Appaloosa breeding and their utilization
Zuzjaková, Eva ; Neumann, Cyril (advisor) ; Starostová, Lucie (referee)
This bachelor thesis is a summary of the basic information about breeding and utilization of appaloosa. A dominant characteristic of appaloosa is an attractive color coat pattern, which is caused through a set of genes called Leopard Complex (Lp). That is the reason, why in this thesis, at first, the history of presence of the Lp in genus Equus takes place. The presence of Lp can be found already in many cave paintings of prehistoric horses. During domestication, the coat pattern became important. The horses of the same color coat pattern, which have today appaloosa, are depicted in the Celts, Scythians and Etruscans tombs, as well as in murals of Chinese and Egyptian cultures. Significant breeding of the spotted horses are also documented in the Ferghana Valley in eastern Uzbekistan. In Europe at 16th century horses were expanded and a lot of them have spotted coat pattern. Then the Spanish settlers imported these horses to the America, where Nez Perce Indian tribe bred horses, which we know today as Appaloosa. Furthermore, in this thesis the Appaloosa breed organization, which belongs complete to Appaloosa Horse club of America, is described. The breeding standard and characteristics, which are color coat pattern, mottled skin, visible white sclera and striped hooves are inducted. Seven various types of coat pattern and tree types of registration are also described. The major problems in the appaloosa breed are inherited diseases. In this thesis the most serious five of them are identified. It is CSNB, congenital stationary night blindnes associated with a homozygous variant of LP gene. HYPP, hyperkalaemic periodic paralysis, which causes muscle spasm of affected horses. This disease was obtained from quarter horses. HERDA - Hereditary Equine Regional Dermal Asthenia which causes irreversible damage to exposed skin. Another serious disease is PSSM, polysaccharide storage myopathy, which is one of many myopathy occuring in horses. The latter disease is lethal GBED. Affected foals lack the enzyme GBE which is necessary for the synthesis and storage of glycogen. In spite of the improving precautions, these diseases have not been fully eliminated from breeding. Further, there are given the possibilities of using Appaloosa horses in sports, hobby and ranch works. The system of shows and their ratings is described as well.

Relationship between Large-scale Chromatin Organization and Nucleolus in Human Cells
Kalmárová, Markéta ; Raška, Ivan (advisor) ; Nedvídek, Josef (referee) ; Smetana, Karel (referee)
In human cells ribosomal genes are organized as clusters called Nucleolus Organizer Regions (NORs) that are situated on the short arms of acrocentric chromosomes. It was found that essential components of the RNA polymerase I transcription machinery, including Upstream Binding Factor (UBF), can be detected on some NORs, termed "competent" NORs, during mitosis. The competent NORs are believed to be transcriptionally active during interphase. But since individual NORs cannot be observed in the cell nucleus, their interphase status remained unclear. To address this problem, we detected the competent NORs by two commonly used methods, UBF immunofluorescence and silver staining, and combined them with FISH for visualization of rDNA and/or specific chromosomes. We found that the numbers of competent NORs on specific chromosomes were largely conserved in the subsequent cell cycles, with certain NOR-bearing homologues displaying a very stable pattern of competence. Importantly, those and only those NORs, which were loaded with UBF, incorporated bromo-uridine in metaphase after stimulation with roscovitine and in telophase, suggesting that competent and only competent NORs contain ribosomal genes transcriptionally active during interphase. Applying premature chromosome condensation with calyculin A, we visualized...

Hemoprotein Nitric Oxide Synthase in Aplysia Californica
Buganová, Michaela ; Martásek, Pavel (advisor) ; Pelouch, Václav (referee) ; Druga, Rastislav (referee)
Nitric oxide (NO) plays a crucial role in neuronal signaling in a variety of eukaryotic and prokaryotic organisms. Nitric oxide synthases (NOS) are heme-containing monooxygenases that catalyze the oxygen dependent oxidation of L-arginine to NO and L-citrulline. The NO produced by NOS activity is a gaseous molecule that diffuses easily through membranes and acts inter or intracellularly. NO activates metal-containing enzymes, including soluble guanylate-cyclase (sGC) that increase levels of the messenger molecule cyclic 3,5-guanosine monophosphate (cGMP) (1, 2) which in turn mediate various pathophysiological or physiological functions in neurons. Nevertheless, many aspects of nitrergic neurons and NO function in the central nervous system (CNS) are unclear. The aim of research described in this thesis was to characterize neuronal NOS, proteins metabolically linked to NOS and NO signaling pathways in the CNS of Aplysia cali/ornica (Aplysia), a popular experimental model in cellular and system neuroscience. The biochemical characteristics of Aplysia NOS (AcNOSj described here revealed its calcium-/calmodulin-(Ca/CaM) and NADPH dependence. A representative set of inhibitors for mammalian NOS isoforms also suppressed NOS activity in Aplysia Polyclonal anti-rat nNOS antibodies hybridized with a putative purified...

Metamorphoses of the text of Arnošt Lustig's short story Tma nemá stín
Levorová, Eva ; Špirit, Michael (referee) ; Holý, Jiří (advisor)
The work deals with tracing development of Lustig's prose Tma nemá stín (Darkness Casts No Shadow) from its first edition as a short story in Věstník židovských náboženských obcí v Československu (Journal of Jewish Congregations in Czechoslovakia) through all other book editions of the short story (as part of collection Démanty noci) up to its elaboration into a nouvelle and its so far the last edition. There is mentioned a list of available editions of the fiction in the Czech language as well as translations into foreign languages (however the work does not pay further attention to foreign language editions). The changes and gradual extension of the text made by the author are typologically classified and there is indicated their significance for impression of the nouvelle. The work focuses on differences in the thematic and stylistic field and their motivation.

The pharmacotherapy of the asthma
Brožová, Lenka ; Melicharová, Ludmila (advisor) ; Pourová, Jana (referee)
Asthma bronchiale is a very common chronic disorder of airways with not fully elucidated pathology, which is not fully curable at the moment. It is estimated that 300 millions of persons suffer from asthma. About 8% of adult population and 10% of children are affected in the Czech republic. The aim of this thesis is to give an overview of contemporary modern pharmacotherapy of asthma. Firstly, this work describes asthma from pathophysiological and epidemiological point of view, among others: risk factors, used classification of asthma and its clinical diagnosis. The main emphasis is given particularly on pharmacotherapy with subdivision on preventive antiastmatic dugs (controllers) and rapid acting bronchodilatators (relievers) including the therapy of asthmatic exacerbation. Overviews of asthmatic pharmacotherapy during pregnancy and inhalatory system in the therapy of asthma at the present are summarized at the end.

Traumatic injury of peripheral nerves represents an important area of present-day clinical and experimental interest. Despite a large quantity of experimental data acquired in an intense research of this topic carried out in recent years, traumatic affection of peripheral nerves continues to be a complex surgical problem. Injury of peripheral nerves often results in a permanent and serious impairment in the patients' lives connected with the loss of the affected peripheral nerve function. If an injured nerve is given a timely and correct treatment, these consequences can be prevented to a certain extent. In the current practice this implies the use of all available methods of peripheral nerves surgery, primarily microsurgical operation technique while observing some of the basic principles essential for a good functional reinervation. This mainly applies to tension-free suture. In some anatomic locations and despite observing the above-mentioned facts, it is still difficult to reach good functional results. This primarily applies to brachial plexus injury which is one of the most complicated and serious of all lesions of peripheral nerves. Out of the total number of nerve injuries they are represented in large sets by three to seven percent. In his set, Midha reports 1.2% of patients affected by the...
