Role of UNHCR in case of asylum seekers, refugees and stateless persons – the case study of Turkey

Inspired by the Kratochvíl, Cibulková and Beník´s model of actorness, the thesis explores the framing power of UNHCR. The critical discourse analysis introduced by Norman Fairclough is applied in order to analyze the textual, contextual and sociocultural dimension of the UNHCR framing power. The case of Turkey was chosen for this study due to the current situation and the fact that Turkey is one of the biggest hosting country in the refugees´ crisis. Through the critical discourse analysis, the thesis explores whether UNHCR is considered as a framing actor in the case of asylum seekers, refugees and stateless people in Turkey. The analysis shows that UNHCR is globally respected and takes important part in the issue. However, not all of the international organizations refer to UNHCR as the lawmaker and the initiator of debates in the issues mentioned above.

International involvement in the peacebuilding process on Sri Lanka

The final months of Sri Lankan civil war, the 26 year military campaign between the Government of Sri Lanka and the insurgent separatist organization Liberation Tigers of Tamil Eelam, are marked by allegations of war crimes committed by both parties of the conflict, including attacks on civilians, summary executions of prisoners, enforced disappearances, restrictions on humanitarian assistance and recruitment of children. Since the attempts to put the Sri Lankan issue on the agenda of the United Nations Security Council failed on the unwillingness of China and Russia, the UN bodies in Geneva have taken a leading role in promoting the peacebuilding process on the island. The thesis aims to analyse the impact of the Geneva-based human rights bodies, in particular of the United Nations Human Rights Council, on the process of reconciliation in Sri Lanka, for which is crucial that the alleged violations of human rights in the last months of the civil war would be properly investigated and that the perpetrators would be held accountable. In order to put the activity of the UN Human Rights Council into broader context, the paper will examine the political development in Sri Lanka since the end of the civil war in 2009 as well as the failed efforts of Sri Lankan governments to launch the process of reconciliation.

Policy development and implementation: case study of social housing

The diploma thesis focuses on the process of creation and implementation of public policy which is examined on the social housing policy. The aim of this thesis is to explore how is a certain policy prepared and put into use, considering the social housing policy as an example. Therefore, the key research question inquires how the process of creation and implementation of public policy looks like. The first chapter of the thesis deals with theoretical background of the public policy process, stressing the role of public policy documents. The second chapter describes the area of Czech social politics and policy as a cover for social housing policy development. The third chapter examines the history of Czech housing policies. The final chapter focuses on the development of social housing policy and examines its connections with certain public policy documents.

T cells labelling by bimodal contrast agent for in vivo studies of stroke

Stroke is a serious brain injury, which causes sudden death or terminates in permanent neurological disability. Nowadays, tissue plasminogen activator (tPA) is used as the only effective treatment of stroke. One of the potential targets for novel therapy are T cells. Even though the explicit role of T cells in the pathogenesis of brain injury, amounts and timing of all T cell subtypes infiltrating into brain during the stroke still needs further investigation. The research in this field is complicated by the lack of efficient methods for in vivo cell tracking. Therefore the aim of this thesis was to develop a method of T cells labelling by MRI contrast agent in order to investigate T cells distribution in ischemic mice model using in vivo MR imaging. T cells were isolated from C57/BL6 mice in two step isolation protocol using gradient centrifugation and magnetic separation with the efficiency of 97 %. The isolated cells were labelled with 100 μg Fe/mL of Molday ION Rhodamine B contrast agent. The labelling efficiency after 17 hours of cells incubation was higher than 99 %. The labelled cells were cultured with CD3 and CD28 antibodies resulting into the 74 % viability of labelled T cells compared to 83 % viability of non labelled T cells. Methods and ways of implementation of changes and innovations in companies through project management are in today's society very well established. There exists methodologies, techniques and tools for the management of individual projects. However in a role of project manager, I very often faced the fact that companies are performing the project portfolio management in very intuitive way. This in itself leads to failed and prematurely terminated projects, initiation of the wrong projects or realisation of correct projects, but at the wrong time. Very often I have also recognized, that there is lack of awareness of fact, that there are also Open Source applications that can help with the organization's portfolio. This thesis deals with the definition of requirements, search and selection of proper OSS application and subsequent implementation. In the first part of this thesis I prepare a theoretical framework about PPM, and on that basis then identify and verify a set of requirements for selection of right OSS PPM application. The next section of thesis describes searching for suitable applications from a variety of sources, assessment against the requirements and the final selection. In the last part the implementation project is drafted, which aims to serve other colleagues in project management as one of the possible implementation paths. The outputs are then continuously confronted with experts in matter of projects and project management in the field of IT in order to maximize symbiosis between theory and real life experiences. Virtually every project management methodology defines the collection of already-proven techniques, best practices or lessons learned from the previous similar implementations. The main contribution of this thesis I see in fact that it contains not only the design of the project implementation, but also describes a logical path, what leads to the result. Therefore this thesis could be used as a base or discussed best practice, when a project of implementation of PPM application in place.

The Finnish Landscape Photography and Its Role in Helsinki School

In my bachelor thesis I am focused on the images of the Finnish landscape photography and relationship to nature due to the harsh geographical conditions. I will describe the political and cultural influences that affected the aesthetic thinking about perception of scanned landscapes. Furthermore, I would like to deal with the influence by art that have shaped and inspired the Finnish authors and compare it with the perception of the country abroad. For example Dieseldorf photography school (Bernd and Hilla Becher, Andreas Gursky), Japanese photographers (Hiroshi Sugimoto, Rinko Kawauchi) Czech photographers (Josef Sudek, Josef Koudelka) and the others .

Mask, Disguise and Role Play in Worldwide Contemporary Self-Portrait

This bachelor thesis deals with the role of mask, disguise and playing with the roles through self-portrait in worldwide contemporary photography. In the first chapters it follows the general development of the self-portrait and a brief history of photography and its forms and tendencies from beginning to the present, it is also focused on the mask in general and role of masks and disguises in the history of photography. Main part of this work is the last chapter, which is devoted to the youngest generation of contemporary photographers. In their self-portraits we can follow tendency which reveals use of mask and disguise for change of their identity. However, they do not create a new identity but takes over, or form the illusion of existing already defined identities. The aim of this thesis is to present the mask as an instrument for role play through a self-portrait.

Cultural support in Prague's districts

Diploma thesis ”Cultural support in Prague's districts” focuses on a current situation in municipal strategical management and support of culture in all Prague districts. The introduction emphasises the importance of each level of government in the management and support of culture through comparing specific cultural policies. The research is divided into two parts. First one depicts how culture is being understood on the municipal level and presents an overview of existing instruments for support and planning. The second part is focused on a few municipal districts that handle their culture policy with a particular strategy and consider it to be an irreplaceable part of a local development. The second part also examines what tactics and tools these municipal districts use for a realization of their strategic goals. In conclusion, this thesis proves why the role of the local official government is so important in the developement and implementation of culture.

Role of Psb28 proteins in the biogenesis of the Photosystem II complex in the cyanobacterium Synechocystis sp. PCC 6803

The thesis focuses on the role of Psb28 proteins, namely the Psb28-1 and its homolog Psb28-2, in the biogenesis of the Photosystem II complex (PSII) in the cyanobacterium Synechocystis PCC 6803. The aims of this work were to localize the proteins within the cells, and to determine their function. A fraction of both Psb28 proteins was identified in the monomeric PSII core complexes but most proteins were found in the unassembled protein fraction associated with thylakoid membranes. Psb28-1 was mostly detected as a dimer while Psb28-2 as a monomer. Psb28-1 also differed from Psb28-2 by its higher affinity to the PSII core complex lacking CP43 antenna. Characterization of Psb28-less mutants suggested regulatory function of the proteins in PSII biogenesis in connection with chlorophyll biosynthetic pathway. Analysis of preparations isolated using FLAG-tagged versions of Psb28 proteins showed their association with Photosystem II - Photosystem I supercomplexes, especially under increased irradiance, and supported a role of Photosystem I in the PSII biogenesis.
