Influence of tissue helminths on the development of cancers in model organisms

Mesocestoides corti and Taenia crassiceps are tapeworms, larvae of which are characterized by their ability to reproduce asexually. In this work, the effect of infection by M. corti and T. crassiceps in BALB/c, C57BL/6J and ICR mice on the growth and metastasis of B16F10 melanoma tumors was investigated. Although an increase in metastatic activities was observed after intravenous administration of melanoma cells to M. corti-infected mice, both tapeworms showed a strong suppressive effect on the size and number of tumors and metastases formed when the cells were administered intraperitoneally. This, in some cases, led to a complete elimination of tumor cells. In vitro cultivation of B16F10 cells in the presence of larval excretory-secretory products led to a decrease in their viability but an increase in their migration ability. Flow cytometry proved that M. corti infection has an effect on the increased number and proportion of macrophage populations in the peritoneum of ICR mice. Our work confirmed the anti-tumor effect of T. crassiceps infection in mice and introduced M. corti as a new helminth species capable of influencing cancer. Diplomová práce Vliv a možnosti fyzioterapie u pacientů po onkologicko-chirurgických zákrocích hrudníku - její specifika v předoperačním, akutním pooperačním a ambulantním období Abstract Lung tumor diseases are the most frequent tumors in the world. In men it is the most common tumoral disease in incidence, in women it occupies the third place. Bronchogenic carcinoma is the most frequent one, which includes tumors of trachea, bronchus and lung parenchyma. In most cases, the treatment requires surgical removal associated with hospitalization and subsequent rehabilitation. The theoretical part addresses oncologic surgical issues of lung tumors. A major part of the theory is dedicated to the options of physiotherapy and its specifics in particular parts of rehabilitation care. The practical part observes two groups of patiens and comparatively evaluates the effect of the therapy in particular parts by objective and subjective tests.
Dysregulation of immune system in the patients with Common Variable Immunodeficiency

This thesis includes set of published experimental results, which were obtained at the Department of Immunology, Second Faculty of Medicine Charles University within the project focused on the basal and applied research of the Primary immunedeficiencies (PID), particularly Common variable immunodeficiency (CVID). The first, theoretical part, is divided into two sections. The first section is dedicated to the general aspects of Primary antibody deficiencies (PAD). The second section is focused on epidemiology, ethiopathogenesis, classification, clinical and therapeutical aspects of CVID. The main consideration is devoted particularly to non-infectious complications, which significantly contribute to morbidity and mortality of CVID patients. The second part consists of the set of publications describing specific mechanisms of immune system dysregulation and their clinical manifestation, which are briefly commented. The spectrum of issues resolved within the project covers following basic aspects: 1) characteristics of lung complications in CVID from the point of view of bronchial asthma, 2) characteristics of associated malignancies, 3) significance of genetic background for the specific treatment, 4) therapy of CVID focused on aspects of immunoglobulin substitution. Charakterizace metalothioneinu pomocí kapilární elektroforézy

Bachelor thesis is focused on the study of metallothionein (MT), a low-molecular mass, cysteine-rich protein, whose presence was confirmed across various organisms such as bacteria, fungi, plants, invertebrates and vertebrates. So far, the four isoforms have been described in mammals. The ability of MT to bind metal ions to its structure results in its functions in a number of physiological and pathological processes of living systems such as the influence of metal-ion homeostasis, preservation of the redox ho-meostasis, detoxification and transport of metal ions and regulation of gene expression. Connection between MT and neurodegenerative diseases, apoptosis and cancer is incre-asingly discussed. This work was aimed to create a comprehensive overview of bio-chemical, analytical and functional characteristics of MT focusing on neurodegenerative diseases. The experimental part was devoted to the optimization of the method of sepa-ration of partial isoforms and qualitative and quantitative analysis of binding properties of MT to selected metal ions using capillary electrophoresis.
Molecular mechanisms of carcinogenic effects of alcohol

Alcohol (ethanol) enters the human body mainly through ingestion of alcoholic beverages and its chronic consumption is considered a worldwide socio-economic problem. Besides others, alcohol consumption increases the risk of development of breast, liver, colorectal and upper aerodigestive tract cancer. In the liver, ethanol is metabolised into toxic acetaldehyde which is the main cause of DNA damage leading to cancer development. Acetaldehyde covalently interacts with nucleotides in the DNA forming DNA adducts such as N2 -ethylidene-2'-deoxyguanosine or S- and R-α-methyl-γ-hydroxy-1,N2 - propano-2′-deoxyguanosine. Acetaldehyde can also interact with proteins and disrupt their function. Ethanol metabolism by cytochrome P450 2E1 leads to production of reactive oxygen species, that subsequently damage cellular molecules such as lipides and DNA. Ethanol also initiates carcinogenesis through aberant DNA methylation or interference with retinoic acid metabolism. In cancer development, alcohol interacts with other environmental and genetic factors, which can increase the risk of developing cancer in predisposed individuals.
Smoked meats in human nutrition

Smoked or processed meat is an important part of dietary pattern of czech citizens. Particulary nowadays, its consumation is rather controversial, mainly because of excessive consumation health impacts. Meat in general play an irreplaceable role in our diet. Its a unique source of whole proteins and some mineral substances such as iron. One of the most discussed negatives is, that excessive consumationnof processed meat is linked to development of certain diseases such as obesity, cardiovascular disease, certain types of cancer and type 2 diabetes. We have researched aspects of consumation of certain types of processed meat products specifically portion ratio, mean portion size and consumation afinity according to gender, age and BMI. Following results were found: Largest portions of smoked meats is consumated during lunch with an exception of "vídeňské párky". Men consume more smoked meats than women. Highest affinity was observed in age group till 25 years. In case of BMI - results varied according to the type of smoked meat. In case of prevention is necessary to examine the issue comprehensively and focus not just on eating habits but also on the whole patient's lifestyle.
The knowledge of certain pupils in secondary schools from the area of North and Central Bohemia about selected diseases of civilisation

This diploma thesis deals with the problems of diseases of civilization. The work is divided into theoretical and practical parts. The theoretical part explains what are diseases of civilization, how they arise and how they can be prevented. For each disease (coronary heart disease, obesity, diabetes, cancer) the thesis describes in more detail how and why they arise, what are the risk factors, what preventive measures exist and how to treat the disease. In the practical part, the thesis is devoted to the research of pupils' knowledge in the field of diseases of civilization of selected schools. Using a questionnaire, we dermined the knowledge of the pupils of two secondary schools and found the variance in their knowledge. Furthermore, this knowledge of pupils was compared with their knowledge two years ago when a similar questionnaire was completed in the same schools. The purpose of this thesis was to find out how the knowledge of identical pupils of these two schools changed after two years. The final discussion shows how the knowledge of pupils has changed after two years. It was found that pupils' knowledge improved overall. Cancer, diabetes and obesity knowledge have all improved. The issue of sexual dysfunctions in patients with cancer

The bachelor thesis deals with the issue of sexual dysfunctions in patients with cancer. Objectives: To determine the influence of cancer on the sexuality of patients and the quality of their lives, to describe the issue of sexual dysfunctions. To find out whether nurses are educating patients on sexuality. Methodology and research set: Four research questions were identified. How do cancer diseases and their treatment affect the sexuality of patients and the quality of their lives? How are patients informed about the issue of sexual dysfunction in connection with cancer and its treatment? What is the level of knowledge and experience of nurses with education in cancer patients in the field of sexuality? The qualitative research was used. Two research files were processed by open coding using the pencil-paper method and the data were categorized. The first group consisted of 10 patients determined by the snowball method. Five categories and 13 subcategories were identified. The second sample consisted of 10 nurses selected by the method of deliberate selection of nurses working in the oncology department or outpatient department in the South Bohemian Region. 4 categories and 11 subcategories have been identified. Results: It has been found that nurses do not educate patients in the field of sexuality. They do not have enough experience with education or knowledge, patients are rather taught by a doctor. Oncological diseases affect the sexuality of patients and the quality of their lives. Patients are not sufficiently informed about the issue of sexual dysfunctions related to cancer, they would welcome more information. In most cases, they are shy to ask their problems, the most common sexual dysfunction is the lack of appetite, thus manifested by a lack of interest in sex. They are more often informed about the treatment. Patients are greatly assisted by a partner or family. Use in practice: The obtained results can be used as a possible educational material for nurses to improve communication with cancer clients in the nursing process or as a suggestion for further research investigations.
Quality of life of children after oncological treatment

Cancer in childhood age comprises less than 1% of all diagnosed cancer cases. But is significant, because the childhood cancer is second in mortality, just after accidents. A theoretical part of the study characterizes childhood cancer and its treatment and introduces the term "quality of life". Two goals are set in this work. The first objective is to map the "quality of life" in cancer survivals. The second objective is to analyze how "the attitude towards life" is changed after cancer treatment and whether it differs from healthy children. In this thesis, a quantitative research was used. The standardized SF - 36 questionnaire "on quality of life conditioned by health" and the socio - demographic questionnaire WP 6 - socio - demographic data were used. Three hypotheses were established. H1: "Childhood cancer survivals appreciate their health more than children who did not go through any cancer treatment." H2: "Childhood cancer survivals suffer from cognitive disabilities in comparison to children who did not go through cancer treatment." H3: "Childhood cancer survivals have a more positive relationship to themselves than children who did not go through cancer treatment." The results of this research reveal that young cancer survivors have a similar quality of life as their healthy peers. This thesis should approach the quality of life of children after cancer treatment. The study will be presented at a Professional conference.
