Scientific technology park Telč

The subject of bachelor thesis is a design of a building of a Scientific Technology Park with laboratories for Energoklastr in Telč. Energoklastr is competency oriented cluster focusing on co-operation of small and medium size businesses, research organizations, universities and public sector in field of applied research and transfer of technologies. It significantly contributes to development of innovations, applied research and transfer of technologies in field of lowering energy demand. Energoklastr especially focuses its activity on technology lowering emission levels and sustainability of energetics in fields like a production of electric devices, aerial industry, car industry, IT and construction. Building site for meant for the construction of VTP with laboratories of effective use of biomass in energetics is to be found in cadastral area of Telč. It is located in north-west part of town, in neighborhood of existing industrial and agricultural buildings. The property is adjacent to a first class road number 23 (Třebíč – Jindřichův Hradec), that is one of the main connection to highway D1. VTP building with the laboratory is designed like a two story wooden structure with a flat roof and no cellar. The building is trapezoidal shaped, which is adjusted to a shape of the property. The building is divided to laboratory part and administrative part according to a story. There is a suspension stainless steel staircase designed in a lobby, which is discussed in part D – Architectonical detail.

Pressure influence on the magnetic and transport properties of the f-electron compounds

In this thesis, we describe the work with a new pressure cell, its implementation in Joint Laboratory for Magnetic Studies, together with testing measurements necessary for ensuring good environmental conditions control and trouble-free operation. We have chosen a single crystal of the CePtSn compound to be the first representative measurement. We measured electrical resistivity along crystallographic b-axis in conditions: T = 2-300 K, p = 0-2.5 GPa and magnetic field B = 0-14 T applied along b-axis. We observed only minimal pressure influence on temperatures of magnetic phase transitions, however interesting and rather unusual behavior in magnetic fields was revealed. Previous works reported two field-induced transitions at low temperatures and ambient pressure, with values of the critical fields BcLF ~ 4 T and BcHF ~ 11.5 T, accompanied with large magnetoresistence step of -30% and +10%, respectively. With application of pressure, we observed shift of the critical field of low-field transition BcLF to the higher fields, whereas the value of critical field of high-field transition BcHF was unaffected with applied pressure. T cells labelling by bimodal contrast agent for in vivo studies of stroke

Stroke is a serious brain injury, which causes sudden death or terminates in permanent neurological disability. Nowadays, tissue plasminogen activator (tPA) is used as the only effective treatment of stroke. One of the potential targets for novel therapy are T cells. Even though the explicit role of T cells in the pathogenesis of brain injury, amounts and timing of all T cell subtypes infiltrating into brain during the stroke still needs further investigation. The research in this field is complicated by the lack of efficient methods for in vivo cell tracking. Therefore the aim of this thesis was to develop a method of T cells labelling by MRI contrast agent in order to investigate T cells distribution in ischemic mice model using in vivo MR imaging. T cells were isolated from C57/BL6 mice in two step isolation protocol using gradient centrifugation and magnetic separation with the efficiency of 97 %. The isolated cells were labelled with 100 μg Fe/mL of Molday ION Rhodamine B contrast agent. The labelling efficiency after 17 hours of cells incubation was higher than 99 %. The labelled cells were cultured with CD3 and CD28 antibodies resulting into the 74 % viability of labelled T cells compared to 83 % viability of non labelled T cells. ELECTRON BEAM REMELTING OF PLASMA SPRAYED ALUMINA COATINGS

Plasma sprayed alumina coatings find numerous applications in various fields, where they enhance the properties of the base material. Examples include thermal barriers, wear resistance, electrical insulation, and diffusion and corrosion barriers. A typical structure of plasma sprayed coatings, containing a multitude of voids and imperfectly bonded interfaces, gives them unique properties - particularly low thermal conductivity, high strain tolerance, etc. However, for certain applications such as permeation barriers or wear resistance, these voids may be detrimental.\nThis paper reports on the first experiments with remelting of plasma sprayed alumina coatings by electron beam technology, with the purpose of densifying the coatings and thereby eliminating the voids. Throughout the study, several parameters of the e-beam device were varied - beam current, traverse velocity and number of passes. The treated coatings were observed by light and electron microscopy and the thickness, structure and surface morphology of the remelted layer were determined and correlated with the process parameters. Based on the first series of experiments, the e-beam settings leading to dense and smooth remelted layer of sufficient thickness were obtained. In this layer, a change of phase composition and a marked increase in hardness were observed.\n

Mechanical and Electrical Properties of Thin Metal Films Deposited by Vacuum Evaporation

Thin layers are widely used in many fields of technology and today we can say that they are found in all modern technologies. Thin layers can be created in two ways, namely by chemical or physical means. This work focuses on the latter method, more particularly a technology of thermal evaporation of thin layers in a vacuum. The work focuses on the process principles during and after the evaporation. Much of the work focuses on the development and design of experiments. These experiments illustrate some of the phenomena that take place on thin films produced by the aforementioned technology. Work helps to better understand processes during formation of thin layers and properties that influence the quality and stability of thin films. In conclusion we describe results of experiments and new developments in the field of thin films deposition using evaporation under vakuum are summarized.

The structural and the fundamental mechanical properties of intermatallics on the base of TiAl

Gamma TiAl alloys are emerging as a pervasive structural material. Remarkable progresses have been made through refining lamellar microstructure and grains, microalloying, chemistry modification, process development and/or their combination. The main thrust was to enhance resistance to creep deformation, oxidation and high cycle fatigue deformation/failure. Less or even no knowledge have been obtained in the field of the intermetalics fracture at uniaxial or bending loading, there are missing data about their behaviour at the presence of cracks and other defects. The reason is simply in the other properties that are key from application point of view. Due to their brittleness at temperature close to room temperature the investigation of the intermetalics fracture is complicated but promissible provided it bring ideas for the further development of the alloying and microstructure. The paper is focused on the first steps in this field.

The analysis of aberrations in the localization of the radiated fields during irradiation of the patients with rectum tumors by EPID method

The presented thesis deals with the analysis of deviations in radiation fields during the irradiation of the patients with rectal tumors. The first part of the thesis is focused on the current problems connected with rectal and rectosigmoid cancer both in the Czech Republic and worldwide. It also includes the description of risk factors, characterisation of the given type oftumor, manifestation of the disease as well as the prognosis. What is also tackled is the prevention, which is important and often neglected.It was necessary to mention the possibilities of screening and dispensarisation programs. The next part deals with the diagnostic process (from anamnesis to specialised examinations) as well as the therapy. Chemotherapy and surgery are mentioned only marginally whereas radiotherapy, including its planning and methods, is dealt with in a more detailed way. The main part consists of the analysis of deviations in radiation fields. The deviations have been obtained by means of EPID (electron portal image device) in a sample of patients with rectal and rectosigmoid cancer who are treated with the use of radiotherapy at the oncological ward of the Hospital České Budějovice, Inc. The maximum, minimum and average deviation in the localization of radiation fields were assessed statistically, by means of the khi-quadrate test. The results obtained will be submitted to the oncological ward of the Hospital České Budějovice, Inc., where they can contribute to the assessment of radiotherapy quality. The thesis includes supplements such as portal images and tables with graphs indicating the development of the incidence and mortality for the chosen diagnosis within the entire population.
