Emerging helminthoses of humans

Emerging infectious diseases (EID) are diseases currently spreading to new areas of the world from the endemic ones. They are rapidly increasing in incidence and their further transmission is highly possible. Mostly they are classified as zoonoses (diseases transmitted from vertebrates to humans). The course of infection can be accompanied by various symptoms; the cases vary from asymptomatic to, chronic conditions, leading in some cases to serious complications and death of the host. Spreading of EID can be influenced by many factors, e.g. different socio-economical, cultural, and abiotical factors. It can be influenced by activity of different vectors, and transport of intermediate, reservoir, and final hosts. This work is focused on the modes of spreading of four parasites: Ancylostoma ceylanicum, Echinococcus multilocularis, Dirofilaria repens and Angiostrongylus cantonensis. Heating of a recreational building using renewable resources

The bachelor thesis deals with the design of heating a recreational building in Vysoké Mýto using a heat pump. The theoretical part deals with the topic of hot water design. The computational part of the bachelor thesis contains a design for heating a recreational building using hot water underfloor heating. The heat source is an air-to-water heat pump. The calculation also includes the preparation of hot water and the design of all necessary system equipment.
Energy Performance Certificate of the Building

This bachelor thesis deals with the evaluation of the energy performance of the Mars hotel in Špindlerův Mlýn. The theoretical part of this thesis is focused on the conversion of sunlight into electric power using solar-powered photovoltaics. The practical part is focused on the analysis of energy consumption and energy flux of the inspected building. The final part C concerns about the result of this thesis – the energy performance certificate of the analyzed building.
The immune response of naïve mice infected with the neuropathogenic schistosome Trichobilharzia regenti

Helminth neuroinfections represent a serious health issue, but the mechanisms of the host immune response often remain neglected despite the fact they might contribute to pathogenesis. This is partly due to the unavailability of clinical samples and the lack of suitable laboratory models. Herein, I focused on the characterization of several aspects of the immune response of mice infected with the neuropathogenic avian schistosome Trichobilharzia regenti. After the percutaneous infection of mice (accidental hosts), most T. regenti schistosomula are entrapped and eliminated in the skin, but the parasite antigens initiating the protective immune reaction are not known. Our in vitro experiments revealed that T. regenti cathepsin B2, a cysteine peptidase used for the skin penetration, activates bone marrow-derived dendritic cells much stronger than the parasite homogenate, suggesting its role in initiating the mixed type1/2 host immune response. However, some schistosomula manage to escape from the skin and continue their migration to the spinal cord. Influence of tissue helminths on the development of cancers in model organisms

Mesocestoides corti and Taenia crassiceps are tapeworms, larvae of which are characterized by their ability to reproduce asexually. In this work, the effect of infection by M. corti and T. crassiceps in BALB/c, C57BL/6J and ICR mice on the growth and metastasis of B16F10 melanoma tumors was investigated. Although an increase in metastatic activities was observed after intravenous administration of melanoma cells to M. corti-infected mice, both tapeworms showed a strong suppressive effect on the size and number of tumors and metastases formed when the cells were administered intraperitoneally. This, in some cases, led to a complete elimination of tumor cells. In vitro cultivation of B16F10 cells in the presence of larval excretory-secretory products led to a decrease in their viability but an increase in their migration ability. Flow cytometry proved that M. corti infection has an effect on the increased number and proportion of macrophage populations in the peritoneum of ICR mice. Our work confirmed the anti-tumor effect of T. crassiceps infection in mice and introduced M. corti as a new helminth species capable of influencing cancer. Solar hot water storage tank

The solar hot water storage tank is off great importance in the solar collector array. It allows transformed energy accumulation thus deals with the inconsistency between supply and demand. The suitable design of the storage tank can improve system efficiency. The storage capacity represents the balance between the amount of stored hot water and the tank's heat losses. The design of the storage capacity is based on three hypotheses. The coverage of hot water demand by solar energy, the ratio between storage capacity and solar thermal collector area and the prediction that the storage capacity corresponds to one- to twofold hot water demand. The purpose of this thesis is to share an understanding of the solar storage tanks design and to improve the design through numerical simulation, experimentations and general calculations. It also focuses on the confirmation of the used hypotheses and determination of the best way to design the solar storage tank for general practice and further potential discussions. The simulation model has three variables – the storage capacity, collector area and the number of occupants. The intent is to find the interdependence of these three variables. The purpose of the simulations is to modify the design of the solar tank based on the mutual influence of studied parameters. The modifications are performed based on the users' needs.
Energy self-sufficient mountain guesthouse

The aim of this master project is to design an off-grid nearly-zero energy mountain guesthouse. First, the building structure is designed. It is a two-storeys building with roof made of lattice trusses. On first floor is dining room, kitchen, playroom and office. On second floor are guest rooms and an owner´s flat. The vertical load-bearing structures are designed from ceramic blocks. Horizontal load-bearing structures are designed from reinforced concrete monolithic slab. The building envelope is insulated with ETICS. Second, HVAC, lighting, photovoltaics and use of rainwater is designed. TZB system controlled remotely with a PC or cellphone are designed. Third, energy study is made. The project is developed using CAD (drawings), DEKsoft (thermal calculations) and Atrea Duplex (air conditioning design).
Heating and cooling systems in a multifunctional building

The master´s thesis is about the design of heating and cooling of a multifunctional house. The building is functionally divided into parts of shops, administration and apartments. Thesis describes various cooling options and basic types and functions of air conditioning systems. Three variants of the system at the level for expanded building permits and their evaluation are developed. The selected variant is developed at the project level for the construction.
Analysis of boundary conditions of ground heat exchangers

Diplomová práca,, ,,Analysis of boundary conditions of ground heat exchangers,, je primárne zameraná na analýzu okrajových podmienok a hlavných faktorov vplývajúcich na spoľahlivý návrh a prestup tepla plošného zemného výmenníka v oblasti geotermálnych aktivít na kúpeľnom ostrove v Piešťanoch. Posúdenie možných vplyvov na životné prostredie, ako je napríklad vznik tepelných zmien v dôsledku extrakcie tepla vrátane zamŕzania pôdy v okolí potrubia a potenciálneho znečistenia podzemných vôd je taktiež rozoberané. Súčasťou tejto záverečnej práce boli spracované tri numerické analýzy metódou konečných prvkov pomocou softvéru COMSOL Multiphysics 5.6., ktoré sa môžu vyskytnúť v tejto konkrétnej oblasti. Prvá numerická analýza je podrobená štúdii dynamických procesov plošného zemného výmenníka tepla a zeminy nasýtenou vodou pri konštantnej teplotnej okrajovej podmienke pôdy. Dynamický proces zemného výmenníka tepla a nasýteného podložia pri nestálej teplotnej okrajovej podmienke je skúmaný v druhej numerickej analýze. Posledná analýza je vyhotovená na posúdenie prevádzky plošného zemného výmenníka tepla s možnosťou prúdenia podzemnej vody v rovnakej vrstve. Vďaka týmto numerickým štúdiám a výsledkom dynamických tepelných modelov, každá prípadová štúdia obsahuje rozloženie teplôt v zemine, výstupné teploty z výmenníka a vlastný čiastočný záver.
Solar collector type influence on the heating solar water tank

The diploma thesis deals in the theoretical part with various types of sys-tems for the preparation of hot water and specifies the types of solar sys-tems. In the calculation part, it solves the design of kindergarten heating by a system of heating elements. It also solves two variants of hot water preparation in the hot water tank. The design also includes all the equip-ment needed for the proper functioning of the heating system. The exper-imental part compares flat panel and evacuated tube solar collectors.
