Abilities and importance long-term cultivation embryonic cells from different species of animals

Embryonic cells inside the blastocyst, which divide and differentiate into cells of all tissues of organism, are called embryonic stem (ES) cells. Using the ES cells for transplantations is one the potential ways in regenerative medicine. The present study deals with two different lines of mouse pluripotent cells: embryonic carcinoma (EC) cells line P19 and ES cells line D3. The in vitro cultivation was performed in basal cultivation medium with fetal bovine serum (FBS) and in addition of leukemia inhibitory factor (LIF) in case of ES cells. The neurodifferentiation was induced by medium without FBS and without LIF in case of ES cells and supported by retinoic acid in case of P19 cells and embryoid bodies cultivation in case of ES cells. Pluripotency and neurodifferentiation of the cells were confirmed by presence of molecular markers of pluripotency and neurodifferentiation in both lines in nondifferentiated and differentiated states. Mouse EC P19 cells were transfected by gene for green fluorescent protein (GFP) and used for transplantation in cerebellum of wild type (WT) and Lurcher (Lc) mutant mice. Nondifferentiated cells (P19) and neuroprogenitors (NPG) derived from these cells were used. The study followed the survival, morphology and localization of the grafts. Security of electronical shops built on PHP platform

This thesis is focused on security of electronical shops built on PHP platform. The goal of this thesis is to analyze electronical shops security and personal data protection from view of czech legislature, trustfulness of these shops from view of customers and also certification of these shops, analyze various possibilities of payments and assuring their security and analyze possible security risks, which are matched with technologies these e-shops are built on. Analysis of these appropriateness is based on theoretically knowledge extracted from scientific sources and from author's experiences. Benefit of this thesis consists in reference to possible inadequacies of e-shops in personal data security, on-line transfers and security risks derived from PHP technology, which they are based on. This thesis is comprehensive survey of things, which quality e-shops should have ensured and contains also summarized survey of security risks, which web developer should minimize during web creation. This thesis offers its own resolution of security risks and also chosen real e-shops evaluation. This thesis can be useful not only for web makers and e-shop programmers, but also for e-shop customers.

Protection of basic Human Rights in the African System (Zimbabwe) - and its Comparison with the Continental legal system

The diploma thesis in question attempts to map the African regional system of human rights protection, its basic elements, central institutions and the results of their cooperation. The work aims to find out whether the current set-up is capable of ensuring an effective protection of human rights of the African citizens. The first part of the thesis focuses at the definition of human rights in their historical and conceptual context. For the needs of the comparative analysis, a part dealing with the protection of human rights in the continental legal system follows, together with a suggestion of the "superstructural" options that the citizens of the member states of the European Union have disposal of. The African legal system as the guiding line of the thesis is applied on the case of Zimbabwe. This is where practice shows that despite the existence of a formally existing system under the African union and its institutions, the African region is facing serious human rights violations for which there is no sufficient safeguard. The thesis concludes with the provision of a generalizing comparison of the two regional mechanisms of human rights protection and the application of this area of study in the sphere of education and teaching practice.

Problematika zpracování a následného využití čistírenských kalů

Sludge treatment and its processing problematics and subsequent use of sewage sludge are nowadays much discussed issues. Requirements for ecologic and environmentally friendly disposal of sludge are continuously increasing. These requirements lead to an increase of energy, technology and economic performance of sludge treatment, particularly in relation to the current legislative conditions in this area. The issue of sludge treatment is therefore one of the main topics in the operation of wastewater treatment plants in Modřice, which serves as the "central" wastewater treatment plant for Brno city and its surroundings. Nowadays is sludge in Modřice stabilized by anaerobic mesophilic fermentation and sanitized by drying or liming. Values of proposed parameters and operational data on the input to the slurry line in some cases differ, e.g. sludge concentration in the reservoir of the primary sludge, and the concentration and volume flows in the sludge thickening tank. This is due to the nature of entering waste water (higher total pollution in them) and limits of subsequent technologies. Despite these deficiencies are achieved proposed and required legislative parameters of sludge in output.

Social Capital: Usefulness of the Concept

The vast amount of theoretical and empirical work on social capital that has inundated the social sciences, since the beginning of the nineties, can be roughly divided in to two categories. Firstly there are those writers, mainly political scientists and economists, who follow Putnam's line of reasoning and tend to see "associational membership" more as a source of social capital than as another form of it. Secondly those, mainly sociologists, who tend to treat the concept of social capital as primarily a social structural variable, which can be used to refer to social network or to the linkages between individuals and/or organizations. The later theorists, represented by writers such as Edwards and Foley (2001b), and Lin (2001a, 2001b), usually associate social capital with facilitating goods and service flows among individuals and organization. Through a historical overview, discussion on the major conceptualizations, as well as special characteristics and perspectives of social capital, we try to clarify the social capital debate and clearly differentiate between the two approaches. Subsequently, we present a thorough critique of Putnam's approach to social capital, as well as various arguments questioning its conceptualization and use. Emergency Call Service 112 and its Possibilities in Receiving Emergency Calls from Handicapped Citizens

This bachelor thesis deals with the possibilities of calls made by handicapped people (deaf, blind or physically handicapped) to emergency lines, including line 112. According to the European Parliament Directive 2002/22/ES of 7 March 2002 on universal service and users' rights relating to electronic communications networks and services, specifies that each citizen, whether healthy or handicapped has the right to call to emergency line 112 free of charge. This thesis involves a questionnaire based research. Sets of questions focused on handicapped citizens asked whether they had sufficient possibilities to call to emergency lines. The questionnaires were distributed to handicapped citizens in South Bohemia. The aim was to find out whether the efforts of the Fire Rescue Service in the field of reception of emergency calls from handicapped people were sufficient or not and whether these citizens are or will be interested in using the line in the future. The research has shown that handicapped people may communicate with the emergency lines by means of mobile phones like healthy people, provided their handicap does not prevent them from doing that. Blind and sand-blind people do not have problems with telephoning either, mostly from mobile telephones. They might only have a problem with specification of the event location. Deaf and hearing impaired people actually have problems with mobile phone communication, but the Fire Rescue Service of the South Bohemia Region enables them to send SMS, thanks to which they can receive assistance of the same quality as any healthy person in emergency situation occurrence. As for legislation, a new draft Fire Rescue Service Act, which contains the issues of emergency calls from handicapped people, is being consulted at present.

Experimental system for the mouse polyomavirus life cycle study Abstract: Murine polyomavirus (MPyV) is the prototype of the Polyomaviridae family. This family includes also some important human pathogens (BKV, JCV, Merkel cell polyomavirus). Due to their specific properties viruses within this family may serve as versatile vectors for gene therapy or recombinant vaccine production. New methodological approaches may help to understand some yet unknown facts about MPyV life cycle. Clarification of some processes during murine polyomavirus life cycle may be also important to fully exploit polyomaviruses for therapeutic purposes. The aim of this diploma thesis was to preparare two innovative experimental systems that extend possibilities of studying the life cycle of MPyV. The first part of the diploma thesis focusses on construction of recombinant MPyV which expresses yellow fluorescent protein (EYFP) in the early stages of infection. Such virus can be very useful for studying the infection spreading by live- cell imaging and Fluorescence-Activated Cell Sorting (FACS) and can be employed for co- localization studies of YFP-tagged LT antigen with certain cellular proteins. "Centrum" Department Store in Brno

The subject of the thesis is an architectural study of the reconstruction and extension of the "Centrum" department store in Brno. It is a dominant building designed by the famous czech architect Vladimir Karfík, which is located at the Urban Conservation Area Brno. Object wasn´t realized with the full extent of the original proposal, so in the project is planned a completion of it. The new solution respects the historical quality of Urban Conservation Area Brno with a concurrent view of the location on the lucrative land in the city center. The architectural concept is based on a return to the original idea of the architect and the addition of my own creative contribution within the extension of the new floors. Draft of the extension is intended so that the individual parts of an object communicate with each other, but they are easily recognizable as well. This idea was filled by both parts linked together visually by horizontal lines and ripple, which is used on balconies of the extension and behind the facade of the current building by shielding walls. However it is emphasized the contrast between an enclosed mass and a free-flowing space. The existing part of the building is used especialy as a leasable area of trade and services. On the upper floors there are areas of administration. Furthermore, there will be a restaurant and cafe with exhibition galleries and apartments are designed on the new added floors. The building is adapted to use by person with limited mobility. Newly added uses in the object will increase the attractiveness of the building and its surroundings.

Clinic for infectious diseases

In this thesis, we address the petition of new infectious clinic at the documentation for construction. Location the building is designed in the northern outskirts of Šumperk. The building is an irregular shape with 3 storeys with 1 basement. Developed building area is 1180,08 m2. The building is a movement divided into three parts, and to: object A- communication area for meals, object B- children's department, object C- building department for adults. The supporting structure is made of masonry materials. Ventilated facade to the building is from the outside, lined with stone lining- slate / wood lining- Siberian larch. Draft internal layout reflects the requirements of standards for hospital then submitted demands to avoid danger to persons health due to the occurrence of infectious diseases. The thesis also includes two special projects: Design of reinforced concrete slabs petition air conditioning. Projects are handled over selected parts of the Clinic of Infectious Diseases.
