Miloslav Macháček Title of diploma thesis: In vitro evaluation of photodynamic activity of tetrapyridoporphyrazine derivatives for treatment of solid tumors Nowadays, cancerous diseases belongs among the main causes of death, incidence continues to rise. Therefore research in the area of tumor treatment significantly focuses on the development of new potential anticancer drugs and therapeutical approaches. Photodynamic therapy (PDT) belongs to clinically approved therapeutic methods for the treatment of the malignant and nonmalignant diseases. PDT is composed of three main components - photosensitiser (PS), light and oxygen, all of them are non-toxic on their own. However, their interaction leads to a generation of highly reactive molecules. This method is based on absorption of light with appropriate wavelenght by PS and subsequent energy transfer from photon to surrounding molecules, especially to oxygen. This transfer leads to a generation of cytotoxic reactive oxygen species (ROS) of which singlet oxygen plays a main role. Based studio study AG35 is processed by the thesis that the study elaborates in grades documentation for building permits and documentation for the execution of the project. The central theme is Music Club in Brno, for which it was entered plot near the Grain Market Square slope below Špilberk. Near the plot is solved square Obilný trh, park Špilberk, Brno University Hospital and building Ombudsman. The land is sloping, which significantly influenced the design. My main idea was to create a project that would include both a monumental building that will meet and represent the function and at the same time that they are in the best compliance with the terrain of the land. Create a public space where people will resort to relax, or walk to the park we were commonplace. Roofs are walkable, offering views of the surrounding area, larger building houses a cafe and music club itself, in a small building with a test, a recording studio and a ballroom. Both buildings are connected to the underground parking. The very architecture of the buildings plays with the principles of the libre and minimalist style.

Linkage and radiation hybrid mapping of the porcine GNAS1 gene to chromosome 17

The genomic fragment of the porcine GNAS1 gene was amplified by PCR and comparatively sequenced. We identified two polymorphisms, SNP at site 314 and insertion/deletion of 10 nucleotides. Linkage analysis in the Hohenheim Meishan x Pietran family placed the GNAS1 gene to chromosome 17, between GHRH and SW2427. In radiation hybrid mapping GNAS1 was most significantly linked to marker SW2431 on chromosome 17. This position corresponds to 17q23. The GNAS1 gene is located in the QTL region for some carcass traits.

Software for CCD camera used for electroluminescence

This thesis studies the optimization of diagnosis defects of the photovoltaic cells in the darkroom by the help of new operating software for the used CCD camera. First the present situation and the possible solutions are analyzed. Then the attention is paid to the theory of light-sensitive CCD sensors, their construction, their principles of operation and the theory of scanning images. The thesis also focuses on the analysis of the function of the operating libraries which were provided by the equipment producer. The key part is the development of the application for the camera G2-3200 of the "Moravske pristroje" company which significantly accelerates the work with the camera in the darkroom.

Envolvement of Quality Control Tests of Treatment Planning System

This Bachelor´s thesis deals with the treatment planning system Short-term Stability Check. They are carried out according to the Recommendations of the SONS using QUASAR phantoms. These phantoms are expensive, difficult to handle and the methodology for these phantoms of yearly Short-term Stability Check is unnecessarily time consuming. Then it is in the interest of the department that the QUASAR phantoms will be replaced by Inhomogeneity Phantom. Also, the National Radiation Protection Institute lends QUASAR phantoms, so they are not always available. The purpose of this thesis is verified if QUASAR phantoms can be replaced by Inhomogeneity Phantom for Short-term Stability Check. The thesis consists of theoretical and practical parts. The theoretical part deals with the treatment planning system, checks of the ionizing radiation sources and technical equipment. The newly developed methodology is derived from Atomic act and the Recommendation of the SONS and as mentioned above. The treatment planning process and its algorithm are described in an independent chapter. The treatment planning is carried out in the system Eclipse. The system includes many functions, which accelerate the creation, selection and verification of treatment plans. In the following chapter are described the individual checks of the ionizing radiation sources, aimed to verify the accuracy of the devices´ parameters. The Quality Assurance of the ionizing radiation is a condition for the treatment quality and radiation protection of patients and personal. Short-term Stability Check is one of these checks. There is also described the technical equipment, which is necessary for the practical part is impossible. It includes TrueBeam linear accelerator, a 2D detector, phantoms and other equipments, such as wedges, filters. In the practical part there are 2 methodologies, the one of research and the second for Short-term Stability Check of the planning system. The research methodology includes sub-chapters that describe the workflow for Short-term Stability Check. This methodology is based upon information from the manuals of technical equipment. Before introducing the proposed methodology into practice it will be necessary to make its independent verification. Thereafter it will be needed to shorten the methodology to send it to the SONS for assessment. The proposed methodology for Short-term Stability Check of the planning system is in agreement with the on Recommendation of the SONS. In the practical part was verified the planned dose in the planning system Eclipse with measured dose only for checks with inhomogeneities. In this study is formulated the hypothesis: "Can be assumed partial or complete replacement phantoms from the firms Modus or AGMECO by Inhomogeneity Phantom supplied by the PTW company". To confirm or refute the hypothesis was collected data of individual doses. Data collection was performed by irradiating the measurement setup: the detector Octavius 729 + Inhomogeneity Phantom + appropriate number of added plates. In total four energies, 6X, 18X, 6XFFF and 10XFFF were used. Data evaluation was provided by gamma analysis, which assesses the location and size of the point in the dose matrix according to previously specified criteria. The criteria were determined by the table Tolerance. These criteria were verified at most of points. The important thing was that the criterion of 10.0 mm; 10.0% required by Recommendation of the SONS was fulfilled by all points, because it concerns a build-up area, penumbra and interfaces with inhomogeneity's. The research was successfull. It confirmed the hypothesis that QUASAR phantoms may be completely replaced by Inhomogeneity Phantom. In case of practical implementation of this methodology, it should facilitate the work and reduce significantly the time needed to perform the checks.

Neuroregeneration after spinal cord injury and in amyotrophic lateral sclerosis -possibilities for stem cell therapy

Adipose-derived MSC could be used as an alternative for bone marrow MSC in the treatment of acute SCI. We used the intraspinal grafting of rat adipose-derived naïve and predifferentiated MSC to improve motor function after a balloon-induced compression lesion of the rat spinal cord. Grafted cells survived for seven weeks after transplantation, improved motor activity and integrated into the host tissue. They expressed the oligodenrocyte precursor marker NG2 and, occasionally, the astrocytic marker GFAP, but did not transdifferentiate into a neuronal phenotype. Bone marrow MSC may change the disease course and extend lifespan in a rat model of ALS. Combined intraspinal and intravenous transplantation of rat BMSC was performed in symptomatic rats overexpressing the SOD1 G93A gene. Cell-treated animals lived longer compared with sham-treated rats and displayed significantly improved motor activity and grip strength. Rat BMSC survived until the end stage of the disease and were migrating along the white matter of the spinal cord. Examination of cerebeller function in patients with scoliosis

Purpose: The aim of this research was to determine whether there is a difference in the presence of minimal cerebellar signs, hypermobility and dyspraxia in the group of patients with idiopathic scoliosis (IS) and a control group of individuals without IS. Methods: 11 patients have been selected for this research with IS and 11 individuals without IS as a control group. The International Cooperative Ataxia Rating Scale (ICARS) was used for examination of cerebellar signs. Hypermobility was evaluated with ten tests according to Janda. The presence of pendulum reflexes was determined by examination of patellar and triceps reflex, while dyspraxia was analyzed by means of the Czechoslovak test. Results: Patients with IS scored significantly worse in ICARS than the control group (p < 0,05). IS patients presented with significantly more pronounced hypermobility in head rotation (p = 0,0384) and forward bending (p = 0,0409) tests comparing to the control group. No significant difference between the two groups was identified in pendulum reflexes and the Czechoslovak dyspraxia test. Clinical evaluation of postural balance functions in patients with chronic obstructive pulmonary disease

Introduction: Chronic obstructive pulmonary disease (COPD) is one of the most common chronic respiratory diseases. Impairments in exercise capacity, kinesiology and skeletal muscle function are well established in these patients. Recently presented data also suggests impairments in postural balance and increased risk of falls in patients with COPD. The aim of this study is to examine postural balance functions in a group of patients and compare the results with a control group. Methods: Twelve patients (the average age 65,6 ± 7,1, 5 women, 7 men) with COPD hospitalized at the Pulmonary Clinic at the Faculty Hospital, Prague Motol and 10 healthy control subjects (the average age 58,6 ± 5,2, 7 women, 3 men) participated in this study. Participants were measured by The Activities-specific Balance Confidence Scale (ABC) and The Balance Evaluation Systems Test (BESTest). Results: COPD patients scored significantly worse (0,0099, p < 0,05) on the ABC scale total score compared to healthy controls, 78,38 ± 21,14 for COPD versus 97,78 ± 3,88 for controls. The total score and the six subsystem categories score of the BESTest were lower in COPD patients, but not significantly, compared to controls. The effect of cognitive training for schizofrenia or schizoaffective disorder

The dissertation deals with effects of cognitive training on a group of 41 probands suffering with schizophrenia or schizoaffective disorder. The twelve months research has covered several psychiatric facilities, where the cognitive training is carried out regularly. Trial method has been based upon a set of three questionnaires filled in by probands: DEX - (Dysexecutive Questionnaire), CFQ - (Cognitive Failures Questionnaire), EMQ - (Everyday Memory Questionnaire) and variant DEX and CFQ questionnaires, which has been filled in via phone interviews with family members or other significant persons. Experimental design test-retest has been applied. Subsequent statistical analysis of the data collected reveals a significant improvement after the training. This statistically significant positive effect has been shown by four of the five questionnaires used. As for the demographic characteristics, only the age of proband at the manifestation of illness correlates significantly with the positive effect of training (the latter age, the greater effect). Age, sex or education has no relationship with the effect of training for a given group of probands.

Utilizing Internet tools in creating and implementing the strategy of TeskaLabs company

This thesis covers the ways of utilizing internet tools for creating and implementing the strategy of TeskaLabs company. The amount of internet tools that provide various services to companies has risen significantly lately. TeskaLabs is a technology company that is aware of the potential of using internet tools instead of expansive boxed products. The thesis describes what inputs were key during choice of internet tools and what tools became part of the company's infrastructure in the end. Testing of internet tools involved communicators, internet storage services, tools for project management customer relationship management and marketing tools that implement email campaigns, creation of web presentation and internet content and social networks. Overview of functionality of various internet tools is provided and it may serve as a manual for companies that want to exploit the potential of internet services for their good.
