PharmDr. Martina Čečková, Ph.D. Consultant: PharmDr. Lenka Ťupová, Ph.D. Title of diploma thesis: Study on expression of selected ABC transporters in placenta The placenta is a temporary organ through which the fetus is supplied with nutrients and oxygen from the mother's blood and conversely waste substances are transferred into the mother's blood during pregnancy. Substance transfer through the placenta is a complex process controlled by a number of physiological mechanisms, including passive diffusion, facilitated diffusion or active transport, which is realized by activity of membrane transporters with energy consumption. Presence of active ABCs efflux transporters in the placenta has been known for a long time and their function is associated primarily with limiting the entry of harmful substances into the placenta and further into the fetus, thus contributing to its protection. PharmDr. Martina Čečková, Ph.D. Title of diploma thesis: Effect of CDK and FLT3 inhibitors on activity of ABC efflux transporters in vitro, relation to multidrug resistance P-gp and BCRP are transmembrane proteins that form part of a large family of ABC transporters. These are ATP-driven transporters, which main task is to eliminate exogenous and endogenous substances and their metabolites from cells of both, healthy and tumour tissues. This activity is often associated with the expulsion of administered therapeutics and multiple drug resistance (MDR) in tumour cells. A promising therapy of cancer represents a newer class of drugs target the tyrosine kinase (TK), and cyclin-dependent kinases (CDK), which are cell enzymes responsible for the processes of proliferation, apoptosis and differentiation. One of the important functions provided by the placenta is the transport of substances between the mother and the fetus. This transfer of substances enabled mainly by membrane transporters, which are located on the apical and basal membranes of the syncytiotrophoblast. During various physiological or pathological changes in the human body, their expression and amount can vary significantly. Inflammatory reactions that may occur during pregnancy are also related to these changes, and therefore we have addressed this issue and believed that this condition may alter the expression of placental transporters. PharmDr. Martina Čečková, Ph.D. Title of rigorous thesis: Study of the effect of roscovitine and its derivatives on the expression of drugs transporters in vitro In this study we aimed to evaluate interactions of selected inhibitors of cyclin-dependent kinases (CDKi) with efflux transporters ABCB1 (P-glycoprotein, P-gp, MDR1) and ABCG2 (breast cancer resistance protein, BCRP) in cell line LS174T derived from human adenocarcinoma of colon. Among the selected compounds belong roskovitin and its derivates designed as BA-12 and BP-14. Previous studies demonstrated that these substances inhibit both mentioned transporters and therefore increase the effect of simultaneously administrated cytotoxic drugs. Since the drug interactions on the level of transporters can arise from induction of their corresponding genes, the aim of this work was to extend the current knowledge on mentioned derivates of roskovitin in terms of possible affection of ABCB1 and ABCG2 expression. Nowadays cancer remains one of the most challenging health issues worldwide. Chemotherapy represents one of the essential approaches in the treatment of malignant diseases. However, multidrug resistance (MDR), a multifactorial phenomenon described as a loss of sensitivity of cancer cells to several diverse chemotherapeutic agents at the same time, often compromises the therapy outcomes. A well-known cause of MDR is an increased expression or/and an enhanced activity of efflux drug transporters of ATP binding cassette (ABC) superfamily, which has been found in many types of cancer. In the last decade, an expanding body of literature suggested a new hallmark of cancer cells - inflammation. Plant sugar metabolism and arbuscular mycorrhizal symbiosis

The study of arbuscular mycorrhizal symbiosis (AMS) - the mutualist relationship between the most of land plants and evolutionary old fungal group Glomeromycota - is becoming a prestigious topic. The prevalence of and extent of physiological action of AMS on plants is very interesting for the plant biology itself, but its importance grows, notably in time of global climate change, frequent soil degradation and ascending exhaustion of mineral fertilizer reserves. Although the flows in AMS of some minerals, like of phosphorus was enlightened, carbon exchange between the symbionts is still poorly understood. In this experimental work, I utilized the boom of molecular and bioinformatic methods in the quest for completely unexplained carbon flows. The organisms used include barrel medic (Medicago truncatula), the model legume for symbiotic relationships, biotic, and abiotic stresses; Rhizophagus irregularis, the widely used fungus for such experimental studies of AMS; and Sinorhizobium meliloti, the nodulating nitrogen-fixing bacterium compatible with the barrel medic. This bachelor work is concerned with design of the remedy, how to improve the market position of the firm. In the first part of this work principles of Economy are used as initial view. Subsequent second part of work utilizes Economy principles for MISVO TRANS s.r.o. company dates. The result of this work is to design changes in the firm to reduce costs and to solve company problems to be able to survive in the time recession and prosper in the future.
Reconstruction of discharging shakeout machine electric installation

This Diploma work is concerned with the design of project documentation for discharging shakeout machine electric installation. This work continues in the Semestral project 2 in which the background of a new machine was made. Then, this work deals with the replacement of a switch room with required devices including the design of a new connecting cables and power streams. In the first part, the work deals with the control part of the device and the design of control and signal cables including the product documentation of the device. In the second part, the work deals with the proposal of building electro installation in which a new lighting and its use is described. The last part focuses on the coordination during the realization of a suggested device. In this part, all problems coming from the particular working procedures including a proposal of possible solutions are described. According to the project documentation and following realization, the documentation of final concept reflecting the real status of the device is described.
