Klimková, Kateřina ; Mladěnka, Přemysl (advisor) ; Hrdina, Radomír (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Pharmacology and Toxicology Candidate: Kateřina Klimková Supervisor: Assoc. Prof. Přemysl Mladěnka, Pharm. D., Ph.D. Consultant: Assoc. Prof. Kateřina Valentová, Ph.D. Title of Thesis: Determination of the stoichiometry of the copper complexes with dehydrosilybin A Silymarin, the standardized extract of the milk thistle (Silybum marianum), is a widely used approved over-the-counter drug that is recommended for a number of liver diseases. Silymarin contains as one of its components 2,3-dehydrosilybinA, which has an appropriate metal binding site in its structure. In general, flavonolignans, due to their structure, can interact with transition metals in the gastrointestinal tract by forming complexes. This property can be useful for the protection against excessive amounts of metals in the body. The aim of this in vitro study was to analyse the interaction of 2,3-dehydrosilybin A with copper, which plays a crucial role in the organism as a cofactor of many enzymes. Although being an essential element, it can, however, be toxic at elevated levels. Stoichiometry, as one of the most important characteristics of the complex, was determined by UV-Vis spectrophotometry in four (patho)physiological pH conditions (4.5; 5.5; 6.8; 7.5)...
Intraspecific variability in seeds nutrient stoichiometry
Kesl, Tomáš ; Mašková, Tereza (advisor) ; Konečná, Marie (referee)
Immediately after germination, the plants depend on the nutrients stored in the seed from which they germinated, until the nutrient intake is completely replaced by the mature leaves and roots. Nutrients in seeds are important for early development of vegetative organs. Their content and concentration in seeds can be influenced by various factors such as nutrient availability in soil, latitude, temperature, photoperiod, granivory and competition. This bachelor's thesis summarizes current knowledge about intraspecific and interspecific variability of seed nutrient stoichiometry, specifically nitrogen, phosphorus and carbon, as they most often limit plant growth. For all three elements, common and different trends can be observed, both at the intra-species and inter-species level. The only trend comparable between the intra-species and inter-species level is correlation of carbon seed concentration and latitude, while at the inter-species level there is also the opposite trend of negative correlation of latitude and carbon concentration in seeds. Other observed trends are not comparable between the intra-species and inter-species level. It is not possible to compare these trends, as there is lack of studies on the variability of seed nutrient stoichiometry. Therefore it is necessary to focus on seed...

Digestate Grate Boiler
Krejčiřík, Jiří ; Milčák, Pavel (referee) ; Lisý, Martin (advisor)
This master thesis deals with the properties of digestate in combustion, design of construction solution of digestate boiler and experimental verification of boiler parameters. The first chapter deals with the properties of digestate as a fuel. It is followed by a chapter dealing with the existing measured results on a similar boiler. This is followed by a stoichiometric calculation of the digestate boiler. The next chapter is devoted to combustion chamber models. This chapter is followed by a chapter with experimental verification of chamber models and combustion process tuning. The last chapter contains design of construction and operational measures.

Assessment of the complex stoichiometry of the active dexrazoxane metabolite ADR-925 with iron and copper by the standard Job's method
Szotáková, Tereza ; Mladěnka, Přemysl (advisor) ; Jirkovský, Eduard (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Pharmacology and Toxicology Candidate: Tereza Morávková Supervisor: Assoc. Prof. Přemysl Mladěnka, Pharm.D., Ph.D. Title of Thesis: Assessment of the complex stoichiometry of the active dexrazoxane metabolite ADR-925 with iron and copper by the standard Job'smethod Iron and copper are important trace elements which participate on many physiological processes in humans. Their kinetics in the organism is tightly regulated since both lack or excess of these elements are associated with pathological states. Free ions of iron and copper can catalyse reactive oxygen species (ROS) production and hence cause damage to proteins and DNA. Imbalance in these metals is linked with diabetes, cardiotoxicity, cirrhosis of the liver and neurodegenerative diseases as Alzheimer's disease or Parkinson's disease. Dexrazoxane is a drug with documented protecting effect against cardiotoxicity of anthracyclines. A former theory associated its protective effects against these cytotoxic drugs with the iron-chelating properties of its active metabolite ADR-925. The goal of this diploma thesis was to explore if ADR-925 is able to chelate Fe2+ , Fe3+ , Cu+ a Cu2+ ions at physiologically and pathophysiologically relevant pH values (4,5; 5,5; 6,8 and 7,5) and...
Food quality impact on growth and survival of Daphnia longispina (Crustacea: Cladocera)
Sýkorová, Veronika ; Sacherová, Veronika (advisor) ; Macháček, Jiří (referee)
This thesis focuses on ecological processes among plankton organisms. It especially questions the influence of food quality on growth and survival of freshwater plankton. I focused on the influence of various ratios of nitrogen and phosphorus in available food. I am also interested in finding out the food requirements of chosen consumer Daphnia longispina. Within this broad topic, I specify a question, whether various quality and quantity of food influences the life history parameters of the species Daphnia longispina and whether the absence of common species of Cladoceran Daphnia longispina in Plešné Lake is caused by unsuitable quality of food. I have bred Daphnia longispina in laboratory conditions for two years. I fed it by algae Monoraphidium dybowskii isolated from Plešné Lake and grown in two media P-limited (C:P 1174), P-unlimited (C:P 290) and three concentration (1,2, 4 mg C/L). Cladocerans fed by food from medium that had a C:N:P ratio approximately the same as can be found in Plešné Lake (C:P 1174) lived for a singificantly shorter period of time and during the experiment they did not reproduce even in the case with the largest amount of food (4 mg C/L). This is a possible reason why Daphnia longispina did not return to Plešné Lake after the period of strong acidification, climax of...
Diet quality impact on growth and survival of freshwater zooplankton species
Sýkorová, Veronika ; Sacherová, Veronika (advisor) ; Nedbalová, Linda (referee)
Food quality has a significant influence on freshwater zooplankon. It influences its growth rate and development generally. By quality we understand a certain stoichiometric ratio of the main biogenic elements (carbon, nitrogen, phosphorus) in the biomass of food. Ecological stechiometry is helping to understand the balance of chemical elements in ecological interactions and processes. Freshwater zooplankton consists mostly of two groups of organisms: rotifers (Rotifera) and crustaceans (Crustacea) - cladocerans (Cladocera) and copepods (Copepoda). Each of this groups requires different quality of food. Most of filtering cladocerans are fast- growing organisms, which need a phosphorus-rich food for their fast development (small C:P ratio in food) and vice versa for most of the copepods. The food offered is not always ideal for all members of zooplankton, that is why various pre- and post- absorptional mechanisms developed to help them to cope with unsuitable food. The ability to survive unvafourable conditions is closely linked to the zoolpankton's reactions to unsuitable C:N:P ratio in its food. The C:N:P ratio in food of zooplankton also influences the species distribution in water bodies. Key words: stoichiometry, food quality, zooplankton
