Design of the tram tunnel lining

The subject of the diploma thesis is the design of an alternative variant of the tram tunnel lining on the Large Urban Ring Road in Brno Žabovřesky. The first part of the thesis describes the actually implemented solution. PLAXIS 2D software was used for the alternative design, thanks to which the necessary parameters were obtained. The alternative solution of the primary lining consists of fibre-reinforced shotcrete reinforced with two composite meshes. The proposed alternative was found to be economically more advantageous and, last but not least, less time consuming.
Analysis of embryotoxic effect of hydrocortisone using chick embryotoxicity screening test (CHEST).

Cleft lip is one of the most common human birth deffects. Its etiopathogenesis is multifactorial and many aspects of its occurrence remain unknown in the fields of both genetics and teratology. One of the set of known negative external factors causing cleft lip is chemical hydrocortisone. Its effect on cell proliferation is highly heterogeneous and depends on attributes of a specific cell population. In this work we studied the cleft beak origin after the hydrocortisone treatment on the basis of Chick Embryotoxicity Screening Test (CHEST). Our main aim was to detect cell cycle changes in the chick frontonasal process after hydrocortisone injection via flow cytometry analysis. Hydrocortisone caused S phase arrest within a minor subpopulation of highly granular cells with specific cell cycle. This sensitive subpopulation was localized in the areas of previously defined proliferative centers within the frontonasal process using immunohistochemistry of frozen sections. Quantitative analysis of cells in these areas revealed significant decrease of M phase portion in the hydrocortisone treated samples in comparison with the control samples. After successfully avoiding of the destructive scenario with the demolition of reservoirs and replacing them with parking house, the second question arises. How to stimulate the people's interest in these underground jewels, the interest that would confirm the decision to save reservoirs. One of the possible ways, in which I see a purpose and the one that I decided to chose is to make the reservoirs accessible alongside the pathway and let the natural curiosity of human to do the rest.

Technical design of an alternative to the small hydropower plant Ejpovice

The main purpose of this bachelor’s thesis is a new design of a small hydroelectric power plant Ejpovice with non-pressure and pressure derivation. The non-pressure part of a waterway includes design of a weir, culvert and an inlet channel. The pressure part of a waterway includes an intake object and an original tunnel. The design review includes two variants of a waterway. In the first variant water is running straight through the tunnel. The second variant is considering with a glass reinforced pipe that will be installed in the tunnel so the tunnel won’t be flooded. At first the thesis analyses all the sources for the preparation of a design and discribes an area of interest. Below the thesis presents a design of all the new building objects and technological equipments. Detailed restoration of the tunnel is also described. In conclusion of the thesis, there are calculations of a power generation and an economic analysis of both variants. The payback period of the second variant is significantly longer than the period of the first variant although an alternative design doesn’t require a comlicated restoration of the tunnel. On the basis of this fact I recommend to implement the first design.
Design of the High Speed Track between Brno and Znojmo

The aim of this diploma thesis is to design a new track route of high speed line from Brno to Znojmo, specifically between the planned terminal of high speed line Unkovice and existing railway station Hrušovany nad Jevišovkou. The track is designed for a maximum speed of 250 km/h. The alignment and profile of the line were designed, including the design of the railway superstructure and track geometry parameters.
Kuřim south bypass - location study

This diploma thesis deals with the solution of the southern bypass of the town of Kuřim, especially its connection to the road I / 43. The main goal was to optimize intersections and tracing affected communications. The work was based on previously prepared studies, from which were taken over the routes of the road I / 43 and the southern bypass of Kuřim.The proposal slightly modified the height and directional solution of the taken routes. Route road I / 43 is designed to affect the inhabitants of Moravské Knínice. Part of the work are also the designs of relocations of adjacent roads and new solutions resulting intersections. The proposal was prepared with regard to the Principles of Territorial Development of the South Moravian Region.
Application of thermography in the diagnostics of transport infrastructure objects

In order to maintain the proper condition of buildings, bridges, roads and other structures, their regular maintenance is essential. Often, if the distress is detected at the beginning of the disorder, the cost of removing it is far lower than after months of uncontrolled degradation. Locating a distress in time, often means detecting it before it is visible to the naked eye. A key means of detecting hidden distresses is non-destructive diagnostic methods (NDT), including thermographic testing. The purpose of the presented methodology is to provide an efficient tool that will allow a rapid assessment of the state of the transport infrastructure objects and help with the identification of the locations where visual inspection or detailed diagnosis is required. The thermographic measurement is non-contact and can be carried out without the need to exclude traffic on the road. The methodology describes possible ways of using infrared thermography, especially in diagnosis of bridges, roads and tunnels. The presented methodology was prepared within project TE01020168 - Centre for Effective and Sustainable Transport Infrastructure (CESTI), with financial support of the Czech Technology Agency within the Centre of Competence Program. This methodology is intended for road managers at all levels (state, county, city), designers, construction companies and other entities that can use data and processed outputs from thermographic measurements.

Connection of the grade-separated interchange Ostrovačice D1 with highway D52 - Location Study

This master’s thesis is a reconnaissance study developed to create a concept of road connection between the road 43 and the highway D52 in the Czech Republic. The possible route of road 43 is planned both in highway and road category in so called Bitysska trail and is connected to highway D1 at Ostrovacice interchange. This thesis suggests highway and road options to connect interchange Ostrovacice and highway D52 at the section between Ledce and Rajhrad. Suggested options were compared and the most convenient draft option was further worked out.
