HPLC Determination of Potential Drugs

1 ABSTRACT Charles University Faculty of Pharmacy in Hradec Králové Department of Biophysics and Physical Chemistry Candidate: Karolina Palickova Consultant: Ing. Vladimir Kubicek, CSc. Title of Thesis: HPLC Determination of Potencial Drugs The diploma thesis describes selection of the most suitable conditions for HPLC determination of a recently prepared potential bronchodilatans. The aim of the thesis was to find a suitable isocratic method for the HPLC determination of the substance in rat plasma to be used during pharmacokinetic experiments. The chromatographic column Ascentis® Express RP - Amide was utilized. Best results were achieved with mobile phase consisting of acetonitrile and phosphate buffer (pH = 3,0) in volume ratio 12:88. The column was thermostated at 20 řC. Fluorescence detection (λex = 228 nm, λem = 380 nm) was applied to get a sufficient sensitivity. 4-quinazolinole was chosen as an internal standard. Biological samples were processed by LLE before the HPLC analyses. The proposed method was validated successfully and then employed in pharmacokinetic studies.
Regulation of advertisements in pharmaceutical industry

Abstract, Jan Metelka The main aim of this diploma thesis was to firstly explain the field of human pharmaceuticals and regulation of advertisement regarding them, from the general point of view and then in details. In order to achieve this it was required to define advertisement and pharmaceuticals because these terms and crucial for the whole thesis. Both definitions may be off the scope of this text however it plays an important role in understanding all necessary issues. Advertisements in pharmaceutical industry have some specifics as described in chapter 2. There are two kinds of recipients - experts (doctors and pharmacists) and ordinary people (mostly patients). Every kind is special and regulated in a different way. Third chapter described all steps in administrative proceedings related to Act on Advertisement, forming a public branch of law against unfair competition. It summarises and divides all misdemeanours, statistically describes how often they occur and deeply analyses possible breaches of Act on Advertisement. The main part of this thesis is Chapter seven, dealing with unfair competition in private law, as mentioned in early chapters, however here it is specifically described how to apply the general clause and the more important kinds of unfair competition such as deceptive adverts and...

Title of Diploma Thesis: Physico-Chemical properties of drugs To test physico-chemical properties of new molecules is necessary during drug development. It could be helpful to understand or predict the pharmacokinetic parametres of a new drug in vivo/in vitro experiments. One of this parameters is a dissociation constant (pK). Dissociation constant is defined as " Number on pH scale, wherein is just fifty percent of molecule in a ionization condition". In real case this number can help us to know where in the gastro-intestinal tract (GIT) the drug will be absorbed. In GIT only molecules exhibiting pK from 3 to 11 could be absorbed. Out of this range it is not possible. In this work I would like to introduce the ways of experimental measurement of pK values. I was working with two methods to measure the pK values of water-soluble compounds. The spectrophotometric method and the potentiometric one. Multicomponent Analysis

Title of Diploma Thesis: Multicomponent analysis This diploma thesis deals with the determination of concentrations of individual dyes in mixtures by multicomponent spectrophotometric analysis. The method was tested on selected dyes which were divided into two groups. The first group contains methyl orange, methyl red, orange II and phenol red. The second group consists of methylene blue, bromocresol green and thymol blue. Within each group mixtures were prepared containing combinations of two, three and four dyes in various ratio. Spectra of prepared mixtures were measured by diode array UV/VIS spectrophotometer. Absorbance values were read at wavelengths of absorption maxima of the individual dyes. Using the pseudoinverse matrix, concentration ratios of components were calculated from the absorbance values. The determination in determined and over determined systems was performed. On the basis of the results obtained by the method of analysis it is possible to determine the individual concentration of the multi-component dyes mixtures without prior separation. Determination of Lipophilicity of Drugs by HPLC

Thesis title: Determination of Lipophilicity of Drugs by HPLC In this thesis a RP-HPLC method for fast and reliable determination of lipophilicity was proposed and tested. Stationary phase was selected by using hydrophobic substraction model. Capacity factors of the chosen substances were initially measured on Zorbax ECLIPSE XDB C18 250x4,6 mm, 5µm column, which exhibits almost identical retention characteristics as the column used for this purpose until now. Then the capacity factors of the same substances were determined by using Zorbax ECLIPSE XDB-C18 50x4,6 mm, 1,8µm column that was selected to reduce retention times significantly. A group of newly synthesised drugs based on structure of pyrazine served as samples for the measurement. The reproducibility of the capacity factor values determined using both columns was compared and the independence of the capacity factor on the mobile phase flow was confirmed. The diploma thesis deals with witchcraft and other mystic phenomena in conception of Edward Evan Evans-Pritchard. It is focused especially on the notion of witchcraft in connection with magic, shamanism and oracles. It describes relation of all these phenomena towards religious belief and various attitudes towards them. It defines respective practices and explains differences between them. Last but not least, it introduces usual reactions to them. Via concrete situation it also shows interconnection of all described phenomena and presents them as a coherent and logical system.
Intravenous Drug Administration and Dilution

The goal of this elaborate work was to describe the most often medication misconducts, errors and risks during the intravenous drug administration and dilution in the field of primary care in the Intensive Care Unit - ICU. My interest was to find out about local ICU's standards and regulations and to survey how knowledgeable nursing stuff is towards medication hazard regulation and its affect. The first theoretical part concentrates on conception of IV drug administration and dilution. It charts and shows the situation at the current time. Empirical section of my elaborate work shows findings based upon questionnaire. It serves the purpose of hypothesis verification and confirms the goals of my study. Obtained information should be a foundation for future processing and utilization standards regulation or recommendation how to safely administrate intravenous drugs.
Electronic auction in healthcare

Working with the theoretical part focuses on the regulation of electronic auctions in terms of legislation both Czech and foreign, given that we are members of the European Union. Furthermore, the work explains the possible ways to use electronic auctions, which are mostly used in both the Czech Republic and abroad. At work I also am trying to shed light on the current use of auctions on the auction portal Aukro examples, which is widely used by people for personal use. Finally, the theoretical part I try to outline the future development of electronic auctions. The practical part is focused primarily on its activities in the hospital Pardubice Region, Inc. where I introduced the auction. In several examples, visible results, which not only brings centralization entire purchase, but also the use of electronic auctions as a tool for communication and bidding suppliers with the lowest purchase price for a group of hospitals. The work also draws from the Ministry for Regional Development, which is responsible for the preparation of the most important regulations for electronic auctions in healthcare, namely the Public Procurement Act. Electronic auctions are a tool very interesting, because it is the most transparent way of evaluating procurement by parameters rates. Whether if electronic auction in health care and efficient purchase option will be discussed in this work.

The purpose of this bachelor tesis is to make an overwiev about contemporary state of knowledge about infusion pumps. Labour is locate to infusoin pump characteristic and constructional solving of kinetic mechanism. First part is a characterization of products from inland and external producers. Second part is pointed to the opinion poll of infusion pumps use. Close of this labour is oriented to knowledge summary and incoming development of infusion pumps.
