Piezoelectric nanofiber materials for wearable electronics

This thesis deals with the influence of production parameters on the morphology of PVDF nanofibers produced by electrospinning. Nanofibers are generally a widely used material, nanofibers are generally a widely used material, they can also have piezoelectric properties, which gives them a specific spectrum of use, eg in biosensors. The production of nanofibers was realized by electrospinning. A scanning electron microscope was used to examine the morphology and surface properties. The aim of this thesis is to compare the properties of PVDF nanofibers produced under different settings of production parameters.
Study on biotechnological potential of thermophilic gram-positive bacterium Brevibacillus sp. Bz

Předmětem předložené diplomové práce je studium biotechnologického potenciálu termofilní grampozitivní bakterie Brevibacillus borstelensis BZ. Teoretická část obsahuje obecnou charakterizaci termofilních organismů a jejich termozymů. Popisuje také adaptivní molekulární mechanismy, které zajišťují termostabilitu těchto proteinů. Závěr teoretické části je věnován biodegradaci odpadních substrátů a polymerů na bázi přírodních a fosilních zdrojů. První část experimentální práce se zabývá produkcí hydrolytických enzymů na různých původních zdrojích a odpadních substrátech. Díky intenzivní enzymatické produkci na původních zdrojích je Brevibacillus borstelensis BZ považován za velmi slibného producenta termostabilních enzymů, konkrétně xylanáz a celuláz. Testovaný bakteriální kmen BZ byl schopen produkce termostabilních enzymů i na odpadních substrátech. Na vybraných substrátech kmen BZ přednostně produkoval xylanázy. Díky tomu byla xylanázám věnována zvláštní pozornost, kdy bylo testováno teplotní a pH optimum. Závěrem experimentální práce byla testována schopnost bakterie Brevibacillus borstelensis BZ degradovat vybrané polymery na bázi přírodních a fosilních zdrojů. Kmen BZ poskytl nový pohled na biologický rozklad polyethylentereftalátu (PET), amorfní frakce kyseliny polymléčné (PLA), semikrystalické PLA a polyhydroxyalkanoátů (PHA). Pozorováním povrchu PET fólie skenovacím elektronovým mikroskopem (SEM) se potvrdilo zdrsnění materiálu, přítomnost rýh a naprosté pronikání bakterie skrz fólii. Pokud jde o polymery na bázi přírodních zdrojů, PHA granule byly zcela degradovány. Studiem morfologie povrchu obou zmíněných PLA bylo prokázáno jasné zhoršení jejich struktury přítomností jam a trhlin na povrchu polymerů.
Condition of dorsoabdominal scent glands in adults of the true bugs from the taxon Pentatomomorpha (Heteroptera)

Existence of larval dorsoabdominal scent glands (DAGs) together with occurence of adult metathoracic scent glands is one of the important autapomorphic characters of taxon Heteroptera within taxon Insecta. DAGs can persist until adults in some species of this taxon. The persistence of DAGs were proved also in members of infraorder Pentatomomorpha. Targets of this thesis are: collect the knowledge about persistent adult DAGs in selected representative members of important families of taxon Pentatomomorpha; study and compare the condition of the DAGs in the oldest larval instars and adults; clarify the significance of existence the openings of conducting ductules of proper glandular units in reservoir intima of DAGs in study of the persistence of these glands. For study were used stereomicroscope, light and scanning electron microscope. Temperature drift compensation for nanostructure analysis

An ultra high vacuum apparatus for nanostructure experiments is being developed by the Tescan company in cooperation with the Institute of Physical Engineering of the Faculty of Mechanical Engineering. The apparatus is designed for preparation, modification, and analysis of nano-scale structures. A sample holder which allows heating and cooling has been developed for the apparatus. A scanning electron microscope is attached to the chamber to provide analytical and manufacturing capabilities. During experiments with heating enabled, the sample moves relative to the SEM column, causing drift of the image. This bachelor thesis proposes a solution to the problem of temperature drift by the means of motion tracking based on image registration using Fourier transform. An application complementary to the SEM control software which implements the algorithm was designed and tested at the instrument.

Design of the electron microscope

The theme of this diploma thesis is the design of a scanning electron microscope with a focused ion beam. Specifically, the thesis is focused on the design of the microscope covers and the adjacent workplace of the operator for Tescan Orsay Holding a.s.. Design is solved as the first proposal aimed at future innovation of the design of the entire product line.
Analysis of non-metalic inclusions in motor heads castings and suggestion on their remove

Improving the quality of metal castings for serial production of automotive industry, leading to improvements in the properties of casts and their lower costs. This publication analyzes the non-metallic inclusions contained in casts of heads of internal combustion engines, molded of Nemak Czech Republic, Ltd. Tackling includes analysis of size, shape, chemical composition and hardness of the specific non-metallic inclusions. The thesis describes the purity metal holding furnaces and proposals to eliminate inclusions from metal.
Characterization of structures fabricated by selective wet etching of silicon

The task of master’s thesis was to perform optimalization process for preparing metal etching mask by electron beam litography and subsequent selective wet ething of silicon with crystalographic orientation (100). Further characterization of etched surface and fabricated structures was performed. In particular, attention was given to the morphology demonstrated by scanning electron microscopy and study changes of the optical properties of gold plasmonic antennas due to their undercut.
Design of electron microscope

The aim of my Master's thesis is the design of a scanning electron microscope. The designed concept presents an innovative approach to the problems and respects all technical, ergonomic and aesthetical demands made on it. The main creative part of design process starting from the concept development and ending up with the final solution is preceded by a background research study including a historical, technical and design analysis of the electron microscope. The conclusion of my Master's thesis analyses the final design, its character and its contribution in broader context.
Freezy sublimation in cryoattachement Gatan Alto 2500 connected to scanning electron microscope JSM-7401F

This thesis deals with freezy sublimation in cryoattachement Gatan Alto 2500 connected to scanning electron microscope (SEM) JEOL JSM-7401F. The thesis is devided into the theoretical and practical parts. In the theoretical part I focus mainly on physical qualities of water and on problematics of freezy-drying. I also describe qualities of subsidiary substances which are being used when working with cryotechnology. I briefly describe technology used: cryoattachement Alto 2500 and SEM JSM-7401F. In the practical part I deal with the description of the method that I have developed in order to measure weight decrease of frozen specimen owning to its sublimation in vacuum. I provide measurement results of the sublimation of these substances: demineralized water, Dextran, glycerol and phosphate buffer saline.
