. From functionality description we designed the user interface. The practical part closes with the use case of application in Vysoká škola ekonomická v Praze. Benefits of the thesis are in development of real, new application for future commercial use. Contribution to the theory of Location Intelligence and analytical solution of development of user interface.

Relationship between practical wisdom and ethical states in Aristotle

In our text we examme the relationship between practical wisdom and ethical states in Aristotle's Nicomachean Ethics. The goal of our work is to define the function of practical wisdom in ethical virtue. We gradually analyze the indiviual parts of definition of ethical virtue from EN II, 6, that is, that virtue is a state concerned with choice which holds the mean relative to us in such a way as a man of practical wisdom would define it. After the analysis of state concerned with choice and practical wisdom we arrive at the determination of practical wisdom as the source of virtuous action; only practical wisdom can be the criterion that pronounces what is bad and good; desire in itself is not capable of deliberation. Following from this a new question arises: how can practical wisdom influence the irrational part of the soul, which is the seat of desire? Practical wisdom, having no direct influence on desire, uses ethical virtue-temperance as it's intermediary. This induces in irrational desire the intention to follow practical wisdom and thus practical wisdom becomes master of desire. We see the contribution of our work firstly in defining the function of practical wisdom in Aristotelian Ethics with greater accuracy and secondly in establishing temperance as a neccessary condition of the other virtues and...

Rehabilitation of cognitive functions in patients after brain injury

The mm of the master thesi s "Rehabilitation of cognitive functions in patients afteľ bľain injury" was to claľify these problems fľom pľactical point of view. In the first chapter readers get acquainted with kinds of brain injury. The second chapter deals with cognitive tlmctions and the third, most extensive, lS dedicated to rehabilitation of patients after brain injury. The ľehabilitation lS conceived as a complex process involving care of patients in the biological, psychicaL social and physical sphere. The empirical part concentrates on training oť attention. Patients after brain injury. who were invol\"ed in the study. vvere divided into two groups. Within three weeks one group practiced on computer (programme RehaCom) and the other attended occupational therapy. We investigated whether there are any diťferences in the influence of training of attention by means oť computer programme or occupational therapy. In regard of a smalI sample (five patients in each group) and other conditions we came to the conclusion that the effect of training oť attention appeared in neither group after three weeks. Kinesiologic Analysis of Selected Muscle Groups in Cross-Country Skiing Using the Classical and Skating Technique

Název: Kineziologická analýza činnosti vybraných svalových skupin při běhu na lyžích klasickou a volnou technikou Kinesiology analysis of activities choice muscular groups at run on cross country skis classical and free technique Cíl práce: Kineziologický rozbor a vytvoření rámcové koordinační mapy vybraných svalových skupin zapojujících se během jednoho pohybové~o cyklu při běhu na lyžích klasickou a volnou technikou. Metoda: Povrchové EMG měření v kombinaci ·s plošnou kinematografickou analýzou pomocí synchronizovaného videozáznamu. Výsledky: U obou běžeckých technik jsme našli fenomén trojextense u odrazové končetiny, který je charakteristický pro lokomoci všech tetrapodů. Nalezli jsme výraznou stabilizační funkci m. gluteus medius a m.adduktor longus. Přítomnost nalezených fenoménů je uváděna i ve volné bipedální chůzi. Z tohoto faktu lze usuzovat, že se jedná o lokomoční aktivitu člověka nacházející se v intencích posturálně pohybové ontogeneze.
