Relationship between Large-scale Chromatin Organization and Nucleolus in Human Cells

In human cells ribosomal genes are organized as clusters called Nucleolus Organizer Regions (NORs) that are situated on the short arms of acrocentric chromosomes. It was found that essential components of the RNA polymerase I transcription machinery, including Upstream Binding Factor (UBF), can be detected on some NORs, termed "competent" NORs, during mitosis. The competent NORs are believed to be transcriptionally active during interphase. But since individual NORs cannot be observed in the cell nucleus, their interphase status remained unclear. To address this problem, we detected the competent NORs by two commonly used methods, UBF immunofluorescence and silver staining, and combined them with FISH for visualization of rDNA and/or specific chromosomes. We found that the numbers of competent NORs on specific chromosomes were largely conserved in the subsequent cell cycles, with certain NOR-bearing homologues displaying a very stable pattern of competence. Importantly, those and only those NORs, which were loaded with UBF, incorporated bromo-uridine in metaphase after stimulation with roscovitine and in telophase, suggesting that competent and only competent NORs contain ribosomal genes transcriptionally active during interphase. Hemoprotein Nitric Oxide Synthase in Aplysia Californica

Nitric oxide (NO) plays a crucial role in neuronal signaling in a variety of eukaryotic and prokaryotic organisms. Nitric oxide synthases (NOS) are heme-containing monooxygenases that catalyze the oxygen dependent oxidation of L-arginine to NO and L-citrulline. The NO produced by NOS activity is a gaseous molecule that diffuses easily through membranes and acts inter or intracellularly. NO activates metal-containing enzymes, including soluble guanylate-cyclase (sGC) that increase levels of the messenger molecule cyclic 3,5-guanosine monophosphate (cGMP) (1, 2) which in turn mediate various pathophysiological or physiological functions in neurons. Nevertheless, many aspects of nitrergic neurons and NO function in the central nervous system (CNS) are unclear. The aim of research described in this thesis was to characterize neuronal NOS, proteins metabolically linked to NOS and NO signaling pathways in the CNS of Aplysia cali/ornica (Aplysia), a popular experimental model in cellular and system neuroscience. Metamorphoses of the text of Arnošt Lustig's short story Tma nemá stín

The work deals with tracing development of Lustig's prose Tma nemá stín (Darkness Casts No Shadow) from its first edition as a short story in Věstník židovských náboženských obcí v Československu (Journal of Jewish Congregations in Czechoslovakia) through all other book editions of the short story (as part of collection Démanty noci) up to its elaboration into a nouvelle and its so far the last edition. There is mentioned a list of available editions of the fiction in the Czech language as well as translations into foreign languages (however the work does not pay further attention to foreign language editions). The changes and gradual extension of the text made by the author are typologically classified and there is indicated their significance for impression of the nouvelle. The work focuses on differences in the thematic and stylistic field and their motivation.

This work aims to map the occurrence and development of infectious occupational diseases (INZP) in 2001-2006 and the possibilities of prevention in selected INZP listed in the list of diseases occupational (NZP) under 1, ie interhumánním transmission. Occupational disease is a disease listed in the List of Occupational Diseases, which is part of Government Regulation No. 290/1995 Coll. The disease referred to in this list are divided into six chapters. Infectious and parasitic diseases are given in Chapter V. It is divided into three items. Traumatic injury of peripheral nerves represents an important area of present-day clinical and experimental interest. Despite a large quantity of experimental data acquired in an intense research of this topic carried out in recent years, traumatic affection of peripheral nerves continues to be a complex surgical problem. Injury of peripheral nerves often results in a permanent and serious impairment in the patients' lives connected with the loss of the affected peripheral nerve function. If an injured nerve is given a timely and correct treatment, these consequences can be prevented to a certain extent. In the current practice this implies the use of all available methods of peripheral nerves surgery, primarily microsurgical operation technique while observing some of the basic principles essential for a good functional reinervation. This mainly applies to tension-free suture. In some anatomic locations and despite observing the above-mentioned facts, it is still difficult to reach good functional results. This primarily applies to brachial plexus injury which is one of the most complicated and serious of all lesions of peripheral nerves. Metody: V práci byla použita metoda Videotrénink interakcí, jakožto metoda intervenční, rozvíjející, vycházející z kvalitativních prvků. Výsledky: Uvedené příklady práce s trenéry mají pouze informativní a ilustrativní charakter. Prostřednictvím videonahrávek jsem chtěla přiblížit způsob práce s touto metodou v prostředí sportu a popsat sledované interakce mezi trenérem a hráčkou. Nevyvozuji tudíž z těchto krátkých ukázek žádné kategorické závěry, jsou jen jistým naznačením tendence vlivu videotréninku interakcí na rozvíjení komunikačních dovedností sportovního trenéra. The proposal fish ladder

This graduation theses describes the design of the fish ladder at the dam in Brno (district Přízřenice). In the introduction I mentioned migration patterns of fish. For a better understanding outlined and fish behaviour associated with migration, some fish undertake long voyages others migrate only a few meters. Extremely important is that for most species of fish migration is important for their survival, whether breeding or feeding. Middle part is focused for water structures and water flow, specifically to the list of dams and their distribution from different perspectives. Nowadays the progress should go ahead with nature and technical buildings. The work also has a list of fish ladders with their description and evaluation of suitability. The last part is dedicated to the design of fish passage in Přízřenice. The weir has been built on a 35,293 river km river Svratka. In this part of the work is summarized description of the location in which it is proposed fish passage. Furthermore, the list of the most abundant fish stocks at a fishing ground Svratka 3.

The concrete with a higher resistance to high temperatures

The result of this master’s thesis is acquaintance with the issues of cement concrete resistance to high temperatures. This work describes the processes ongoing in the structure of concrete at extreme thermal loads and analysis of effects of high temperatures on the individual components of reinforced concrete structures is given, then influence of high temperatures and fire on the aggregates, matrix, reinforcement steel and the resulting changes in their properties. In the experimental part is given design of composition of concrete with a higher resistance to high temperatures and subsequent verification of the physico-mechanical properties of sample of concrete after exposure to temperatures at intervals of 200 ° C, 400 ° C, 600 ° C and 900 ° C. In individual recipes were observed changes of volume weight, changes of compressive strength and tensile strength flexural , tensile strength of surface layers and the occurrence of cracks and then were compared the results of thermally loaded and unloaded samples. The benefit in this issue is to evaluate the surface appearance of samples after heat load - study of area of crack and measurement of their width - was used according to the methodology's authors Xing, Hebert, Noumowe a Ledesert given in Cement and Concrete Research. This methodology allows to quantify changes of surface, after temperature load.
