Study of the composition and organization of cytochrome P450 system by covalent crosslinking

The system of mixed function oxygenase (MFO system) participates in significant roles in the metabolism of endogenous compounds and xenobiotics. This system contains cytochrome P450, NADPH:cytochrome P450 reductase, and also there are assigned NADH: cytochrome b5 reductase and cytochrome b5. It was proved that cytochrome b5 can stimulate or inhibit cytochrome P450 (CYP)-dependent reactions and even change the ratio of resulting metabolites. The mechanism of cytochrome b5 action has not been fully elucidated yet. Elucidation of protein-protein interactions in MFO system and determination of topology of this system could explain the mechanism of cyt b5 action. The covalent cross- linking technique is suitable method for identifying protein-protein interactions within the membrane. Cytochrome b5 contains 3 methionines and in 2 cases the methionines are localized in a short hydrophobic C-terminal membrane anchor. Interactions with cytochrome P450 in the membrane environment can be identified by substitution of two methionine for photoactivatable analogue of methionine (photo-methionine) and subsequent photoactivation. Hooligan as an object of film analysis

Final thesis reflects the preproduction and production of thesis film with the topic of radical football fans. Work defines the theme of violence in the world of rowdies and film work with them. Further analyzes not only the history of football fans, but their categorization in terms of sociology and their impact on contemporary society. Work is devoted to the history of Czech cinema, working with the motif of the football world. The reasoning at the conclusion of the thesis reflects the impact of the films on this topic at the general audience and selected subculture.

Court Interpreting in the Czech Republic: the status quo

This thesis addresses the current state of court interpretation in the Czech Republic. The work consists of two parts, theoretical and empirical. The theoretical part describes EU legislation on court interpretation and projects that aim to improve the quality of court interpretation. The thesis also discusses the development of Czech legislation regarding court interpretation from its inception to the present and expands to expected future development in the field of court interpretation. Major institutions such as EULITA at the European level and KST ČR in the Czech Republic are alluded to in the work. The empirical part consists of research conducted by a questionnaire among court interpreters, judges and investigators. The aim of the research is to answer the question who the present Czech court interpreters are and what their co-operation with the judges and investigators is like.

The Literary Works of G. Angiolini and J.-G. Noverre in the context of the 18th century

This dissertation offers a commented translation into Czech language of works written by two most important choreographers of the 18th century, Italian Gasparo Angiolini and French Jean-Georges Noverre. It is a selection of their most important texts where they develop an inovative aesthetics of the dramatic ballet – ballet en action. An introductory chapter about theatre aesthetics of the first half of the 18th century is included in the beginning of the work where I summarize the most important discussions of the period and topics that inspired the choreographers to their reform in the art of dance. Futhermore, a chapter refering to the sources for this work is attached to each commented translation.

THE BACCHAE, FAUST AND LADYBIRDS

This bachelor thesis focuses on themes like the collapse of the myth of God, crisis of faith and seeking God, as well as on way to stage them in a theatrical performance. The first part of this work tries to depict changes in a relationship between a man and God and their mutual combat. The chapter dealing with the cult of Dionysus shows a clash of the two fanaticisms; the religious and the rational one. The Faust chapter is concerned with the idea of free decision making and the difficulties and the possibility of a personal crisis this freedom might cause. The final chapter The Ladybirds opens the problematic of suffering of the innocent, which is the most common objection against the existence of God. The practical part of this work reflects on the school projects and the process of making. It also asks whether it is possible to stage themes like faith in one of the most atheistic nations in the world.

Family housing decision-making

This thesis is concerned with choosing the best and most real estate mortgage loans for family with children. The family should be chosen property near his residence momentary, then it should be recommended for this property the best mortgage loan. Selecting the property was affected by the demands of all family members and subsequent mortgage loans was selected from the products offered on the Czech market in November 2016. The thesis can be thematically divided into three main parts. The first part includes a literature search, which is divided into two areas. First described methods of multi-criteria decision making, which will be used in the decision making process. Along with these methods includes a literature review and theoretical part of the program area Super Decisions, which will be used in the practical part. Furthermore, theoretically they recognized concepts related to mortgage loans, according to which are then laid down criteria for assessing mortgage loans. The second part contains already a practical perspective on family issues, which therefore includes the selection of the best and most real estate mortgage loan program with the help of Super Decisions. The third part includes a summary of the work and the interpretation of results obtained through the program Super Decisions.

Effect of snowpack on runoff generation during rain on snow event.

During a winter season, when snow covers the watershed, the frequency of rain-on-snow (ROS) events is still raising. ROS can cause severe natural hazards like floods or wet avalanches. Prediction of ROS effects is linked to better understanding of snowpack runoff dynamics and its composition. Deploying rainfall simulation together with hydrological tracers was tested as a convenient tool for this purpose. Overall 18 sprinkling experiments were conducted on snow featuring different initial conditions in mountainous regions over middle and western Europe.
Dye tracer brilliant blue (FCF) was used for flow regime determination, because it enables to visualise preferential paths and layers interface. Snowpack runoff composition was assessed by hydrograph separation method, which provided appropriate results with acceptable uncertainty. It was not possible to use concurrently these two techniques because of technical reasons, however it would extend our gained knowledge. Snowmelt water amount in the snowpack runoff was estimated by energy balance (EB) equation, which is very efficient but quality inputs demanding. This was also the reason, why EB was deployed within only single experiment.
Timing of snowpack runoff onset decrease mainly with the rain intensity. Initial snowpack properties like bulk density or wetness are less important for time of runoff generation compared to the rain intensity. On the other het when same rain intensity was applied, non-ripe snowpack featuring less bulk density created runoff faster than the ripe snowpack featuring higher bulk density. Snowpack runoff magnitude mainly depends on the snowpack initial saturation. Ripe snowpack with higher saturation enabled to generate higher cumulative runoff where contributed by max 50 %. In contrary, rainwater travelled through the non-ripe snowpack relatively fast and contributed runoff by approx. 80 %.
Runoff prediction was tested by deploying Richards equation included in SNOWPACK model. The model was modified using a dual-domain approach to better simulate snowpack runoff under preferential flow conditions. Presented approach demonstrated an improvement in all simulated aspects compared to the more traditional method when only matrix flow is considered.

Technical Analysis in Foreign Exchange

The bachelor thesis deals with trading in the forex market. The theoretical part describes the Forex market, its history, when it traded, what is traded, which entities are involved in the market and selected basic concepts, that you must know in this market. There are also basic procedures and rules as the speculative individual should behave on the market in different situations and how to distribute the funds, to avoid a large percentage losses of the capital, as the foundation of financial literacy should think about knowledge. At the conclusion of the practical part, there is the technical analysis, that justifies and predicts the future development of exchange rates. Technical analysis is followed by a practical part, which describes the transactions, that were carried out under real conditions on the currency pair EUR / USD. These transactions were carried out using two technical indicators, Moving averages and Bollinger bands. At the conclusion of the work, there is the evaluation of these two indicators, which one was more profitable for us.

Puppies and young dogs nutritions

Puppies and young dogs nutritions Summary In this work I have attempted to summarize the most important aspects in nutrition of puppies and dogs in growth. The introduction describes the digestive tract and also the changes that have occurred due to domestication. Next is explained the importance of nutrition and water in nutritional requirements of a dog. Important are needs of nutrients and energy that are necessary to maintain the provision of the necessary vital functions. The basic components of food are proteins, saccharides, lipids, minerals and vitamins. I explain the necessary proportions, the amounts and the possible consequences of their lack in food. I focus on essential fatty acids, which in recent years are subject to frequent explorations and their pros and cons are not yet fully understood. I also mention ballast and significant specific active substances. Nutrition of pregnant and lactating females is often neglected, but for puppies it is a very crucial period. The influence of nutrition of pregnant females to the size and weight of newborn puppies has been proved. Further I deal with nutrition of puppies immediately after birth, I explain the composition of colostrum and its effect on the immunity system of the puppy. I also compare ratios of nutrients in the colostrum and milk. Further, I deal with the feeding of the individual at the time of weaning and in different growth periods. There are many possibilities of feeding puppies from after weaning until adulthood described in books. With the new knowledge about nutrition the ways of feeding our dogs have inevitably changed , but also the dogs' nutritional requirements have changed due to the conditions where and how they live and what their nutrient needs are according to their sport, breeding or other use. Currently, there is a possibility of a commercially produced complete nutritional diet or home-prepared fresh food - cooked or raw- barf. In the last chapter, I summarized possible aspects of malnutrition and its potential consequences. I described the most common problems associated with inadequate nutrition of growing dogs such as obesity as well as malnutrition, which also adversely affects the individual. The influence of proper nutrition of the puppy on its growth is clear. If we neglect the nutrition of the dog in the first two years of its life, the consequences are often irreversible.

The Contrast of life and death in works of J. K. Šlejhar

The thesis focuses on J. K. Šlejhar's literary work from two points of view. The first part introduces Šlejhar's work in the contemporary literary context as a result of a synthesis of new literary styles. In the second part the thesis concentrates on a textual analysis and reconstruction of the author's "artistic world". The first part of the thesis gives insight into the contemporary discussions aimed at revising and redefining the function of art with respect to the new development in society and new human needs. The critical reception of realism and naturalism and the reflection of the then wide-spread adoption of new influences from foreign literatures are also dicussed. To draw a comparison, we quote both period and current studies dealing with J. K. Šlejhar's work. Thereby we want to demonstrate the changing requirements of literature and art and the changing perception of a literary work throughout time. The second part of the thesis comprises the textual analysis of Šlejhar's selected writings from the point of view of semiotic literary theory and the reconstruction of the author's "artistic world". Such approach enables us to draw comparisons to Russian literature of the 19th century, primarily with the works of N. V. Gogol and F. M. Dostoyevsky, and to discover a common basis for both...
